Egr-1 induced gene expression changes in HUVECs

ABSTRACT: The target genes of the transcription factor early growth response-1 (EGR-1) were studied in human umbilical vein endothelial cells (HUVEC). Densely cultured HUVEC were infected with recombinant adenoviruses expressing EGR-1 or empty control viruses at a MOI of 100. RNA was isolated from the cells at the time points 16 h, 24 h and 48 h post infection. Control cells without infection were cultured in parallel and harvested at the 24 h time point. Total RNA was isolated using TRIzol (InVitrogen) and Alater (Ambion) reagents and cDNA synthesis, hybridization and microarray analysis using U133 GeneChips performed according to the Affymetrix manual. Gene expression changes were calculated as the ratio of EGR-1 or control virus (CV) infected cells to non-infected control cells. Keywords: time-course

Egr-3, a member of early growth response family, is immediately and dramatically induced by VEGF in HUVEC, which regulates expression of many genes related to endothelial activation. Experiment Overall Design: Total 21 samples were derived from triplicate arrays of VEGF-treated si-Control-transfected cells and duplicate arrays of each of the VEGF-treated si-Egr-3-transfected cells.

A profibrotic gene expression program induced by Egr-1 in skin fibroblasts

Project description:Systemic sclerosis (SSc) is characterized by vascular damage, autoimmunity and fibrosis and is associated with highly variable clinical presentation and disease course. Aberrant transforming growth factor-ß (TGF-ß) signaling via the early immediate transcription factor Egr-1 is implicated in the pathogenesis of SSc. To shed light on the role of Egr-1 in fibrosis, regulation of gene expression in human skin fibroblasts overexpressing Egr-1 was examined by genome-wide expression analysis. Over 600 genes were found to be regulated by Egr-1. The Egr-1-responsive gene signature is largely comprised of genes involved in cell proliferation, TGF-ß signaling, wound healing, extracellular matrix synthesis and vascular development. Expression of the Egr-1 responsive genes was evaluated in a microarray dataset comprising skin biopsies from 17 patients with scleroderma and six healthy controls (GEO GSE9285; PMID 18648520). The “Egr-1 responsive gene signature” was enriched in the ‘diffuse-proliferation’ subset of skin biopsies in the patients with diffuse cutaneous SSc (dcSSc), but was not associated with other forms of scleroderma, or with healthy controls. Skin biopsies from patients with scleroderma can provide more insights into the relevant pathological processes in the subset of disease and could be developed into a diagnostic tool for identifying a subset of diffuse scleroderma patients who may be responsive to Egr-1 therapy. Cultures of primary fibroblasts from neonatal foreskin treated by Egr1 and Tgfb1 were measured at 24 and 48 hours. Control samples without any treatment were also measured at the same time points. Two biological replicates per condition/time point were measured using the Illumina HumanRef-8 V2 Expression BeadChip.

Transcription profiling of mouse Egr-1 knock-outs during development (p30 and p42)

Project description:In chicks, the avian homologue of the early growth response protein-1 (ZENK) has been shown to be increased in a special cell type of the retina, the glucagonergic amacrine cells, under conditions that lead to a reduction in eye growth (myopic defocus, recovery of myopia) and decreased under conditions that enhance ocular growth (hyperopic defocus, form-deprivation). The investigation of Egr-1 knock-out mice showed that homozygous knock-out mice with no functional Egr-1 protein developed relative axial myopia at the age of 42 and 56 days, compared to heterozygous- and wildtype Egr-1 knock-out mice. To clarify the role of Egr-1 in the retinal regulation of eye growth, and to get an idea about the biochemical pathways underlying this mechanism, we studied the role of Egr-1 in more detail using Affymetrix microarrays. Experiment Overall Design: Retinal samples of young homozygous Egr-1 knock-out and wildtype mice at the age of 30 days (hm30 and wt30; no difference in axial eye length yet) and 42 days (hm42 and wt42; already a difference in axial eye length of 59 Âµm) were taken to compare the mRNA expression changes over time between these two genotypes and within the same genotype between the two age groups.

Hepatic circadian gene expression in WT and liver specific Egr-1 knockout mice aged 2month, 6month and 12month

Project description:The circadian rhythms of older individuals are normally shifted, and the metabolic functions of these individuals are degraded. How the link between metabolism and circadian rhythms is decoupled in peripheral metabolic organs with age increased is still largely unknown. Here, we analyze hepatic transcriptome in the liver at zeitgeber time (ZT) 1, 5, 9, 13, 17, 21 from C57BL/6J aged 2month, 6month and 12month. Transcriptional changes suggest that the peripheral circadian system is shifted and that the rhythmic lipid metabolism is disrupted in the liver with aging. Furthermore, we figure out liver EGR-1 might integrate the central and peripheral rhythms and regulate metabolic homeostasis in the liver. Another hepatic transcriptome of WT and Egr-1-KO liver samples at EGR-1 highest(H) and lowest(L) zeitgeber time in 2month, 6month and 12month were analyzed. The results indicated that the rhythmic coupling of EGR-1 and CIDEA regulates age-related metabolic dysfunction in the mouse liver.
