MiR184 and miR499 in HSPC proliferation and differentiation

ABSTRACT: We observed that platelet-derived extracellular vesicles (pEV) are enriched in miR184 and miR499. These pEVs are taken up by hematopoietic stem and progentior cells (HSPC). We wanted to know the effects of miR184 and miR499 on HSPC proliferation and differentiation.

Proteomic analysis of human platelet-derived extracellular vesicles generated via activation by different receptors

Project description:Renowned for their role in hemostasis and thrombosis, platelets are increasingly recognized for their involvement in innate immunity, immunothrombosis and inflammatory diseases. Platelets, heterogeneous in size and molecular content, express a wide range of receptors enabling versatile activation endpoints relevant for immune functions. Upon activation, platelet release extracellular vesicles (PEVs), whose formation and molecular cargo exhibit remarkable tunability based on receptor-mediated activation or environmental cues. This study aimed to compare the immune modulatory roles of PEVs released through activation by specific platelet receptors, GPVI, CLEC-2, and thrombin-collagen (TC). Functional assays in vivo in zebrafish and using an in vitro human macrophage models highlighted distinct immune-inflammatory responses triggered by PEVs. Surprisingly, despite extensive characterization using omics analyses and basic EV particle characterization, the differences in the protein and miRNA cargo and the EV physicochemical properties between the PEV types were subtle and insufficient to precisely define their mechanism of action to distinguish a distinct functional profile. Moreover, the constitutively formed PEVs used as controls displayed a disparate activation profile from the receptor-induced PEVs.

Proteome profiling of extracellular vesicles enriched from plasma of patients with lung adenocarcinoma and malignant melanoma.

Project description:We present an optimized HiRIEF LC-MS workflow to perform an in-depth proteome profiling of plasma derived extracellular vesicles (pEVs). Here, we have applied the HiRIEF pre-fractionation and tandem mass tags (TMT)-based peptide quantification to generate pEV proteomes of 6 malignant melanoma (MM) and 6 lung adenocarcinoma (LUAD) patients. We have detected traditional EV-marker proteins, tetraspanins, ESCRTs, and several other proteins associated to EV cargo selection, trafficking/sorting, and exosome biogenesis. Importantly, we also detected many LUAD and MM related proteins in the cancer pEVs. Our advanced proteomics workflow exhibits high pEV proteome coverage and the ability to detect potential disease-specific markers in pEVs enriched from clinical plasma samples.

Identification of Proteins in Streptococcus pneumoniae extracellular vesicles (pEVs)

Project description:Extracellular vesicles (EVs) have recently garnered attention for their participation in host-microbe interactions in Streptococcus pneumoniae infections. However, the effect of pEVs on the disruption of alveolar epithelial barrier remain poorly understood. Our studies focus on EVs produced by Streptococcus pneumoniae (pEVs), and reveal that pEVs are internalized by alveolar epithelial cells. In vitro, pEVs induce autophagy activation in a dosage-dependent manner and decrease the alveolar epithelial barrier’s trans-epithelium electrical resistance (TEER). In addition, pEV-containing bacterial peotein serine/threonine-protein kinase StkP may act as an activator for Streptococcus pneumoniae-induced autophagy activation. When administered systemically in mice, Streptococcus pneumoniae wild type strain induced acute lung injury, the deletion of stkP deletion strain attenuated this injury. Taken together, pEVs cargos emerge as critical contributors to tissue damage in mammalian hosts.

Project description:Our understanding of cell fate decisions in hematopoietic stem cells is incomplete. Here, we show that the transcription factor Helios is highly expressed in murine hematopoietic stem and progenitor cells (HSPCs), where it is required to suppress the separation of the platelet/megakaryocyte lineage from the HSPC pool. Helios acts mainly in quiescent cells, where it directly represses the megakaryocyte gene expression program in cells as early as the stem cell stage. Helios binding promotes chromatin compaction, notably at the regulatory regions of platelet-specific genes recognized by the Gata2 and Runx1 transcriptional activators, implicated in megakaryocyte priming. Helios null HSPCs are biased toward the megakaryocyte lineage at the expense of the lymphoid and partially resemble cells of aging animals. We propose that Helios acts as a guardian of HSPC pluripotency by continuously repressing the megakaryocyte fate, which in turn allows downstream lymphoid priming to take place. These results highlight the importance of negative and positive priming events in lineage commitment.

Protein profiling of plasma extracellular vesicles from rheumatoid arthritis patients.

Project description:Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic synovial inflammation and cartilage/bone damage. Cytokines like IL-1 and TNF, play a crucial role in the pathophysiology of RA but have limited diagnostic and prognostic value. Recently, extracellular vesicles (EVs) have gained attention in the pathogenesis of this disease, but which processes they reflect in RA is unknown. Therefore, we identified and quantified proteins in plasma-derived EVs from RA patients (RA-pEVs) by mass spectrophotometry (MS) and compared these proteins to pEVs from healthy controls (HC). The observed RA-pEV proteins were coupled to clinical or laboratory parameters to investigate which RA disease process is reflecting by circulating EVs.

Therapeutic effect of platelet-derived extracellular vesicles on viral myocarditis correlate with biomolecular content

Project description:Extracellular vesicles (EVs) can potently inhibit inflammation yet there is a lack of understanding about the impact of donor characteristics on the efficacy of EVs. The goal of this study was to determine whether the sex and age of donor platelet-derived EVs (PEV) affected their ability to inhibit viral myocarditis. PEV, isolated from men and women of all ages, was compared to PEV obtained from women under 50 years of age, which we termed premenopausal PEV (pmPEV). Because of the protective effect of estrogen against myocardial inflammation, we hypothesized that pmPEV would be more effective than PEV at inhibiting myocarditis. We injected PEV, pmPEV, or vehicle control in a mouse model of viral myocarditis and examined histology, gene expression, protein profiles, and performed proteome and microRNA (miR) sequencing of EVs. We found that both PEV and pmPEV significantly inhibited myocarditis; however, PEV was more effective, which was confirmed by a greater reduction of inflammatory cells and proinflammatory and profibrotic markers determined using gene expression and immunohistochemistry. Proteome and miR sequencing of EVs revealed that PEV miRs specifically targeted antiviral, Toll-like receptor (TLR)4, and inflammasome pathways known to contribute to myocarditis while pmPEV contained general immunoregulatory miRs. These differences in EV content corresponded to the differing anti-inflammatory effects of the two types of EVs on viral myocarditis.
