Gene expression analysis in human scalp biopsies from lichen planopilaris patients

ABSTRACT: We performed a small clinical trial using levia, theradome and plaquenil for the treatment of lichen planopilaris (LPP). Scalp biopsies were obtained at baseline and 6 months, and gene expression was assessed with the NanoString fibrosis panel with additional custom spike-in genes.

Project description:Lichen planopilaris (LPP) is a chronic inflammatory disease of unknown pathogenesis that leads to permanent hair loss. Whilst destruction of epithelial hair follicle stem cells (eHFSCs) that reside in an immunologically protected niche of the HF epithelium, the bulge, is a likely key event in LPP pathogenesis, this remains to be demonstrated. Laser capture microdissection of bulge cells from biopsies of lesional and non-lesional scalp skin from adult LPP patients were analyzed by microarray analysis.

Lichen Planopilaris and Pseudopelade of Brocq Involve Distinct Disease Associated Gene Expression Patterns by Microarray

Project description:Lichen planopilaris (LPP) and pseudopelade of Brocq (PPB) are two scarring alopecia diagnoses that often exhibit similar clinical features. Some suggest LPP and PPB are not distinct diseases, but rather different clinical presentations in a spectrum derived from the same underlying pathogenetic mechanism. We explored the degree of similarity between LPP and PPB gene expression patterns and the potential for common and unique gene pathway and gene activity in LPP and PPB using microarrays. Microarray analysis, using a 21K cDNA set was performed on pairs of biopsies obtained from affected and unaffected scalp of untreated patients (4 LPP and 4 PPB biopsy pairs). Diagnosis was confirmed by histopathology. Significantly differentially expressed genes were identified by analysis of microarray results in various data sets and screened for signaling pathway involvement. Selected genes were validated by quantitative PCR and immunohistology. The global gene expression profiles in LPP and PPB versus comparative intra-control scalp tissue were distinguishable by Significance Analysis of Microarrays (SAM). Specific genes, such as MMP11, were identified with significantly differential expression in association with LPP versus PPB. There was very limited commonality in the gene expression profiles between LPP and PPB. Our findings may have important implications for understanding the pathogenesis of LPP and PPB at the molecular level. Results suggest LPP and PPB involve different mechanisms of disease development and can be regarded as biologically distinct cicatricial alopecia diagnoses. Genes that we have identified may potentially be useful as markers of the respective diagnoses.

Project description:EGFR/MEK inhibitor therapy induces a distinct inflammatory hair follicle response that includes a collapse of hair follicle immune privilege and differential modulation of IL-33 and IL-37 expression. Our findings suggest that successful future management of EGFRi/MEKi-induced folliculitis requires restoration of hair follicle immune privilege. In this RNAseq parietal scalp (rather than truncal skin) biopsies were taken from long-term (3-9 months) EGFRi-treated patients exhibiting folliculitis (Chronic-EGFRi) or from patients prior to commencing and after two weeks of EGFRi therapy (Acute-EGFRi), compared to normal scalp skin.

In this sub-study, we evaluated the molecular responses to PF-06651600, an oral inhibitor of JAK3 and the tyrosine kinase expressed in hepatocellular carcinoma (TEC) kinase family, and PF-06700841, an oral TYK2/JAK1 inhibitor, versus placebo in nonlesional and lesional scalp biopsies of biopsy samples from patients with AA.

Project description:We have utilized the raw sequence data from our earlier investigation of the lichen transcriptome to design a custom DNA microarray for C. rangiferina in order to study the transcripts expressed in lichen thallus during dehydration and rehydration. The aim of this study was to identify the genes most differentially expressed during the rehydration and drying processes and also to get a more integrative view of the molecular players who may play roles in the processes required for lichen desiccation tolerance and the rapid re-establishment of photosynthesis through functional annotation. 8 samples with three biological replicates for each sample, altogether 24 samples. D1h samples have been drying for 1 hour, D3h samples for 3 hours, D6h samples for 6 hours and Dry samples for 24 hours. W15m samples have been wetted for 15 minutes, W30m samples for 30 minutes, W1h samples for 1 hour and Wet samples for 3 hours.
