Shentong zhuyu decoction treatments regulate expression of inflammatory gene by potentially targeting transcription factors in dorsal root ganglia of rats with neuropathic pain


ABSTRACT: Shentong Zhuyu Decoction (STZYD) is a prescription of traditional Chinese medicine, which is widely used in many painful diseases including neuropathic pain (NPP). However, the potential pharmacological mechanism of STZYD is still unclear. The model of spinal nerve ligation (SNL) in rats was applied in this study. Then, RNA sequencing (RNA-seq) was used to screen differentially expressed genes (DEGs) between NPP model rats treated with STZYD and those without STZYD, and 301 DEGs and 14 transcription factors (TFs) were found. The differential expression of TFs is involved in the inflammatory reaction, apoptosis, nerve invasion and neurotoxicity of NPP. Then, the correlation between DEGs obtained in this project and these 14 TFs was analyzed. It was found that 14 TFs and 170 DEGs were co-expressed, with a total of 715 relationship pairs. Some DEGs are enriched into multiple inflammatory immune response biological processes. Therefore, we speculate that the differential expression of immune inflammation-related TFs may participate in the treatment of NPP by STZYD through transcriptional regulation of the differential expression of its target. The findings of this study provide a reference for further exploring the therapeutic targets of NPP.
