MiRNA expression profiles in plasma-derived exosomes of patients with premature ovarian failure

ABSTRACT: Exosomes are extracellular vesicles that have been shown to have therapeutic effects. However, the role of plasma-derived exosomal miRNA in premature ovarian failure (POF) remains ambiguous. To investigate the epigenetic pathogenesis in POF, we analyzed the expression profile of miRNA in exosomes derived from plasma of health control and POF patients using RNA sequencing.

Project description:Despite a significant progress in the treatment of Acute Respiratory Distress Syndrome (ARDS), our ability to identify early patients and predict outcome remains limited. In this study, we aimed to characterize small RNA content of plasma exosomes from ARDS patients in order to identify potential diagnostic biomarkers of the disease. For the first time, we profiled miRNA expression levels in plasma-derived exosomes from ARDS patients (n=8) compared to healthy subjects (n=10) by small RNA-seq. It allowed us to identify 12 exosomal miRNAs differentially expressed in ARDS context (padj<0.05).

The miRNAs in plasma exosomes may serve as markers for non-segmental vitiligo

Project description:More and more studies have showed that plasma exosomal miRNAs are biomarkers for disease. The aim of the study were to investigate the miRNA profiling in plasma exosomes of patients with non-segmental vitiligo (NSV) and to find biomarkers in plasma exosomes for patients with NSV. Plasma exosomes and exosomal RNA of 10 NSV patients and 10 health persons were purified by exoRNeasy Serum/Plasma Maxi Kit. The miRNA profiles of the 20 samples were sequenced using HiSeq 2500 (Illumina) and analyzed by Reads Per Million (RPM) values and edgeR algorithm. Some differently expressed miRNAs in plasma exosomes and skin tissues of the two sets were validated by qRT–PCR.Several miRNAs were confirmed by qRT–PCR and showed similar expression patterns between plasma exosomes and skin tissues. Our study depict the miRNAs expression profiles in plasma exosomes of NSV patients and suggest that several miRNAs in plasma exosomes may serve as biomarkers for NSV.

Some differently expressed miRNAs in plasma exosomes and skin tissues of the two sets were validated by qRT–PCR.A total of 85 miRNAs in plasma exosomes showed differential expression between SV patients and health persons, with a |log2（Fold Change）|≥1 and P-value < 0.05. Several miRNAs were confirmed by qRT–PCR and showed similar expression patterns between plasma exosomes and skin tissues. Our study depict the miRNAs expression profiles in plasma exosomes of SV patients and suggest that several miRNAs in plasma exosomes may serve as biomarkers for SV.

Serum Exosomal Omics Analysis: Revealing Energy Metabolism Anomalies and Promising Diagnostic Markers for Premature Ovarian Failure

Project description:Background: Premature ovarian failure (POF) is a clinical condition characterized by the cessation of ovarian function, leading to infertility. The underlying molecular mechanisms remain unclear, and no predictable biomarkers have been identified. This study aimed to investigate the protein and metabolite contents of serum exosomes to identify underlying molecular mechanisms and explore potential biomarkers. Methods: This study was conducted on a cohort consisting of 14 POF patients and 16 healthy controls. The exosomes extracted from the serum of each group were subjected to label-free proteomic and unbiased metabolomic analysis. Differentially expressed proteins and metabolites were annotated. Pathway network clustering was conducted with further correlation analysis. The biomarkers were confirmed by ROC analysis and random forest machine learning. Results: The proteomic and metabolomic profiles of POF patients and healthy controls were compared. Two subgroups of POF patients, Pre-POF and Pro-POF, were identified based on the proteomic profile, while all patients displayed a distinguishable metabolomic profile. Signaling pathway clustering revealed progression of dysfunctional energy metabolism during the development of POF. The differentially expressed proteins and metabolites were highly correlated, with six of them selected as potential biomarkers. ROC curve analysis and random forest machine learning suggested that AFM combined with GE was a diagnostic biomarker for POF. Conclusion: Omic analysis revealed that inflammation and oxidative stress are factors that damage ovarian tissue, but progressive dysfunction of energy metabolism might be the critical pathogenetic pathway contributing to the development of POF. AFM combined with GE together serves as the best biomarker for clinical POF diagnosis.
