Neural stem and progenitor cells shorten S-phase upon commitment to neuron production

ABSTRACT: The dorsal telencephalon from E14.5 Tis21–GFP heterozygous mice embryos was subjected to microarray analysis. To identify genes whose expression were changed in Tis21–GFP positive S phase cells compared to Tis21–GFP negative S phase cells, Tis21–GFP positive or negative S phase cells labeled with a vital dye were separated by FACS corresponding to the GFP expression and DNA contents of the cells.

Project description:The dorsal telencephalon from E14.5 Tis21–GFP heterozygous mice embryos was subjected to microarray analysis. To identify genes whose expression were changed in Tis21–GFP positive S phase cells compared to Tis21–GFP negative S phase cells, Tis21–GFP positive or negative S phase cells labeled with a vital dye were separated by FACS corresponding to the GFP expression and DNA contents of the cells. 30 telencephalic hemispheres from 15 embryos of the Tis21–GFP heterozygous mice at E14.5 were dissected to collect approximately 200,000 S phase cells either from Tis21–GFP positive or negative for one replicate. Four independent experiments were performed from 3 different pregnant mice at least for each experiment set. Finally three best experiment sets were used for data analyses.

TIS21/BTG2/PC3 and cyclin D1 are key determinants of nuclear diacylglycerol kinase-zeta-dependent cell cycle arrest

Project description:In addition to lipid second messengers derived from the plasma membrane, increasing evidence supports the existence of nuclear lipid-dependent signaling networks. Diacylglycerol is a key second messenger, generated at the nuclear level, which is metabolized by diacylglycerol kinases (DGKs). It has been demonstrated that nuclear DGK-ζ negatively regulates cell cycle progression. The aim of this study was to identify key determinants of nuclear DGK-ζ-dependent cell cycle arrest in C2C12 mouse myoblasts. Using DNA microarrays, Real-Time RT-PCR and western blot, we demonstrated that nuclear DGK-ζ downregulated the expression of cyclin D1 and increased the expression of TIS21/BTG2/PC3, a transcriptional regulator of cyclin D1 with a strong anti-proliferative function. Overexpression of TIS21/BTG2/PC3 blocked the cells in G1 phase of the cell cycle and decreased the levels of Ser807/811 phosphorylated retinoblastoma protein, similarly to overexpression of DGK-ζ. Moreover, during myogenic differentiation of C2C12 cells, we showed an increase of TIS21/BTG2/PC3 expression and a decrease in cyclin D1 levels. siRNA downregulation of TIS21/BTG2/PC3 impaired myogenic differentiation by opposing cell cycle arrest. In summary, these data identify TIS21/BTG2/PC3 and cyclin D1 as downstream effectors of the nuclear DGK-ζ and highlight the importance of this DGK isoform in the regulation of myoblast proliferation and differentiation.

RNA-Seq analysis of microRNA expression profiles in mouse primary CFU-E late erythroid progenitors and Ter119+ mature erythroblasts

Project description:Using RNA-seq technology, we quantitatively determined the expression profile of microRNAs during mouse terminal erythroid differentiation. CFU-E erythroid progenitors were isolated from E14.5 fetal liver as the Ter119, B220, Mac-1, CD3 and Gr-1 negative, C-Kit positive and 20% high CD71 population. Mature Ter119+ erythroblasts were isolated from E14.5 fetal liver as C-Kit negative and Ter119 positive population. Consistent with nuclear condensation and global gene expression shut down during terminal erythroid differentiation, we found that the majority of microRNAs are downregulated in more mature Ter119+ erythroblasts compared with CFU-E erythroid progenitors.
