Secretome analysis in CAF-dependent conditioned mediums with FaDu cells

ABSTRACT: We co-cultured primary cultured CAF (CAF-P), which promotes progression more actively, and CAF (CAF-D), which have less cancer-promoting properties, with OSCC cells, and analyzed the differential secretion through antibody microarray.

Project description:Gene expression profiling of CAF co-cultured with human oral squamous cell carcinoma (OSCC) cells compared with mono-cultured CAF. To identify key molecular regulators expressed by carcinoma-associated fibroblasts (CAF) that promote cancer cell invasion, microarrays were performed by comparing co-cultured OSCC cells and CAF with monoculture controls. comparison 1: CAF co-cultured with OSCC vs. mono-cultured CAF comparison 2: OSCC co-cultured with CAF vs. mono-cultured OSCC 1.7X10(5) YD-10B OSCC cells and 1.7X10(5) CAF were seeded in the upper chamber and lower chamber, respectively, of 6-transwell plates containing collagen-coated 1 micrometer pore transmembrane filters (Becton Dickinson, Franklin Lakes, NJ, USA). Monoculture control samples were generated by culturing only CAF or OSCC on the same side of the filter as in the co-culture design.

Proteome and secretome of human mammary normal fibroblasts and CAF

Project description:The secretome of cancer and stromal cells generates a microenvironment which contributes to tumour cell invasion and angiogenesis. Here, we compare the secretome of human mammary normal fibroblasts and cancer-associated fibroblasts (CAF). We discover that the chloride intracellular channel protein 3 (CLIC3) is an abundant component of the CAF secretome. Secreted CLIC3 promotes invasive behaviour of endothelial cells to drive angiogenesis and increases invasiveness of cancer cells both in vivo and in 3D cell culture models, and this requires active transglutaminse-2 (TGM2). CLIC3 acts as a glutathione-dependent oxidoreductase which reduces TGM2 and regulate TGM2 binding to its cofactors. Finally, CLIC3 is secreted also by cancer cells, is abundant in the stromal and tumour compartments of aggressive ovarian cancers, and its levels correlate with poor clinical outcome. This work reveals an unprecedented invasive mechanism whereby the secretion of a glutathione-dependent oxidoreductase drives angiogenesis and cancer progression by promoting TGM2-dependent invasion.

Project description:Purpose : Primary cultured fibroblast cultures can be classified based on their tumor promoting ability. Normal associated fibroblasts (NAF) cannot transform benign epithelia. Cancer associated fibroblasts (CAF) can transform benign epithelia into cancer. As described in Kato and Placencio et al., High passage CAF (CAF_HiP) cells have lost the ability to potentiate tumor growth. This study sought to identify the differences among three groups of fibroblasts: NAF, CAF, and CAF_HiP Methods :Human male prostatectomy tissues were cultured to generate primary cultured fibroblast cultures. Results and Conclusions : Differential gene expression among the top 200 genes analzed as described in Kato and Placencio et al. revealed 33 secreted proteins. SFRP1 was confirmed to be highly expressed in CAF and was shown to play a role in prostate cancer tumor progression and neuroendocrine differentiation.

Project description:Molecular mechanisms of the cancer cells-carcinoma associated fibroblasts (CAF) interactions growing in vitro conditions as a co-culture. The five canine mammary cancer cell lines were cultured with CAF (isolated from canine mammary cancer) for 72hrs. Then, the cancer cells and CAFs were sorted and the gene expression analysis was conducted. The control for each co-cultured cell line was the same cell line growing as a single culture.
