Evaluating the effects of polyoma small T antigen on gene expression

ABSTRACT: We developed an inducible degradation system for murine polyoma small T antigen (PyST) to study the effects of PyST on gene expression. We examined changes in expression following ectopic expression of tagged antigen and following their degradation in the MCF10A human mammary epithelial cell line.

Small RNA sequencing of murine pancreatic tumors driven by polyoma middle-T antigen with wild-type or single-copy loss of Dicer

Project description:We report the small RNA sequencing results of mouse pancreatic tumor cell lines driven by polyoma middle-T antigen, which have additionally been engineered to have either wild-type (herein "PDwt") or single-copy loss (herein "PDhet") of the Dicer gene. As expected, PDhet cell lines exhibit lower miRNA levels compared to PDwt cell lines. Principle component analysis, detailed in the published manuscript, reveals PDwt cell lines to be more widely varying in miRNA signature compared to PDhet cell lines. Grouping miRNAs by family reveals several families to be significantly downregulated in PDhet cells lines below 50% of PDwt levels, including miR-10, -148, -17, -28, -299, -30, -34, and -379.

Evaluating the effects of PTEN loss-driven cell transformation

Project description:We developed a unique in vitro transformation model utilizing PTEN CRISPR in the MCF10A human mammary epithelial cell line to study PTEN loss-induced transformation and profiled genome-wide expression in these cells to understand how PTEN loss alters the transcription landscape. We also examined changes in gene expression following inhibition of the p110b catalytic subunit of PI3 kinase, which is activated in the transformed cells. In addition, we profiled gene expression in MCF10A cells ectopically expressing PD-L1 (CD274), which is suppressed in cells harboring PTEN CRISPR.

Genome-wide characterization of the transcriptional program of Myc-dependent transformation

Project description:We profiled Myc binding in a normal breast cell-line (MCF10A) under basal conditions after ectopic expression of Myc. We showed that ectopic Myc expression increases tumour formation in vivo and in vitro. We then profiled genome-wide Myc binding using Agilent promoter arrays in MCF10A cells expressing wild-type and ectopic Myc. We show that Myc binds to a greater number of spots in wild-type than in Myc cells, but that some targets are unique to each condition. Genetic deletion or pharmacologic blockade of the amino acid transporter Slc6a14 in mice suppresses breast cancer induced by Polyoma middle T oncogene

Project description:Tumor cells have an increased need for amino acids. Mammalian cells cannot synthesize essential amino acids; they must obtain these amino acids via specific transporters. Glutamine, though a non-essential amino acid, is critical for tumor cells (glutamine addiction). Entry of amino acids into tumor cells is enhanced by upregulation of specific transporters. If the transporters that are specifically induced in tumor cells are identified, blockade of the induced transporters would constitute a logical strategy for cancer treatment. The transporter SLC6A14 is unique and transports all essential amino acids as well as glutamine and is expressed only at low levels in normal tissues, but induced in colon cancer and in ER+ breast cancer. We have now established the potential of this transporter as a drug target for breast cancer treatment using genetic and pharmacologic approaches. We then examined the progression of breast cancer in Polyoma middle T antigen (Py-MT) Tg mouse on Slc6a14+/+ and Slc6a14-/- background using microarray analysis.

Consequences of sEH deletion in the polyoma middle T (PyMT) breast cancer tumor proteome

Project description:The aim of this investigation was to study the consequences of interfering with soluble epoxide hydrolase (sEH) expression on tumor growth and metastasis in genetically modified animals that spontaneously generate tumors without the exogenous application of high concentrations of epoxide mediators or inhibitors. Therefore, breast cancer development was studied in mice expressing the polyoma middle T oncogene (PyMT) under the control of the mouse mammary tumor virus promoter, to induce spontaneous mammary tumors. To facilitate the study of endogenous sEH activity in tumor growth, PyMT mice were then crossed with sEH-/- mice to generate sEH-deficient mice that spontaneously generate breast tumors (so called PyMTsEH mice). For these analyses, primary tumors were removed from 20 week old mice.
