Adult skull bone marrow is an expanding and resilient hematopoietic reservoir

Project description:The bone marrow microenvironment is a critical regulator of hematopoietic stem cell self-renewal and fate. While it is appreciated that aging, chronic inflammation and other insults compromise bone marrow function and thereby negatively affect hematopoiesis, it is not known whether different bone compartments exhibit distinct microenvironmental properties and functional resilience. Here, we have employed imaging, pharmacological approaches, and mouse genetics to uncover specialized and highly surprising properties of bone marrow in adult and aging skull. Specifically, we show that the skull bone marrow undergoes lifelong expansion involving vascular growth, which results in an increasing contribution to total hematopoietic output. Furthermore, skull is largely protected against major hallmarks of aging, including upregulation of pro-inflammatory cytokines, adipogenesis and loss of vascular integrity. Striking dynamic and rapid changes to the skull vasculature and bone marrow are induced by physiological alterations, namely pregnancy, but also pathological challenges, such as stroke and experimental chronic myeloid leukemia. These responses are highly distinct from femur, the most extensively studied bone marrow compartment. We propose that skull harbors a protected and dynamically expanding bone marrow microenvironment, which is relevant for experimental studies but, potentially, also clinical treatments in humans.

Brain-Engrafted Monocyte-derived Macrophages from Blood and Skull-Bone Marrow Exhibit Distinct Properties [scRNA_FIRE-ctrl-blood-skull]

Project description:Microglia arise from yolk sac progenitors and are thought to persist throughout life with minimal input from adult hematopoiesis. However, whether brain-engrafted monocyte-derived macrophages (MDM) exist at homeostasis and during turnover, and how they function relative to yolk sac-derived microglia (YSM) remain unsettled. Here, we combine lineage tracing, pharmacological microglia depletion, and multi-omics profiling to define the ontogeny, identity, and function of brain parenchymal macrophages. Despite sharing the parenchymal milieu, MDM display transcriptional and epigenetic landscapes distinct from YSM. Fate-mapping reveals that brain-engrafted MDM transiently express CD206, echoing a developmental stage of microglial precursors. MDM engraftment and polarization are modulated by IL-34 and CCR2. Furthermore, parabiosis and skull‑flap transplantation reveal that both blood and skull marrow supply the niche, yielding origin‑biased MDM states. Functionally, MDM engraftment enhanced cuprizone-mediated demyelination. Together, our study defines the origins, molecular features, and context-dependent roles of brain parenchymal macrophages across homeostasis, turnover, and CNS pathology.

Dynamic Release of Exosomes After Opening of the Blood-Brain Barrier Predicts Glioblastoma Susceptibility to Paclitaxel

Project description:Liquid biopsies hold promise to improve the diagnosis, response to therapy, and ultimately guide the management of cancer patients, however implementation of this approach in brain tumors has proven challenging due to the limited passage of molecules across the blood-brain barrier (BBB). We recently reported results from a phase I clinical trial in which the BBB was transiently opened in glioblastoma (GBM) patients using skull-implantable low-intensity ultrasound combined with microbubbles (LIPU/MB), performed every 3 weeks with paclitaxel administration until disease progression (NCT04528680). Here, we investigate circulating exosomes, a potential biomarker for susceptibility to treatment, as an exploratory endpoint of this trial. After developing and validating a microfluidic device designed to capture tumor-specific exosomes in glioblastoma patients (GlioExoChip), we studied longitudinal changes in exosome concentrations during LIPU/MB-based BBB disruption and chemotherapy delivery. Exosomes were characterized using nanoparticle tracking analysis, proteomics, western blot, and scanning electron microscopy. GlioExoChip had high capture efficiency for GBM-specific exosomes with phosphatidylserine and Annexin-V chemistry. Longitudinal analysis of exosome changes after BBB opening correlated with MRI enhancement among patients. In vitro, paclitaxel-susceptible glioma cells treated with this drug exhibited apoptosis and exosome release. In concordance, we found changes in exosome release following initiation of paclitaxel with LIPU/MB correlated with overall survival in GBM patients. Our work presents an efficient microfluidic platform for capturing of circulating GBM exosomes , prospectively characterizes the release of tumor material after the opening of the BBB in a clinical trial , and demonstrates that longitudinal liquid biopsies can provide practical insights to improve the care of brain tumor patients
