ABSTRACT: Angiogenesis in collagen gel cultures of rat aorta begins with neovessels sprouting from the aortic explant within the first three days of culture. We used microarrays to detail the pattern of gene expression underlying initial 24 hours of growth, prior to the sprouting of visible neovessles, and identified distinct classes of up-regulated genes during this process.

Either freshly harvested aortic rings, representing day 0, or collagen gel cultures of rat aorta were grown in serum free medium and used to prepare total RNA.

We used microarrys to examine the effects of TNF-alpha on gene expression in both fibrin and collagen gels during the first 48 hours or culture. Rat aortic rings were cultured in either collagen or fibrin maticies. Half of the cultures from each matrix group were treated with 10ng/ml recombinant rat TNF-alpha, and half were left untreated. AGM hematopoietic stem cells are differentially regulated by BMP and Hedgehog signalling pathways

Project description:The first HSCs are produced in the aorta-gonadmesonephros (AGM) region of the embryo through endothelial to a hematopoietic transition. BMP4 and Hedgehog affect their production/expansion, but it is unknown whether they act to affect the same HSCs. In this study using the BRE GFP reporter mouse strain that identifies BMP/Smad-activated cells, we find that the AGM harbors two types of adult-repopulating HSCs upon explant culture. Embryonic day 11 AGM are dissected and either analyzed directly, or after explant culture in conditions containing BMP/Hedgehog with or without cyclopamine. Transcription profiling of mouse dorsal aorta derived mesangioblast clone comparison

Project description:Mouse dorsal aorta from E9.5 mouse embryos (C57BL6) was grown as an explant culture as described elsewhere (De Angelis et al., 1999). After 5 days the explant was dissociated into a single cell suspension, and plated at limiting dilution on a feeder layer of Mitomycin C-treated primary embryonic mouse fibroblasts, in 96 multiwell plates in complete medium, RPMI 20% Fetal Calf Serum. After 1 week, mesangioblast clones appeared approximately 2-4% of the wells. When the clones had grown to approximately 103 cells they were passed on gelatin-coated dishes, and further expanded. Clones D16 and D351 were used for microarray analysis.
