ABSTRACT: egr-2 is an injury-responsive gene that is upregulated in the anterior of planarian tail fragments and required for intestinal remodeling. The goal of this study was to identify differentially expressed transcripts in the anterior region of egr-2(RNAi) tail fragments relative to egfp(RNAi) control fragments.

Project description:To investigate the role of alx-3 in activating wound-induced transciptional programs during anterior regneneraiton, control(RNAi) and alx-3(RNAi) planarians were amputated below the pharynx and were allowed to regenerate for 0.5 hours post-amputation (HPA), 6HPA, 24HPA, 48HPA and 72HPA. Intact animals were are also included.Tissue from tail fragments and intact animals was collected at each time point for RNA extraction and cDNA synthesis. Gene expression profiling analysis was performed using data obtained from RNA-seq of planarian tissues at 5 timepoints after regeneraiton and in uninjured animals .

Vu2019 - Planarian double-head regeneration

Project description:This spatio-temporal model explores tissue polarity re-organization during planarian double-head formation upon regeneration with beta-catenin-1 RNAi treatment. Initial conditions for A and M polarity fields are chosen near the wild type pattern and the first 100 time steps are used to establish the typical wild type splay pattern. The induction of the ectopic head occurs at time-step 100 (interpreted as tail amputation time) at the position where the former tail was (right end), subsequently affects the nearby tissue with some lag and then re-polarizes the posterior half of the tissue from time-step 200 onward. Of these polarity fields, A represents the anterior-posterior polarization system provided by the core PCP pathway, M represents the medio-lateral polarization system provided by the Ft/Ds pathway and P represents the resulting superimposed and normalized polarization. P can be compared to the observable polarity data of ciliary rootlets. This simulation reproduces Figure 4D of the referenced paper. Parameter values correspond to the beta-catenin-1(RNAi) pattern as given in Table S2 of the referenced paper. The model file is in MorpheusML format and can be opened in the free, open-source multicellular modeling software Morpheus (https://morpheus.gitlab.io) to simulate the time course (movie) of the spatio-temporal dynamics.

iTRAQ-Based Quantitative Proteomic Analysis of the Initiation of Head Regeneration in Planarians

Project description:The planarian Dugesia japonica has amazing ability to regenerate a head from the anterior ends of the amputated stump with maintenance of the original anterior-posterior polarity. Although planarians present an attractive system for molecular investigation of regeneration and research has focused on clarifying the molecular mechanism of regeneration initiation in planarians, but proteomic studies are still in the early stages. Here, a global proteomics analysis of regenerating head fragments in planarians at 0h, 2h and 6h after amputation was performed using isobaric tags for relative and absolute quantitation (iTRAQ)-based quantitative proteomics coupled with LC-MS/MS strategy. Then the significantly changed proteins were identified. The correlation betwwen protein expression profiles and signaling pathways/biological processes was analyzed by bioinformatics and systems biology.

Multiplex single cell analysis of serotonergic neuron function in planarians reveals widespread effects in diverse cell types

Project description:We used scRNA-seq to examine serotonergic function in asexual planarians. We profiled 47,292 single cell transcriptomes of planarian of 3 different RNAi categories: gfp(RNAi), lhx1/5-1(RNAi) and pitx(RNAi). We detected changes in cell type frequency and gene expression patterns in planarian of different knock-down conditions, capturing regulatory programs of distinct cell types in response to loss of serotonergic function.
