Project description:High levels of GLI (GLI1 and GLI2) mRNA and GLI luciferase reporter activity were detected in the androgen independent prostate cancer cell lines DU145 and PC-3 compared to the androgen-dependent LNCaP prostate cancer cell line. Subsequently, we observed that ectopic GLI1 promoted hormone independence in LNCaP cells (LNCaP-GLI1). We compared the gene expression profile of LNCaP-pBP (empty vector), LNCaP-GLI1, DU145, and PC-3 cells globally as well as to identify GLI1-regulated genes that may contribute to hormone independence. Project description:Bisphenol A (BPA) analogs, bisphenol B (BPB) and bisphenol AF (BPAF) have been widely detected in the environment and human products with increasing frequency. However, uterine health risks caused by BPBBisphenol A (BPA) analogs, bisphenol B (BPB) and bisphenol AF (BPAF) have been widely detected in the environment and human products with increasing frequency. However, uterine health risks caused by BPB and BPAF exposure need to be further elucidated. The study aimed to explore whether BPB or BPAF exposure will induce adverse outcomes in uterus. We then performed gene expression profiling using data obtained from mouse uterus exposed to BPB and BPAF at 28 days.

Microarray Analysis of DU 145, LNCaP and PC-3 Prostate Cancer Cell Lines upon Docetaxel treatment

Project description:We measured the effect of docetaxel treatment to three differentially responsive prostate cancer cell lines, LNCaP, DU145 and PC-3, based on a transcriptional time course response by microarray analysis. These cell lines represent both androgen independent (DU145 and PC-3) and androgen sensitive (LNCaP) cells Hybridized arrays were scanned with Agilentâs dual laser-based scanner. Feature Extraction software version 10.5 (Agilent Technologies) was used to link a feature to a design file and to determine the relative fluorescence intensity between two samples. Dye swap strategy with alternate cy3 and cy5 labeling on docetaxel treated and control groups over four time points was used to have technical replicates and decrease dye bias.

Project description:Prostaste cancer (PC) is the most commonly diagnosed male malignancy and an important cause of mortality. Androgen signaling plays a key role in PC pathogenesis and the understanding of the mechanisms involved in the regulation of AR function might pave the ground for the development of more efficacious and long-lasting therapeutic strategies for PC patients. DNA/RNA helicases are emerging as important regulators of many cellular processes. Among them, DHX9 has been implicated in the control of genomic stability, transcription and DNA replication. To investigate the role of DHX9 in PC we perform RNA-sequencing analysis in androgen-sensitive LNCaP cells affects upon DHX9 silencing.

Mapping of DHT-responsive or -independent AR-binding sites induced by activated Src in prostate cancer cell lines [ChIP-seq]

Project description:Building on the observation that metastatic, castration-resistant prostate cancer (CRPC) correlates with activation of Src-family tyrosine kinases, we showed that the expression of activated Src renders LNCaP androgen-independent. Here, we report on RNA-seq and/or AR ChIP-seq analyses of LNCaP, LNCaP[Src], VCaP, 22Rv1 cells grown in the presence or absence of 10 nM DHT for 16h, or LuCaP35.1 tumors grown in androgen-supplemented vs. castrated mice (androgen-dependent vs. castration-resistant). We identify an 11-gene Src-induced signature found only in CRPC in response to DHT, and moreover, the differentail expression of a subset (DPP4, BCAT1, CNTNAP4, CDH3) correlates with earlier PC metastasis onset and poorer survival.

SWATH proteomic profiling of prostate cancer cells identifies NUSAP1 as a biomarker of response to Galiellalactone

Project description:Galiellalactone (GL) is a fungal metabolite that presents antitumor and anti-inflammatory activities in vitro and in vivo. Previous studies have shown that GL targets NF-KB and STAT3 pathways and induces G2/M cell cycle arrest in androgen-insensitive prostate cancer cells. In this study, we show that GL-induced cell cycle arrest is independent of the NF-KB and STAT3 pathways in DU145 and PC-3 cells, and also that GL did not affect cell cycling in androgen-sensitive prostate cancer cell such as LNCaP and 22Rv1 cells. In addition, we showed confluence resistance to GL in DU145 cells. Using a SWATH proteomic approach we identified the down-regulation of Nucleolar and spindle associated protein 1 (NUSAP1) under DU145 confluence and in LNCaP cells. Also, the inhibition of NUSAP1 by siRNAs induced resistance to GL in DU145 cells, suggesting that NUSAP1 may be a target for GL and could be useful as biomarker for responsiveness of the antitumor activity of GL.

Expression data from prostate cancer cell lines

Project description:We have investigated Perineural invasion (PNI) and epithelial-mesenchymal transition (EMT) related gene expression profiles of PC cell lines. In this dataset, we include the expression data obtained from laminin binding O-glycan high or low androgen-independent PC cells. These data are used to obtain 70 genes that are differentially expressed in response to cell migraton signal via laminin receptors. 4 Total samples were analyzed. We generated the following pairwise comparisons using GeneSpring Gx Version12.6: PC3-H vs PC3-L; LNCaP vs LNCaP-AI. Genes with an FDRâ¤10% and a fold-change â¥2 were selected.
