Project description:To understand the transcripts regulated by inflammation and how this regulation may differ between different individuals, the transcriptome of HUVECs stimulated with inflammatory mediators was examined in 9 different individuals. In the study present here, HUVEC isolates from 9 different individuals were cultured until passage 4 in fully supplemented growth conditions. Passage 4 HUVEC isolates were then treated with a cocktail of inflammatory mediators (10ng/ml TNF-α, Il-1β, Il-8) for a period of 24 hours prior to RNA extraction. RNA was extracted and hybridised onto CodeLink microarrays. The expression profiles in the inflammatory mediator treated condition were examined along side a superset of the same biological replicates that were untreated (also submitted), to facilitate assessment of the resting transcriptome.

RNA sequencing of HECTD3 knockdown during TNFalpha stimulation in primary HUVEC cells

Project description:We isolated and validated primary HUVECs from the neonatal umbilical cord vein, and performed genome-wide expression analysis to profile differentially expressed genes in control and HECTD3-KD HUVECs treated with or without TNFα for 2 hours. Compared with control HUVECs, 59 genes showed lower expression in HECTD3-KD HUVECs in response to TNFα. Most of them are NF-κB target genes, including adhesion molecules SELE (E-selectin), ICAM-1, and VCAM-1.

Gene expression profiling of HUVEC under basal (resting) conditions

Project description:To understand the biological variability that exists between different individuals, the resting (untreated) transcriptome from 15 different individuals was examined. Transcripts expressed in a bimodal manor across the 15 biological replicates were identified for further examination. In the study presented here, HUVEC isolates from 15 different individuals were cultured until passage 4 in fully supplemented growth conditions. RNA was extracted and hybridised onto CodeLink microarrays. The expression profiles in the untreated condition were examined along side a subset of the same biological replicates treated with a cocktail of inflammatory mediators (10ng/ml TNF-α, Il-1β, Il-8) for a period of 24 hours prior to RNA extraction (also submitted).

Modifications of the composition of the hyphal outerlayer of Aspergillus fumigatus affects HUVEC inflammatory and immune response

Project description:Aspergillus fumigatus is an angioinvasive fungal pathogen and the main etiologic agent causing invasive aspergillosis. Upon contact with human umbilical vein endothelial cells (HUVECs), this fungus induces cellular injury, inflammatory response and a pro-thrombotic phenotype. The pathogen molecules involved in this processes are however still unknown. A. fumigatus hyphae have been shown to produce, both in vivo and in vitro, an extracellular matrix composed of galactomannan, galactosaminogalactan and α-(1,3)-glucan. The deletion of the UDP-Galp mutase results in a mutant with a very sticky phenotype, due to the increase in the galactosaminogalactan content in the cell surface. The overexpression of galactosaminogalactan in this mutant was confirmed by immunofluorescence microscopy with a monoclonal antibody. Interestingly, the adhesion of ∆ugm1 germlings to HUVECs was significantly higher in comparison to the wild type strain. The ∆ugm1 adhesion capacity was followed by an increment in the cytokine secretion and tissue factor expression by infected endothelial cells. Using a shotgun proteomic approach with label-free quantification, we were able to search and compare pathways regulated in endothelial cells challenged with A. fumigatus, wild type and ∆ugm1. The deletion of the UGM1 gene in A. fumigatus induced changes in the endothelial cell response, maily related to immune and stress responses. In addition, the purified urea soluble fraction of galactosaminogalactan was able to induce TNF-α secretion by HUVECs and also regulate coincident pathways identified in the mutant interaction condition. This work describes for the first time the impact of the modifications of the hyphal surface galactosaminogalactan on the endothelial cell responses to A. fumigatus.

hOB Transcriptome after HUVEC-Cocultivation

Project description:Vascularization represents an important issue in bone development, fracture healing and engineering of artificial bone tissue. In the context of bone tissue engineering, it was shown that coimplantation of human primary umbilical vein endothelial cells (HUVECs) and human osteoblasts (hOBs) results in the formation of functional blood vessels and enhanced bone regeneration. Implanted endothelial cells do not only contribute to blood vessel formation, but also support proliferation, cell survival and osteogenic differentiation of coimplanted hOBs. These effects are partially mediated by direct heterotypic cell contacts. In a previous report we could show that cocultivated hOBs strongly increase the expression of genes involved in extracellular matrix (ECM) formation in HUVECs, suggesting that ECM may be involved in the intercellular communication between hOBs and HUVECs. The present study aimed at investigating whether comparable changes occur in hOBs. We therefore performed a microarray analysis of hOBs cultivated in direct contact with HUVECs, revealing 1121 differentially expressed genes. The differentially expressed genes could be assigned to the functional clusters ECM, proliferation, apoptosis and osteogenic differentiation. In summary, our data demonstrate that HUVECs provoke complex changes in gene expression patterns in cocultivated hOBs and that ECM plays and important role in this interaction.
