T cell stimulation of resolved and non lesional epidermis- psoriasis patients

ABSTRACT: Biopsies were stimulated with OKT3 16 hours, epidermis was separated from dermis, RNA was purified and nanostring analysis was performed on a custom panel of transcripts.

Project description:The objective of this study was to investigate how active dermal connective tissue composition dynamics affect long term protein expression in the epidermis. After surgically removing damaged skin to its full thickness at the site of a deep burn in ten patients, we randomized three adjacent test areas within the wounds to receive either: 1) no dermal component as control, 2) a permanent artificial dermal matrix or 3) a temporary dressing to induce formation of granulation tissue. Each area was then covered with an autologous superficial healty skin transplant as the epidermal indicator. One year later, biopsy samples were taken from each site. The epidermis and dermis were selectively microdissected by laser-capture from cut tissue biopsy sections, and analyzed using untargeted non-labelled quantitative proteomics.

Project description:Analysis of the transcriptional response to aneuploidy in mouse epidermis. In this study we measured the transcriptional response to aneuploidy by aboragting the spindle checkpoint in mouse epidermis. We found that, whereas spindle checkpoint inactivation in the epidermis is tolerated, but results in metabolic deranged cells, SAC abrogation kills bulge stem cells Mad2; K14-Cre mice were sacrificed at indicated timepoints and epidermis was separated from dermis using overnight trypsin. RNA was isolated and expression patterns were compared between K14-Cre; Mad2f/f and Cfre negative animals for all timepoints.

Project description:Purpose: To deeply understand the clinical as well as pathogenetic differences, we have analyzed the genes involved in the epidermal lipid synthesis, epidermal development process and immune responses in the skin of AD and PSO patients. Materials and Methods: We have compared the transcriptomes of lesional skin (epidermis+dermis) from 2 adults AD patients and 2 PSO patients by high-throughput complementary DNA sequencing (RNA-seq). For the gene expression estimation, Cufflinks v2.1.1 was used that is the gene annotation database of Ensembl release 72.
