Piezoeletric cold atmospheric plasma sensitizes head and neck cancer to chemotherapy and immune checkpoint blockade therapy

ABSTRACT: To investigate the effects of Piezo-CAP on various types of HNC cells and elucidate the underlying mechanisms. Additionally, we examined the efficacy of combining Piezo-CAP with chemotherapy drugs (cisplatin) or immune checkpoint blockade (ICB) in head and neck tumor treatment. RNA-sequence (RNA-seq), Western blot and quantitative real-time PCR (qRT-PCR) were utilized to investigate the mechanism of Piezo-CAP treatment HNC cells.

Project description:Head and neck cancer (HNC) is the seventh most prevalent cancer globally, often characterized by chemo-resistance and immunosuppression, which significantly hampers treatment efficacy. Cold atmospheric plasma (CAP) has recently emerged as a promising adjuvant oncotherapy with substantial potential and advantages. In this study, Piezobrush® PZ2, a handheld CAP unit based on the piezoelectric direct discharge technology, was used to generate and deliver non-thermal plasma. We aimed to investigate the effects of CAPPZ2 on various types of HNC cells and elucidate the underlying mechanisms. In addition, we endeavored to examine the efficacy of combining CAPPZ2 with chemotherapy drugs (i.e., cisplatin) or immune checkpoint blockade (ICB, i.e., PD1 antibody) in HNC treatment. Firstly, the results demonstrated that CAPPZ2 exerted anti-neoplastic functions through inhibiting cell proliferation, migration and invasion, and promoting apoptosis and autophagy. Secondly, using transcriptomic sequencing, Western blotting, and quantitative real-time PCR, the mechanisms underlying CAPPZ2 treatment in vitro was presumed to be a multitargeted blockade of major cancer survival pathways, such as redox balance, glycolysis, and PI3K/AKT/mTOR/HIF-1α signaling. Lastly, combinatorial thearpy containing CAPPZ2 and cisplatin or PD-1 antibody significantly suppressed tumor growth and prolonged recipient survival in vivo. Collectively, the synergistic effects of CAPPZ2 and cisplatin or PD-1 antibody could serve as a promising solution to enhance head and neck tumor elimination.

Effect of cold atmospheric plasma (CAP) in MCF-7 breast cancer cells [dataset 1]

Project description:Paclitaxel is a widely used therapeutic chemical for breast cancer treatment, however, cancer recurrence remains obstacle for improved prognosis of cancer patients. In the current study, cold atmospheric plasma (CAP) was examined for its potential use to overcome the drug resistance, which mainly comprised of reactive oxygen and nitrogen species. After developing a paclitaxel (Tx)-resistant MCF-7 (MCF-7/TxR) breast cancer cell, CAP was applied to the cells and its effect on recovery of drug sensitivity was examined in cellular as well as molecular aspect. The MCF-7/TxR cells restored sensitivity against Tx up to 70% by CAP. Comparison of genome-wide expression profiles between the Tx resistance cell and its CAP-treated cell identified 86 genes that commonly appeared with significant change. Notably, 48 genes including DAGLA and CEACAM1, which are known to contribute to the acquisition of Tx resistance, showed opposite expression in the two cellular status. The protein expression level of selected genes, DAGLA and CEACAM1, was recovered to that of their parental cell by CAP. Furthermore, the dysregulation of DAGLA and CEACAM1 in MCF-7/TxR alleviated the drug sensitivity recovery effect of CAP. Taken together, CAP inhibited the growth of Tx-resistant MCF-7 cancer cells and recovered Tx sensitivity by resetting expression of multiple drug resistance–related genes. These findings may contribute to extending the application of CAP to the treatment of Tx-resistant cancer.

Identification of genes involved in apoptosis-induced by cold atmospheric pressure plasma in human lymphoma U937 cells

Project description:Cold atmospheric pressure plasma (CAP) is known as a source of biologically active agents, such as reactive oxygen species (ROS) and reactive nitrogen species (RNS). Recent medical investigations have focused on applying CAP to cancer treatment. There is also growing evidence that exposure of cells to CAP or CAP-activated medium induces apoptosis in cancer cells, and ROS and/or RNS are considered to be effective agents to CAP-induced apoptosis. More recently, we demonstrated that Ar-CAP or Ar containing 2.5 % of N2 (Ar-N2-CAP) significantly induced apoptosis in human lymphoma U937 cells. However, a detailed molecular mechanism underling the induction of apoptosis by CAP in cancer cells is unclear. In the present study, to identify genes involved in the induction of apoptosis by CAP in human lymphoma U937 cells, global-scale gene expression analysis was performed using a GeneChip® system.
