Translational Insights from Cell-type variation across Amygdala Subnuclei in Rhesus Monkeys and Humans

Project description:Theories of amygdala function are central to our understanding of psychiatric and neurodevelopmental disorders. However, our limited knowledge of the molecular and cellular composition of the amygdala impedes translational research aimed at developing new treatments and interventions. Here, we use single-nucleus RNA-sequencing from multiple amygdala subnuclei in both humans (n=3, Male) and rhesus macaques (n=3, Male) to begin to bridge the gap between preclinical rodent models and human disorders. Our results reveal substantial heterogeneity between regions, even when restricted to inhibitory or excitatory neurons. Consistent with prior work, our data highlight the complexities of individual marker genes for uniquely targeting specific cell-types. Cross-species analyses suggest the rhesus monkey model to be well-suited to understanding the human amygdala, but also identify limitations. For example, we identified a cell-cluster in the ventral lateral nucleus (vLA) of the amygdala that is enriched in humans, relative to rhesus macaques. Additionally, we identify specific cell-clusters with relative enrichment of disorder-related genes. These analyses point to the human-enriched vLa cell-cluster as relevant to ASD, potentially highlighting a vulnerability to neurodevelopmental disorders that has emerged in recent primate evolution. Further, we identified a cluster of cells expressing markers for intercalated cells (ITCs) that is enriched for genes reported in human genome-wide association studies of neuroticism, anxiety disorders, and depressive disorders. Together, these data shed light on the composition of the amygdala and identify specific cell-types that can be prioritized in basic science research to better understand human psychopathology and guide the development of potential treatments

Interhemispheric differences in gene expression of the cerebral cortex in humans and macaque monkeys

Project description:Handedness and language are two well-studied examples of asymmetrical brain function in humans. Approximately 90% of humans exhibit a right-hand preference, and the vast majority shows left-hemisphere dominance for language function. Although genetic models of human handedness and language have been proposed, the actual gene expression differences between cerebral hemispheres in humans remain to be fully defined. In the present study, gene expression profiles were examined in both hemispheres of three cortical regions involved in handedness and language in humans and their homologues in rhesus macaques: ventrolateral prefrontal cortex, posterior superior temporal cortex (STC), and primary motor cortex. Although the overall pattern of gene expression was very similar between hemispheres in both humans and macaques, weighted gene correlation network analysis revealed gene co-expression modules associated with hemisphere, which are different among the three cortical regions examined. Notably, a receptor-enriched gene module in STC was particularly associated with hemisphere and showed different expression levels between hemispheres only in humans.

LFQ quantitative analysis of 6 rhesus macaques prefrontal cortex samples from Shanghai Ninth People's Hospital

Project description:Rhesus macaques that were exposed to sevoflurane on three occasions during the early postnatal period (days 7, 21, and 35) showed signs of anxiety and impaired visual recognition memory, according to previous studies. By using this animal model, we were able to study the changes in proteomics and RNA sequencing in the prefrontal cortex of rhesus macaques after sevoflurane anesthesia.Proteomics analysis of the brain found that the functions of different proteins expressed after sevoflurane exposure in rhesus macaques were mainly related to mitochondrial respiration.

A cytoplasmic tail motif directs HLA-E to clathrin-mediated endocytosis and valosin-containing-protein-mediated recycling

Project description:Recent discoveries highlight the effectiveness of major histocompatibility complex (MHC)-E-restricted CD8+ T cell responses in controlling certain infections, particularly in a rhesus cytomegalovirus (RhCMV68-1)-vectored simian immunodeficiency virus (SIV) vaccine. In this context, these responses are preferentially elicited and are crucial for enabling vaccinated rhesus macaques to eradicate an SIV challenge. To harness human leukocyte antigen (HLA)-E-restricted CD8+ T cell responses for therapeutic purposes, it is essential to understand how these responses are primed, especially the mechanisms regulating MHC-E trafficking within endosomal pathways. Here, we identified a novel lysine/tryptophan-based motif in the HLA-E cytoplasmic tail that promotes rapid surface turnover via clathrin-mediated endocytosis. This motif, combined with strong binding peptides, also facilitates HLA-E recycling through a distinct valosin-containing protein-dependent pathway. Additionally, we show that this motif and its associated endosomal transport mechanisms are conserved in rhesus macaques and Mauritian-origin cynomolgus macaques. These findings advance our understanding of how MHC-E regulates immune functions through unconventional transport processes and offer insights for stimulating HLA-E-restricted CD8+ T cell responses in immunotherapy development.

Multi-organ Proteomic and Phosphoproteomic Profiles of Mpox Virus Infection in SIVmac239-infected Rhesus Macaques

Project description:Macaque study was conducted in an ABSL-3 facility, adhering to protocols approved by the Institutional Animal Care and Use Committee (IACUC) under the guidelines of the Institute of Laboratory Animal Science, Chinese Academy of Medical Sciences (Approval Number XJ23004). All animals were single-housed and cared for in a facility fully accredited by the Association for Assessment and Accreditation of Laboratory Animal Care. Six rhesus macaques (3 males and 3 females), aged 6–8 years, were inoculated with SIVmac239 at a dose of 100 TCID50 via the intravenous (I.V.) route. On day 115 post-challenge, these 6 rhesus macaques, along with 6 new rhesus macaques (3 males and 3 females, aged 6–8 years), were challenged with 1×107 TCID50 of MPXV through the I.V. route. Half of the rhesus macaques were necropsied on day 10 post-MPXV infection for tissue viral load assessment, histopathology, and proteomic and phosphoproteomic analyses.
