ABSTRACT: In order to study changes in gene expression during mushroom development in Schizophyllum commune, genome wide gene expression was analysed in 4 developmental stages: vegetative mycelium, stage I aggregates, stage II primordia and mature mushrooms. In order to study the effects of absence of expression of sc3 and sc4, genome-wide expression was analysed in a dikaryon in which those genes have been inactivated by targeted gene deletion.

Smelling the difference: Transcriptome, proteome and volatilome changes after mating

Project description:Mushrooms, such as Schizophyllum commune, have a specific odor. Whether this is linked to mating, prerequisite for mushroom formation, or also found in monokaryotic, unmated strains, was investigated with a comprehensive study on the transcriptome and proteome of this model organism. Mating interactions were investigated using a complete, cytosolic proteome map for unmated, monokaryotic, as well as for mated, dikaryotic mycelia. The regulations of the proteome were compared to transcriptional changes upon mating and to changes in smell by volatilome studies. We could show a good overlap between proteome and transcriptome data, but extensive posttranslational regulation was identified for more than 80 % of transcripts. This suggests down-stream regulation upon interaction of mating partners and formation of the dikaryon that is competent to form fruiting bodies. The volatilome was shown to respond to mating by a broader spectrum of volatiles and increased emission of the mushroom smell molecules 3-octanone and 1-octen-3-ol, as well as ethanol and ß-bisabolol in the dikaryon. Putatively involved biosynthetic proteins like alcohol dehydrogenases, Ppo-like oxygenases, or sesquiterpene synthases showed correlating transcriptional regulation depending on either mono- or dikaryotic stages.

Studies of target genes for signal transduction pathways (using a Ras1-, a Cdc42- and a Gap1-mutant) in the basidiomycete Schizophyllum commune.

Project description:Transcriptome changes associated with signal transduction mutants were analysed in order to identify targets of the distinct pathways of Ras1, Cdc42 and Gap1. Two monokaryons (12-43 and 4-39), a dikaryon (W22 x 12-43), a constitutive active Ras1 mutant (T2G12V, monokaryon), a Cdc42 mutant (G12V, monokaryon) and a M-bM-^HM-^Fgap1 x M-bM-^HM-^Fgap1 dikaryon (F15 x F28, deletion of Gap1) were analyzed for that purpose. With microarray analysis mRNA expression in different mutants of the basidiomycete S. commune was analyzed. Genes which show regulation in the comparison either of the two monokaryons 12-43 and 4-39 (for Ras2 and Cdc42 mutant) or the dikaryon W22 x 12-43 (for Gap1 x Gap1 dikaryon) have been eliminated from other comparisons as stage-specific (mono- or dikaryon) differences. Two biological replicate samples were analyzed for each condition.

Study of mating type genes and their roles in mating interactions of the basidiomycete Schizophyllum commune

Project description:Transcriptome changes associated with mating interactions were performed in order to identify signaling components and targets of pheromone response. Two monokaryons (S. commune 12-43 and 4-39), a dikaryon (W22 x 12-43), both semi-compatible mating interactions (Aon: W22 x 4-39; Bon, flat: W21 x 4-39), and different S. commune pheromone receptor recipient strains (Vbar2f, Vbar2t) were analyzed for that purpose. In addition, a S. commune strain showing the thin-phenotype (W22-thin) was investigated to figure out the role of Thn1 in mating.
