GTP-dependent regulation of heterochromatin fluctuations at subtelomeric regions in S. cerevisiae

ABSTRACT: Analysis of gene expression changes following deletion of SIR genes at subtelomeric region. A total of 4 samples were analyzed : Wild type (BY4742) strain, SIR2 deletion strain, SIR3 deletion strain, SIR4 deletion strain.

Analysis of a novel existence region of Sir3p in S. cerevisiae

Project description:In the budding yeast, HMR, HML, telomere and rDNA domain are known as a silencing region. Sir2 need to make it at rDNA and, HMR, HML and the telomere need to Sir2, Sir3, Sir4 complex to control internal gene repression. In this report, we found a newly Sir3 binding domain, CN domain (Chromosome New region) 1~14, by the ChIP on chip analysis on S.cerevisiae chromosome. In addition, we also performed ChIP on chip analysis with anti-Sir3 antibody using G1 phase synchronized cell to find Sir3 distribution difference of stage of cell cycle and we found CN15~CN25 which was G1 phase specific Sir3 binding region. Furthermore, we analyzed difference of gene expression at CN region in sir3 strain, and some regions did not change level of gene expression. In the conventional report, Sir3 had recruited by Sir2 and Sir4 on chromosome, but recruit of Sir3 was independent on Sir2 and Sir4 at some CN regions. These data suggested that we found a newly Sir3 function and Sir3 recruited system on chromosome.

Sir4 H-BRCT domain interacts with multiple phospho-proteins to regulate heterochromatic repression

Project description:In Saccharomyces cerevisiae, the Silent Information Regulator (SIR) proteins Sir2/3/4 form a complex suppressing transcription of genes at subtelomeric regions and the homothallic mating type (HM) loci. Here we identify a non-canonical BRCA1 C-terminal domain (H-BRCT) in Sir4 which is responsible for tethering telomeres to the nuclear periphery. We show that Sir4 H-BRCT and the closely related Dbf4 H-BRCT serve as selective phospho-epitope recognition domains that bind to a variety of phosphorylated target peptides. We present detailed structural information about the binding mode of established Sir4 interactors (Esc1, Ty5, Ubp10) and identify several novel interactors of Sir4 H-BRCT, including the E3 ubiquitin ligase Tom1. Based on these findings, we propose a phospho-peptide consensus motif for interaction with Sir4 H-BRCT and Dbf4 H-BRCT. Ablation of the Sir4 H-BRCT phospho-peptide interaction disrupts SIR-mediated repression and perinuclear localization. In conclusion, the Sir4 H-BRCT domain serves as a hub for recruitment of phosphorylated target proteins to heterochromatin to properly regulate silencing and nuclear order.

Spt3 and Spt8 are involved in the formation of a silencing boundary by interacting with TATA-binding protein

Project description:In Saccharomyces cerevisiae, a heterochromatin-like chromatin structure called the silencing region is present at the telomere as a complex of Sir2, Sir3, and Sir4. Sir2 is a histone deacetylase, and Sir3 and Sir4 promote spreading of the silencing region at the telomere. Histone acetyltransferases restrict the silencing region. Here, we show that Spt3 and Spt8 block the spread of silencing regions. Spt3 and Spt8 are members of the Spt-Ada-Gcn5-acetyltransferase (SAGA) complex, which has histone acetyltransferase activity. Although both Spt3 and Spt8 interact with telomere-binding proteins (TBPs), Spt3 had a greater effect on genome-wide transcription. Mutant analysis showed that TBPs play an important role in boundary formation, in particular the interaction between Spt3 and TBPs.

Sir2-family of histone deacetylase regulated gene expression in Candida glabrata

Project description:Sir2-family of NAD-dependent histone deacetylases are involved in gene transcription repression in yeast. The C. glabrata genes encoding members of this family are SIR2 (CAGL0K01463g), HST1 (CAGL0C05357g), HST2 (CAGL0L08668g), HST3 (CAGL0H08239g) and HST4 (CAGL0F05621g). SIR4 (CAGL0K11396g) encodes a factor that forms a functional complex with Sir2. In this experiment, the transcription profile of C. glabrata HST1 null, HST2 null, SIR2 null or SIR4 null strain individually comparing with that of wild type strain was determined.
