HFD versus LFD [RNA-seq]

ABSTRACT: Obesity alters the transcriptional profile of the adipose tissue. Our aim was to characterize the metabolic adaptations induced by diet-induced obesity in the adipose tissue. To this end wild-type mice were fed for 20 weeks a high-fat diet and subcutaneous adipose tissue was analyzed by RNA sequencing.

Project description:Here, we evaluated the molecular effects of dietary DINCH exposure, on the proteome, phosphoproteome and acetylome profiles of visceral and subcutaneous adipose tissue in a model of diet-induced obesity in male and female C57BL/6N mice. This study includes data on visceral and subcutaneous adipose tissue of male and female mice that were either fed a standard high-fat diet (HFD) or two HFD diets including doses of DINCH (4,500 ppm and 15,000 ppm).

Gpc3 Selectively Suppresses Subcutaneous Adipogenesis in Diet-Induced Obesity

Project description:Subcutaneous and visceral adipose depots employ distinct expansion strategies in response to dietary cues, yet the molecular regulators underlying these depot-specific adaptations remain poorly understood. Through integrated proteomic profiling of human subcutaneous and visceral adipose tissues from paired obese/non-obese donors and temporal transcriptomic analysis of mouse adipose stem and progenitor cells (ASPCs) during diet switching, we identified Gpc3 as an obesity-responsive gene exhibiting reciprocal expression patterns between depots. ASPC-specific Gpc3 deletion in mice amplified high-fat diet (HFD)-induced weight and fat mass gain by selectively enhancing hyperplastic expansion in inguinal white adipose tissue (iWAT), without affecting epididymal WAT (eWAT). Mechanistically, Gpc3 loss biased ASPC fate toward proliferation over adipogenesis via depot-specific modulation of canonical Wnt signaling. These findings establish Gpc3 as a regulator for regional adipose plasticity, offering a molecular target for reprogramming pathological fat distribution in obesity and related metabolic disorders.

Human s.c adipose tissue gene expression during diet-induced weight loss

Project description:Obesity has considerable effects on morbidity and mortality and the prevalence of obesity has been increasing rapidly worldwide during the past two decades. Even if obesity affects the entire individual, adipose tissue play a central role in the development of obesity. Expression profiling of adipose tissue may give insights into mechanisms contributing to obesity and obesity-related disorders. 24 obese subjects (6 women and 18 men, BMI 37.6 ± 4.9) received a very low calorie diet (450 kcal/d, Cambridge diet or Modifast) for 16 weeks. On average, patients lost 27.7 kg during the diet. Microarray expression analysis (Affymetrix U133A) in subcutaneous adipose tissue was performed before diet and after 16 weeks of diet.

Transcriptomic of Visceral Adipose Tissue in HFD-induced obesity: evaluation of Resveratrol

Project description:Visceral adipose tissue (VAT) expansion during the development of obesity is a hallmark of metabolic syndrome, characterized by hypoxia, oxidative stress, and a shift toward a pro-inflammatory secretome. This series examines the transcriptomic profile of VAT in male C57BL/6 mice subjected to long-term obesity induced by a high-fat diet (HFD, 60% of kcal from fat). To investigate new therapeutic avenues, we evaluated the impact of daily resveratrol administration (2.25 mg/kg) for 28 days, beginning at week 16 of life (8 weeks on the HFD diet). Using a transcriptomic approach (RNA-seq), we mapped the expression changes associated with the transition from healthy to dysfunctional adipose tissue. This study provides a detailed molecular resource for understanding how obesity-induced adipocyte stress can be attenuated by natural bioactives.

The aim of this study was to identify transcriptomic signatures in the gonadal WAT of ApoE3L.CETP mice in discrete stages of diet-induced MetS.

Adipose tissue biology and effect of weight loss in women with lipedema

Project description:Lipedema is a lipodystrophic disease characterized by marked increases in lower-body subcutaneous adipose tissue, anecdotally reported to: i) increase inflammation and fibrosis and impair microvascular and lymphatic circulation in the affected adipose tissue, ii) reduce risk of developing obesity-related cardiometabolic abnormalities; and iii) be resistant to diet-induced weight loss. To further our understanding of lipedema, we examined body composition, metabolic health and adipose tissue bology in women with obesity and lipedema (Obese-LIP) at baseline and following ~9% diet-induced weight loss. At baseline, people with Obese-LIP had ~23% greater leg fat mass, ~11% lower android-to-gynoid ratio and ~54% greater insulin sensitivity compared to women matched on age, body mass index and whole-body fat mass. In the Obese-LIP group, total and proinflammatory macrophage content and expression of inflammation and fibrosis-related genes were greater while lymph/angiogenesis-related genes were lower in subcutaneous femoral compared to abdominal adipose tissue. Diet-induced weight loss improved insulin sensitivity and decreased total fat mass due to similar reductions in abdominal and leg fat masses, with minimal effect on markers of adipose tissue inflammation/fibrosis and lymph/angiogenesis. Our study provides important insights into the pathophysiology of lipedema and suggests diet-induced weight loss should be the cornerstone therapy in people with Obese-LIP.

Myeloid lineage (CD45+ CD11b+ cells) analysis sorted from stromal vascular fraction of visceral and subcutaneous adiposetissue in mice in different diet conditions or after rosiglitazone treatment

Project description:Unlike visceral adipose tissue (VAT), subcutaneous adipose tissue (SAT) can play a protective role against the development of insulin resistance and metabolic dysfunction in obesity. These changes are associated with higher induction of pro-inflammatory programs and macrophage infiltration in VAT compared with SAT in obesity. Moreover, our results suggest that subcutaneous adipose tissue macrophages (ATMs) release small extracellular vesicles (sEVs) that can improve insulin sensitivity, opposite to the effect of visceral ATM sEVs in obesity. This functional difference was associated with changes in the proportion of resident ATMs, as well as proinflammatory, recruited ATMs. To study the mechanism by which adipose tissue inflammation and macrophage infiltration are differentially regulated in SAT vs VAT, we performed single cell RNA sequencing (scRNA-seq) analysis in sorted live CD45+ CD11b+ myeloid cells from stromal vascular fraction (SVCs) of SAT and VAT of mice fed normal chow diet, high fat diet, or SVCs of HFD mice after the diet switch from high fat diet to normal chow diet, or treatment with insulin sensitizing, rosiglitazone.

Role of Adipocyte Na,K-ATPase Oxidant Amplification Loop in Cognitive Decline and Neurodegeneration - Expression profiling in Brain

Project description:Adipocyte-specific expression of Na,K-ATPase signaling antagonist, NaKtide, has been shown to ameliorate the pathophysiological consequences of obesity and improve diseased phenotype. Recent studies have established that the release of adipocytokines and other bioactive mediators in obesity contributes to neurodegeneration. This study utilizes a mechanistic approach and characterizes the hippocampal transcriptome in comparison with transcriptomic changes in liver, visceral and subcutaneous adipose tissue. RNAseq analysis in high fat diet fed transgenic mice showed large scale differential gene expression as well as modulation of several biological pathways in hippocampus, liver, visceral and subcutaneous adipose tissue, which were attenuated by doxycycline induced adipocyte specific NaKtide expression.
