Project description:Though the rhesus monkey is one of the most valuable non-human primate animal models for various human diseases because of its manageable size and genetic and proteomic similarities with humans, proteomic research using rhesus monkeys still remains challenging due to the lack of a complete protein sequence database and sufficient proteomic information. In this project, proteomic profiling of multiple organ tissues, 9 male and 11 female were performed in an automated, high-throughput manner employing annotated UniProtKB human database. Based on the success of this alternative interpretation of MS data, the list of proteins identified from total 12 organs of male and female subjects will benefit future rhesus monkey proteome research.

Project description:Chikungunya virus (CHIKV) is a mosquito-borne alphavirus that has emerged as a significant public health concern due to its ability to cause severe arthralgia and occasionally fatal cases. The virus has been shown to infect a wide range of tissues, including the heart, liver, spleen, lungs, kidneys, and brain, leading to a spectrum of clinical manifestations.This study aims to investigate the impact of CHIKV infection on the transcriptome of various organs in rhesus monkeys and mice, providing insights into the molecular mechanisms underlying the pathogenesis of CHIKV in different host species.The transcriptome analysis revealed significant alterations in gene expression across the infected tissues in both species.

Project description:Type 2 diabetes mellitus (TM) is a severely metabolic disorder that affects above 10% worldwide population. Obesity is a major cause of insulin resistance and contributes to the development of TM. Liver is an essential metabolic organ that plays crucial roles in the pathogenesis of obesity and diabetes. However, the underlying mechanisms of liver in the transition of obesity to diabetes are not fully understood. Nonhuman primate (NHP) rhesus monkey is an appropriate animal for research of human diseases. Here, we first screened and selected three individual spontaneous and diabetic rhesus monkeys. Interestingly, the diabetic monkeys were obese with high BMI at beginning, but gradually lost their body weight during one-year observation. Furthermore, we performed a SILAC-based quantitative proteomics to identify proteins and signaling pathways with altered expression in the liver of obese and diabetic monkeys. Totally, 3509 proteins were identified and quantified, and of which 185 proteins displayed altered expression level. GO analysis revealed that the expression of proteins involved in fatty acids β-oxidation and galactose metabolism was increased in obese monkeys; while proteins involved in oxidative phosphorylation (OXPHOS) and branched chain amino acid (BCAA) degradation was upregulated in diabetic monkeys. In addition, we observed a mild impaired mitophagy and apoptosis in the liver of diabetic monkeys, suggesting a dysfunction of mitochondria and liver injury in the late onset of diabetics. Taken together, our liver proteomics may reveal a distinct metabolic transition from fatty acids β-oxidation in obese monkey to BCAA degradation in diabetic monkeys.

Project description:Embryonic stem cells (ESCs) may be able to cure or alleviate the symptoms of various degenerative diseases. However, unresolved issues regarding apoptosis, maintaining function and tumor formation mean a prudent approach should be taken towards advancing ESCs into human clinical trials. The rhesus monkey provides the ideal model organism for developing strategies to prevent immune rejection and test the feasibility, safety and efficacy of ESC-based medical treatments. Transcriptional profiling of rhesus ESCs provides a foundation for future pre-clinical ESC research using non-human primates as the model organism. In this research we use microarray, immunocytochemistry, real-time and standard RT-PCR to characterize and transcriptionally profile rhesus monkey embryonic stem cells. We identify 367 rhesus monkey stemness genes, we demonstrate the high level (>85%) of conservation of rhesus monkey stemness gene expression across five different rhesus monkey embryonic stem cell lines, we demonstrate that rhesus monkey ESC lines maintain a pluripotent undifferentiated state over a wide range of Pou5f1 (Oct-4) expression levels and we compare rhesus monkey, human and murine stemness genes to identify the key mammalian stemness genes. The supplementary tables list the genes that have been upregulated in each undifferentiated rhesus monkey embryonic stem cell line (GSM99998, GSM99999,GSM100000, GSM100001, GSM100002, GSM99965, GSM99966) in comparison analysis with the pooled differentiated embryonic stem cells (GSM99840). Supplemental Table 1 contains the comparison analysis for all 52,865 probe sets on the rhesus monkey gene chip, Supplemental Table 2 contains the rhesus monkey genes that were significantly upregulated (FC>3) in the ORMES-6 biological replicates, Supplemental Table 3 contains the rhesus monkey genes that were significantly upregulated (FC>3) in the pooled differentiated EBs and Supplemental Tables 4-8 represent genes that were significantly upregulated in ORMES 6A, 7, 9, 10 and 13 respectively. Project description:We report Transcription Activator-like Effector Nucleases (TALEN)-mediated mutagenesis of the Rett Syndrome gene, methyl-CpG binding protein 2 (Mecp2), in nonhuman primates (NHPs). Microinjection of Mecp2-targeting TALEN plasmids into rhesus and cynomolgus zygotes caused 40-50% of the embryos harboring mutations when examined beyond 8-cell stage. The surrogate pregnancy rates were 28.6% (2/7) and 16.7% (1/6) for rhesus and cynomolgus, respectively. The 2 rhesus surrogates went through miscarriage at 30- and 63/64-days of gestation. Analyses of two 63/64-day-old liter-mate embryos indicated both were male and carried MeCP2 mutations, yet chimeric. Mutation rate was higher in one embryonic brain than the other. RNA sequencing and anatomical analyses indicated that the brain with higher-degree mutations was underdeveloped, which, together with mid-gestation-stage male lethality likely recapitulate the disease. One live delivery of a female cynomolgus monkey occurred after 162 days of gestation, with abundant mutagenesis in peripheral tissues examined. We conclude that TALEN-mediated mutagenesis greatly facilitates disease modeling in NHPs.

Project description:We report the application of RNA sequencing technology for high-throughput profiling of RNA expression in monkey tissues. By obtaining over four billion bases of sequence of brain, liver and muscle from MCPH1 trasngenic monkey and two control monkeys. We find that brain tissue showed more differentially expressed genes compared with those in liver and muscle. Finally, we show that This result implies that the overexpressed huMCPH1 may have a global impact on multiple organs of the transgenic monkey, and it has the largest impact on the brain in terms of gene numbers.
