Project description:Sprobolus virginicus is a halophytic C4 grass found in worldwide from tropical to warm temperate regions. A Japanese genotype showed a salinity tolerance up to 1,500 mM NaCl, a three-fold higher concentration than seawater salinity. To identify key genes involved in the regulation of salt tolerance in S. virginicus, random cDNA libraries were constructed from salt-treated leaves, and were introduced into Arabidopsis for salt tolerant plant screening. Eight independent transgenic lines were found to be more salt tolerant than wild type from the screen of 3011 lines on the medium containing 175 mM NaCl. Among the selected lines, two contained cDNAs encoding glycine-rich RNA-binding proteins (GRPs). To identify transcriptomic change in the GRP-transgenic line, we performed microarray analysis of the transgenic line and WTunder salt stress.

Project description:Wild halophytic tomato has long been considered as an ideal gene donor for improving salt tolerance in tomato cultivars. Here, a wild tomato genotype, Solanum pimpinellifolium ‘PI365967’ is significantly more salt-tolerant than a cultivar, Solanum lycopersicom ‘moneymaker’. Affymetrix Tomato Genome Arrays was used to compare the transcriptome change of PI365967 and Moneymaker by salt treatment.After treatment with 200 mM NaCl for 5 h, PI365967 showed relatively fewer responsive genes compared to Moneymaker. Salt Overly Sensitive (SOS) pathway was found to be more active in PI365967 than in Moneymaker, coinciding with relatively less accumulation of Na+ in shoots of PI365967. A gene encoding salicylic acid-binding protein 2 (SABP2) was induced by salinity only in PI365967, suggesting a possible role of salicylic acid signaling in salt response of PI365967. The fact that two genes encoding lactoylglutathione lyase were salt-inducible only in PI365967, together with much higher basal expression of several glutathione S-transferase genes, suggested a more effective detoxification system in PI365967. In one treatment, 9 six-leaf-old plants of one tomato genotype (PI365967 or Moneymaker) were divided into three biological replicates and treated hydroponically under sterile condition with (or without) 200 mM NaCl for 5 h. RNA isolated from pooled three individual plants was used in hybridization to one chip, resulting in 12 chips in total.

Project description:Salinity stress poses a significant risk to agricultural yield and productivity. Therefore, elucidation of plant salt-response mechanisms has become essential to identify stress-tolerance genes. In the present study, two pearl millet genotypes with contrasting salt-tolerance showed differential morpho-physiological and proteomic responses under 150 mM NaCl, and the genotype IC 325825 could withstand salt-stress better than IP 17224. The salt-tolerance potential of IC 325825 was associated with its ability to maintain ionic, osmotic balance and membrane integrity under stress. The IC 325825 exhibited better growth under salinity as compared to IP 17224 due to higher expression of C4 photosynthesis enzymes, efficient antioxidant system, and lower Na+/K+ ratio. Comparative proteomics analysis revealed greater metabolic perturbation in IP 17224 under salinity, in contrast to IC 325825 that harbored pro-active stress-responsive machinery, allowing its survival and better adaptability under salt-stress. The differentially expressed proteins were in-silico characterized for their functions, subcellular-localization, pathway/ interaction analysis, and relative transcript levels. This study has provided novel insights into salinity stress adaptive mechanisms in pearl millet, demonstrating the power of proteomics-based approaches. The critical proteins identified in the present study could be further explored as potential objects for increasing salt-tolerance in sensitive crop plants.

Project description:Rice (Oryza sativa) stands among the world's most important crop species and is salt-sensitive. The undue accumulation of sodium ions (Na+) in shoots has the strongest negative correlation with rice productivity under long-term salinity. The plasma membrane Na+/H+ exchanger protein SOS1 is the only Na+ efflux transporter that has to date been genetically characterized and only in dicot plants. Here, the importance of Na+ fluxes governed by the SOS system in the salt tolerance of rice was analyzed by a reverse-genetics approach. A sos1 loss-of-function mutant displayed exceptional salt sensitivity that correlated with excessive Na+ intake and impaired Na+ loading into the xylem. Thus, SOS1 controls net Na+ uptake by roots and the long-distance transport to shoots. The acute Na+ sensitivity of sos1 plants at low NaCl concentrations allowed the inspection of the transcriptional response to sodicity stress, without interference by the osmotic challenge intrinsic to high salinity treatments. The transcriptional response to salt of the sos1 mutant roots involved the preferential down-regulation of stress-related genes compared to the wild-type despite the greater intensity of the stress imposed to the mutant, which suggested impaired stress detection or inability to mount a comprehensive response to salinity.

Project description:The global emergence of soil salinization poses a serious challenge to many countries and regions. γ-Aminobutyric acid (GABA) is involved in systemic regulation of plant adaptation to salt stress, but the underling molecular and metabolic mechanism still remains largely unknown. The elevated endogenous GABA level by exogenous application of GABA could significantly improve salt tolerance in creeping bentgrass with the enhancement of antioxidant capacity, photosynthetic characteristics, osmotic adjustment (OA), and water use efficiency. GABA strongly upregulated transcript levels of AsPPa2, AsATPaB2, AsNHX2/4/6, and AsSOS1/20 in roots involved in enhanced capacity of Na+ compartmentalization and mitigation of Na+ toxicity in cytosol. Significant downregulation of AsHKT1/4 expression could be induced by GABA in leaves in relation to maintenance of significantly lower Na+ accumulation and higher K+/Na+ ratio. GABA-depressed aquaporins (AQPs) expression and accumulation induced declines in stomatal conductance and transpiration, thereby reducing water loss in leaves during salt stress. For metabolic regulation, GABA primarily enhanced sugars and amino acids accumulation and metabolism largely contributing to improved salt tolerance through maintaining OA and metabolic homeostasis. Other major pathways could be responsible for GABA-induced salt tolerance including increases in antioxidant defense, heat shock proteins, dehydrins, and myo-inositol accumulation in leaves. Integrative analyses of molecular, protein, metabolic, and physiological changes reveal systemic function of GABA on regulating ions, water, and metabolic homeostasis in non-halophytic creeping bentgrass under salt stress.

Project description:Salinity seriously reduces the yield and quality of crops. Silicon (Si) has been widely reported to have beneficial effects on plant growth and development under salt stress. However, the mechanism is still poorly understood. In an attempt to identify genes or gene networks that may be orchestrated to improve salt tolerance of cucumber plants, we profiled the RNA-seq transcriptomes of both control and salt-stressed cucumber leaves in the presence or absence of added Si. The comparative transcriptome analysis revealed that Si plays an important role in shaping the transcriptome of cucumber: the expression levels of >1,000 genes (differtially expressed genes, DEGs) were changed in response to Si treatment as compared with the control, and these genes were mainly involved in ion transport, hormone and signal transduction, biosynthesis and metabolic processes, stress and defense responses, and antioxidant activity. Under salt stress, many genes functionally associated with metabolic processes and responses to environmental stimuli were strongly up- or down-regulated in their expressions. Si treatment showed a tendency that induced the transcriptomic profile of salt-stressed cucumber back to that of the control with large majority of Na down-regulated DEGs and about half of Na up-regulated DEGs being adjusted back to CT levels. This study provides a novel insight into the mechanism for Si-mediated alleviation of salt stress in plants at the transcriptome level, and it suggests that Si may act as an elicitor to precondition cucumber plants and induce salt tolerance.

Project description:Kharchia local is an Indian tall landrace wheat cultivar. It is native to sodic-saline soils of Kharchia tehsil of the Pali district of Rajasthan, and is a line developed from selections from farmer's fields. It is the most salt tolerant wheat genotype found in India. No systematic study has been carried out in this direction so far. The gaps in understanding of the mechanism underlying salt tolerance limit our ability to improve the salt tolerance in other crop plants. Transcriptome analysis of Kharchia Local under salt stress will provide the insight into the genes involved in salinity tolerance.

Project description:Both barley (Hordeum vulgare) and rice (Oryza sativa) belong to Poaceae family, but differ greatly in salt tolerance. In order to understand molecular mechanisms in the difference of salt tolerance between the two species, the responses of transcriptomic profiles to salt stress were compared between rice (cultivar Nipponbare) and barley (accession XZ26) to reveal how alternative splicing (AS) coordinates with transcriptional regulation in adaptation to salt stress. Physiological study showed that XZ26 had higher salt tolerance than Nipponbare, as reflected by less growth inhibition, lower shoot Na+ concentration and higher K+/Na+ ratio when exposed to salt stress. Transcriptomic analysis showed that XZ26 had higher ROS scavenging ability, less degradation of protein kinases and enhanced anti-oxidation. Moreover, AS genes related to ion transporter genes and transcription factors could enhance and amplify K+/Na+ homeostasis and signal transduction cascades. We proposed that higher salt tolerance of barley accession XZ26 is attributed to its superior K+/Na+ homeostasis, tissue detoxication and less energy consumption. The present results provide insights at transcriptomic levels into reasons why barley has higher salt tolerance than rice.

Project description:Soil salinity is one of the major factors that limits area of cultivable land and yield potential of crops. Considered as a moderately salt sensitive economically-important crop, biological processes involved in salt stress response in peanut remain elusive. Publishing of the genomic sequence of cultivar peanut represent a new opportunity for further research. ABA INSENSITIVE 4 (ABI4) is an evolutionary conserved transcription factor. Mutation of ABI4 in Arabidopsis enhanced salt tolerance of seedlings significantly by targeting and down-regulating of HKT1;1. However, few achievements were reported in other aspects. In this study, AhABI4s in peanut were first cloned and silenced via virus-induced gene silencing method. Phenotype analysis showed that down-regulation of AhABI4s enhanced salt tolerance of peanut significantly. According to proteome and phosphoproteome analysis, expression of 1,900 proteins and 2,620 phosphorylation sites were predicted to be affected by silencing of AhABI4s under salt stress. Proteins with ion transporting activity were enriched by GO analysis. Further analysis showed that, 31 proteins may participate in homeostasis maintain of Na+, K+, Ca2+, H+ and Cl- in plant cells. Combined transcriptome and proteome analysis indicated that 63 genes may regulated by AhABI4s directly and 7 of them, ABI5-like5, RABA1f, SCAMP3, PK, RP-S26e, HSP70, cysteine proteinase inhibitor, were able to interact with the predicted ion transporters. Silencing of AhABI4s altered down-stream protein-protein interaction network to regulate expression/phosphorylation level of ion transporters, and finally influence homeostasis under salt stress. This work provides novel insights for salt response mechanism research and offer important evidence for protein function study.
