Project description:A MIBC patient with aggressive disease course was selected for ex vivo model establishment and growth dynamic studies. After standard neoadjuvant treatment with gemcitabine and cisplatin, cystectomy was performed, and excised urothelial carcinoma tissue was implanted subcutaneously into male SCID mice to generate CoCaB1 PDXs. Established PDXs were passaged subcutaneously in SCID mice to generate early and late passage models for functional and molecular characterization. Representative early and late passage PDXs were collected for organoid establishment.

Project description:In this paper, a GelMA/TPGS@Tea Polyphenols (TP) bioactive hydrogel system was successfully prepared. The system utilizes the injectability and mechanical support of GelMA hydrogel, combined with the temperature-sensitive gel-forming property of TPGS and the specific inhibitory effect of tea polyphenols (TP) on NLRP3 inflammatory vesicles, with the aim of solving the problem of induced bridge formation and deformity after growth plate injury in pediatric orthopaedic clinics. In vitro and in vivo experiments showed that the hydrogel system could rapidly gel and fill the defects at physiological temperature (37°C), and the TP component significantly slowed down CASP1-mediated chondrocyte pyroptosis by inhibiting the activation of NLRP3 inflammasome, thus preventing the occurrence of bone bridges. Our study provides a bioactive, injectable, and NLRP3-targeted filler material for growth plate injury repair in pediatric orthopedics, which has great potential for preventing bone bridge formation!.

Project description:Bone defect repair remains a major clinical challenge. This study presents a novel strategy using a 3D-printed piezoelectric hydrogel scaffold—composed of gelatin methacrylate (GelMA), hydroxyapatite (HA), and barium titanate (BTO)—for functional bone tissue engineering. The GelMA/HA/BTO (GelHABT) scaffold exhibited a well-defined porous structure, enhanced mechanical stability, and, crucially, reliable piezoelectric responsiveness. This key feature enables the material to convert external mechanical stimuli, such as low-intensity pulsed ultrasound (LIPUS), into endogenous electrical signals. In vitro, the scaffold promoted BMSCs adhesion, proliferation, and osteogenic differentiation, with performance significantly enhanced under LIPUS stimulation. Mechanistic insights revealed that the piezoelectric microenvironment remodeled the cellular miRNA expression profile, particularly up-regulating osteogenesis-related miR-29b-3p and activating the AMPK signaling pathway. Collectively, this ultrasound-responsive, gene-regulating scaffold represents a promising approach for treating bone defects by leveraging piezoelectricity to actively stimulate bone regeneration.

Project description:Caloric restriction (CR) without malnutrition is one of the most consistent strategies for increasing mean and maximal lifespan and delaying the onset of age-associated diseases. Stress resistance is a common trait of many long-lived mutants and life-extending interventions, including CR. Indeed, better protection against heat shock and other genotoxic insults have helped explain the pro-survival properties of CR. In this study, both in vitro and in vivo responses to heat shock were investigated using two different models of CR. Murine B16F10 melanoma cells treated with serum from CR-fed rats showed lower proliferation, increased tolerance to heat shock and enhanced HSP-70 expression, compared to serum from ad libitum-fed animals. Similar effects were observed in B16F10 cells implanted subcutaneously in male C57BL/6 mice subjected to CR. Microarray analysis identified a number of genes and pathways whose expression profile were similar in both models. These results suggest that the use of an in vitro model could be a good alternative to study the mechanisms by which CR exerts its anti-tumorigenic effects. Project description:Tissue engineering strategies that combine human pluripotent stem cell-derived myogenic progenitors (hPDMs) with advanced biomaterials provide promising tools for engineering 3D skeletal muscle grafts to model tissue development in vitro and promote muscle regeneration in vivo. We recently demonstrated (i) the potential for obtaining large numbers of hPDMs using a combination of two small molecules and (ii) the application of electrospun fibrin microfiber bundles for functional skeletal muscle restoration following volumetric muscle loss. In this study, we demonstrate that the biophysical cues provided by the microfiber bundles can induce unsorted hPDMs to form multinucleated myotubes that express desmin and myosin heavy chain. To reduce the batch-to-batch variability of these samples, we tested a genetic PAX7 reporter line (PAX7::GFP) to sort for more homogenous populations of hPDMs. RNA sequencing and gene set enrichment analyses confirmed that PAX7::GFP-sorted hPDMs exhibited higher expression of myogenic genes while unsorted hPDMs demonstrated increased expression of genes associated with embryonic, neural crest, and fibroadipogenic progenitor lineages. We tested engineered skeletal muscle grafts derived from either PAX7::GFP-sorted or unsorted hPDMs within in vivo skeletal muscle defects. The unsorted hPDM-derived grafts also exhibited greater fibrosis and more proinflammatory responses. In contrast, the PAX7::GFP-sorted groups had moderately high vascular infiltration, more implanted cell association with embryonic myosin heavy chain (eMHC) regions, and greater CD206:CD86 ratios compared to unsorted hPDMs and acellular fibers suggesting they induced more pro-regenerative microenvironments. These findings demonstrated some promise for the use of PAX7::GFP-sorted hPDMS on fibrin microfiber bundles and provided some insights for improving the cell-biomaterial system to stimulate more robust in vivo skeletal muscle regeneration.
