PRL-3 induces Kupffer cells to recruit MDSCs in liver through exosome ITGαvβ5

ABSTRACT: Previous studies have found that PRL-3 promotes liver metastasis of colon cancer, but whether it is involved in promoting liver PMN formation remains unclear.It was found that MDSC is the main cellular component of PMN formation and plays an immunosuppressive role.We found that PRL-3 can up-regulate the expression of ITGαvβ5 in exosomes of colon cancer cells through proteomics identification and extractor identification, and further verified that exosome ITGαvβ5 uptake by Kupffer cells can participate in inducing liver infiltration of MDSCs by exosome pre-education and mouse liver metastasis model construction,thus promoting liver metastasis of colon cancer. Then we extracted exosomes from colon cancer cells, induced Kupffer cells in vitro, and conducted RNA-seq sequencing to screen and identify genes involved in regulating MDSC infiltration.
