An adipokine derived from subcutaneous adipose tissue

ABSTRACT: Adipokines are proteins secreted by the adipose tissue and are associated with obesity-related metabolic disorders; however, most studies have focused on adipokines expressed by visceral adipose tissue. This study aimed to identify the adipokine potentially derived from subcutaneous adipose tissue and its clinical significance.

LRG1 is an adipokine that promotes insulin sensitivity and suppresses inflammation

Project description:While dysregulation of adipocyte endocrine function plays a central role in obesity and its complications, the vast majority of adipokines remain uncharacterized. We employed bio-orthogonal non-canonical amino acid tagging (BONCAT) and mass spectrometry to comprehensively characterize the secretome of murine visceral and subcutaneous white and interscapular brown adipocytes. Over 600 proteins were identified, the majority of which showed cell type-specific enrichment. We here describe a metabolic role for leucine-rich α-2 glycoprotein 1 (LRG1) as an obesity-regulated adipokine secreted by mature adipocytes. LRG1 overexpression significantly improved glucose homeostasis in diet-induced and genetically obese mice. This was associated with markedly reduced white adipose tissue macrophage accumulation and systemic inflammation. Mechanistically, we found LRG1 binds cytochrome c in circulation to dampen its pro-inflammatory effect. These data support a new role for LRG1 as an insulin sensitizer with therapeutic potential given its immunomodulatory function at the nexus of obesity, inflammation, and associated pathology.

Project description:In addition to total body fat, the regional distribution and inflammatory status of enlarged adipose tissue are strongly linked to metabolic and cardiovascular complications of obesity. We recently showed that the severity of liver non-alcoholic histopathology in obese subjects increased with the amounts of macrophages in visceral adipose tissue (VAT), while no relation was found with the subcutaneous adipose tissue (SAT). In the abdominal region, SAT is anatomically divided into superficial (sSAT) and deep (dSAT) layers. The aim of the present study was to test the hypothesis that these distinct compartments differentially contribute to hepatic alteration in obesity. Whole tissue mRNA expression from severely obese individuals: subcutaneous and visceral adipose tissues

Project description:Subcutaneous adipose tissue and visceral adipose tissue samples were obtained from severely obese individuals that underwent bariatric surgery. The goal of this study was to compare genome-wide gene expression levels in the two tissue types from healthy and unhealthy severely obese individuals. Whole-transcriptome subcutaneous adipose tissue gene expression levels were determined in 73 individuals with a BMI >35 kg/m2. Whole-transcriptome visceral adipose tissue gene expression levels were determined in 69 individuals with a BMI >35 kg/m2. Modules of co-expressed genes likely to be functionally related were identfied and correlated with BMI, plasma levels of glucose, insulin, HbA1c, triglycerides, non-esterified fatty acids, ALAT, ASAT, C-reactive protein, and LDL- and HDL cholesterol.

LC-MS/MS analysis of visceral and subcutaneous adipose tissue proteomes in young goats with focus on innate immunity and inflammation related proteins

Project description:The aim of this study was to undertake an in-depth and comparative study of the protein expression patterns of subcutaneous and visceral adipose tissues in goats, combined to an mRNA expression study of proteins involved in immune and inflammatory response. Samples were obtained from four healthy goat-kids, Alpine breed, naturally reared by their mothers. Animals were slaughtered at the age of 30 days, during routinely slaughtering procedures, and four different adipose tissues were collected from each animal. Subcutaneous fat was taken from sternum and base of the tail; visceral fat was taken from perirenal and omental depots. The four adipose tissues deposits were selected due to their frequent use in experimental studies on fat tissue. Liver samples were also collected, and used as reference samples. Tissue samples were snap frozen in liquid nitrogen and stored at –80 degrees C until analysis. The 20 runs included in this dataset are from the 4 animals x 5 tissues (sternum, base of the tail, perirenal area, omentum and liver). A 2-D label free LC-MS/MS approach followed by cluster analysis was used for comparing the subcutaneous and visceral fat tissue proteomes. Each sample was analysed in ~12 SCX fractions which correspond to the WIFF files included in each RAW_GOAT0xx.zip files.
