Different gene expression in cisplatin chemo-resistant or sensitive of esophageal squamous carcinoma patients.

ABSTRACT: To investigate mRNA expression difference in cisplatin chemo-sensitivity in esophageal squamous carcinoma patients. We performed gene expression profiling analysis using data obtained from RNA-seq of 6 different patients.

Project description:In this study, esophageal squamous cell carcinoma (ESCC) patients’ endoscopic biopsy tumor tissues obtained before their concomitant chemo-radiation therapy (CCRT) were used to analyze the transcriptomes by using the RNA-sequencing approach. According to retrospective study of patients’ responses to CCRT, the treatment-naive specimen can be divided into two groups (16 good CCRT response and 12 poor CCRT response). This systematic profiling uncovered a gene set that is differentially expressed between the two patient groups.

MIcroRNA expression profiling of chemotherapy resistant esophageal adeno- and squamous cell carcinoma cell lines

Project description:Chemotherapy resistance adversely impacts the treatment of some individuals with esophageal cancer. Identifying chemotherapy resistance might help tailor clinical treatments. In this study the impact of microRNAs on chemotherapy resistance in esophageal cancer was investigated. We used microarrays to detail the global programme of microRNA expression underlying chemotherapy resistance and identified distinct classes of up-regulated microRNAs in generated chemotherapy resistant cell lines. An in-vitro model of acquired chemotherapy resistance in esophageal adeno- (EAC) and squamous cell carcinoma (SCC) cells was used, and microRNA expression profiles for cisplatin or 5-fluorouracil (FU) resistant variants vs. chemotherapy sensitive controls were compared using microarray. These results were further validated using qRT-PCR techniques (not shown in this submission).

Project description:Esophageal cancer is one of the most serious health problems globally because of its high incidence and mortality rate. Cisplatin (CDDP) is the primary treatment but limited by the drug resistance. However, the mechanism of cisplatin resistance remains unclear. Herein, the CDDP-resistant cell line TE-1/CDDP was established to compared with TE-1, the expression profiles of circRNAs mRNA and lncRNA were compared between TE-1/CDDP and TE-1 using microarray analysis, and used this as a basis for exploring possible mechanisms of drug resistance in genes such as HDAC9.

Proteomic profiling of cisplatin-resistant and cisplatin-sensitive germ cell tumor cell lines using quantitative mass spectrometry

Project description:Testicular germ cell tumors (GCTs) are the most common malignant solid neoplasms in men between the age of 15 and 40 with an increasing incidence seen over the last four decades. Histologically, type II GCT are divided into seminomas (SEM) and non-seminomatous GCT (NSGCT) according to the WHO. NSGCT comprise distinct subentities, e.g. embry-onal carcinomas (EC), yolk sac tumors, choriocarcinomas and teratomas (TER). The distinction between SEM and NSGCT is important because of varying treatment approaches, treatment responses and patients’ prognosis. The treatment of GCTs primarily comprises a radical inguinal orchiectomy of the affected testis. Subsequent cisplatin-based combination chemotherapy is required in metastatic GCTs. The introduction of cisplatin-based com-bination chemotherapy has led to cure rates of up to 90% even in metastatic disease stages. Teratomas are of particular interest, as they are uniformly cisplatin-resistant and sur-gery is the only successful therapy. The number of patients with recurring disease that finally fail several lines of platinum-based chemotherapy is about 3-5% of all GCT patients and about 15% of patients with primary metastatic disease, but they have an exceptionally poor prognosis with a life-expectancy of only a few months. In this study, we compared cisplatin-resistant and cisplatin-sensitive cell lineages of pluripotent NTERA-2 and NCCIT as well as the nullipotent 2102EP cells (with NCCIT deriving from a TP53 mutated mixed mediastinal GCT) by high-resolution mass spectrometric analy-sis (MS) combined with stable isotope labelling with amino acids in cell culture (SILAC). The NTERA-2 and NCCIT cell lines represent EC, which can develop in all other types of NSGCT. Both cell lines represent a very well-studied in vitro model for NSGCTs.

Expression profiling of cisplatin resistant cells derived from H460 lung cell line.

Project description:Combination of platinum-based chemotherapy and radiation is currently the standard treatment for locally advanced lung cancer patients. However, therapeutic resistance to these therapies may arise from the presence of cancer stem cells (CSCs). To investigate the CSCs hypothesis of chemo-radiation resistance, we used microarray assay to profile CSCs-like cisplatin-resistant lung cancer cells (CDDP-R) versus its parental cells. CDDP-R cells were established by exposing H460 lung cancer cells to 3¬µM cisplatin for 7 days, followed by 0.8% methylcellulose selection over 14 consecutive days.¬†We found that CDDP-R cells expressed higher levels of stem cell markers, including CD133 and ALDH. They are more resistant to cisplatin- and etoposide-induced apoptosis and to high radiation dose (20Gy). Clonogenic assays suggest that CDDP-R cells were more resistant to radiation than parental H460 cells (DER=1.21, p<0.01). Xenograft studies suggest that CDDP-R cells were more tumorigenic (p<0.001). Microarray and comprehensive protein interaction networks analyses revealed IGFBP3 as a highly ranked hub protein which plays an important role in the mechanism of cisplatin resistance. We found reduced level of IGFBP3 and enhanced IGFR-1 activation upon IGF stimulation in CDDP-R cells. The specific targeting of IGF-1R using siRNA resulted in significant sensitization of CDDP-cells (DER=1.17, p<0.05) to radiation compared with the parental H460 cells. Our findings suggest that CDDP-R cells have the characteristics of CSCs and constitute a ‚Äúsuitable‚Äù model to study lung CSCs. Evidence suggests that microRNAs are key players in the initiation, progression, and drug resistance of cancer cells. However, the precise miRNAs dysregulated and contributing to platinum resistance in ovarian cancer cells have not been fully elucidated. Here, we conducted a miRNA expression profiling of cisplatin-sensitive (A2780) and cisplatin-resistant (CP20 and CIS) ovarian cancer cells to identify potential miRNAs involved in platinum resistance.
