Project description:Arabidopsis MPK4 is involved in the control of antagonism between salicylic acid (SA) and ethylene (ET)/jasmonic acid (JA) pathways in the plant innate immune system as a repressor of the SA pathway, but an activator of the ET/JA pathway. Here we and use comparative microarray analysis of ctr1, ctr1/mpk4, mpk4 and wild type to show that MPK4 is required for only a narrow subset of ET regulated genes.

MPK4 phosphorylation dynamics and interacting proteins in plant immunity

Project description:The Arabidopsis MAP kinase 4 (MPK4) has been proposed as negative player in plant immunity, but it is activated by pathogen-associated molecular patterns (PAMPs), such as flg22. The molecular mechanisms by which MPK4 is activated and controls plant defense remain elusive. In this study, we investigated the Arabidopsis defense against a bacterial pathogen Pseudomonas syringae pv. tomato (Pst) DC3000 when the MPK4 is overexpressed. We showed an increase in pathogen resistance and suppression of jasmonic acid (JA) signaling in the MPK4 overexpressing (OE) plants. We also showed that the OE plants are very sensitive to flg22-triggered reactive oxygen species (ROS) burst in guard cells, which resulted in an enhanced stomatal closure, compared to wild-type (WT). During the flg22 activation, dynamic phosphorylation of MPK4 within and outside of the conserved TEY activation loop was observed. To elucidate how MPK4 functions during the defense response, we used immunoprecipitation coupled with mass spectrometry (IP-MS) to identify MPK4 interacting proteins in the absence and presence of flg22. Quantitative proteomic analysis revealed a shift in MPK4-associated protein network, providing insight into the molecular functions of MPK4 at a systems level.

Project description:af01_cul3a2_cul3b - comparison col0 vs 003d - Cullin proteins belong to a multigene family that includes at least three members in budding yeast, six in human, five in C. elegans and five in Arabidopsis thaliana. All cullins analyzed so far directly interact with RBX1/HRT1/ROC1, a RING finger protein, thereby forming the core module of different ubiquitin ligase complexes, which specifically recruit substrate proteins for ubiquitylation and subsequent degradation by the proteasome. CULLIN1, the only cullin protein studied so far in plants, has been shown to be essential for plant development and to play a role in multiple signalling cascades. Here, we report on the first molecular and genetic characterisation of a plant Cullin 3. In contrast to fungi and animals, the genome of the model plant Arabidopsis thaliana contains two related CULLIN 3 genes, called CUL3A and CUL3B. We found that CUL3A is ubiquitously expressed in plants and that the CUL3A protein is also able to interact with RBX1 and certain Arabidopsis BTB-domain proteins. In order to determine the role of CUL3A in plant development, we used a reverse genetic approach and identified T-DNA insertion mutants. cul3a null mutants flower slightly latter than the control plants and their hypocotyls exhibit a reduced sensitivity to far red light. Transcription profiling of Arabidopsis wild type, 35S:mks1 and mpk4 plants

Project description:In plants, MAPKs are implicated in signalling cascades regulating immediate as well as systemic responses to pathogens. Arabidopsis MAP kinase 4 (MPK4) negatively regulates systemic acquired resistance (SAR) accompanied by the expression of pathogenesis-related (PR) genes. MPK4 interacts with and phosphorylates MKS1. Plants that over-express MKS1 exhibit constitutive SAR. RNA was independently isolated three times from each of the three genotypes, wild-type, mpk4 and 35S:mks1 and hybridized to nine Affymetrix ATH1 GeneChips.
