MicroRNA profiling of mammary gland and liver tissue of cows under different feeding conditions (control, restricted amount of milk replacer, milk replacer ad libitum)

ABSTRACT: To investigate the influence of the feed additive (milk replacer) on biological processes occuring in cow mammary parenchyma, mammary fat pad and liver via miRNA-mediated regulation. To this end, we performed microRNA expression profiling using next-generation sequencing.

Different Diets Change the Bovine Milk Extracellular Vesicle-miRNA Expression

Project description:Given that different diets could alter cow milk yield and composition, the effects of different feed formula on milk extracellular vesicle (EV) miRNAs were detected. Cow milk EVs contained various small RNAs, including miRNAs, snRNAs, tiRNAs, Cis-regulatory elements, and piRNAs. Two hundred and seventy-six known bos taurus miRNAs were identified by sequencing in bovine milk EVs. There were 13 immune-related miRNAs in the top 20 miRNAs in milk EVs. Nine differently expressed known miRNAs were detected in responding to different feed formulations. Cow milk EVs are abundant of small RNAs, especially miRNAs, which might be closely related to the development of maternal mammary gland and neonatal immune maturity.

Project description:Female Ames dwarf mouse (+/+ or +/df) with ad libitum access to water and standard pelleted food (LabDiet, PMI Feeds, Inc., St Louis, MO) caged with microisolator filter tops. Mice were killed between 24 to 26 months of age, tissues removed, rapidly frozen on dry ice, and stored in liquid nitrogen. Total liver RNA was isolated from frozen tissue as described (T. Tsuchiya, J.M. Dhahbi, X. Cui, P.L. Mote, A. Bartke, S.R. Spindler, Physiological Genomics, Submitted). mRNA levels were measured using the Affymetrix mouse U74Av2 array according to standard protocols. After hybridization, arrays were scanned using a Hewlett-Packard GeneArray Scanner. Image analysis was performed as described (Cao SX, Dhahbi JM, Mote PL, and Spindler SR. Secreted milk transcriptome from early lactation dairy cows

Project description:Our objective was to characterize the protein coding portion of the bovine mammary gland transcriptome by magnitude of RNA-seq read counts. EBseq analysis determined no differentially expressed genes due to postruminal lysine infusion, therefore, read counts were averaged across treatment and block. DAVID functional annotation analysis was utilized to determine specific gene ontologies and KEGG pathways to describe the data by magnitude of reads.
