Detection on the effect of TLR stimulation and NOS inhibition on human chondrocytes.

ABSTRACT: To enlarge our understanding on the effect of TLR1/2 stimulation to human chondrocytes and how NOS inhibition blocks this stimulatory effects, we planned to apply RNAseq analysis to human chondrocytes that are treated with TLR1/2 agonists and NOS inhibitor.

Project description:We report the gene expression differences between OT-1 T cells activated alone or in the presence of TLR1/2 (Pam3CSK4) or TLR7 (Gardiquimod). By activating OT-1 splenocytes in the presence or absence of the TLR agonists, isolating RNA from purified CD8+ T cells we show that cells activated in the presence of either TLR1/2 or TLR7 agonists had similar transcriptional profiles demonstrating an increase in Th1 cytokine expression over cells that were activated alone. This study provides a key piece of information for those designing T cell mediated therapies.

Project description:This study was performed to identify gene expression differences in not otherwise specified soft tissue sarcomas (NOS, malignant fibrous histiocytomas) and correlate them to histological findings and the clinical course. RNA was isolated and differential gene expression was analysed by the microarray technique. We used microarrays to detail the global gene expression differences between primary soft tissue sarcoma samples, harvested intraoperatively. Keywords: response to chemotherapeutic agent We compared the gene expression of ten different tumor samples of MFH/NOS sarcoma to determine global gene expression differences by microarray analysis.
