Candida albicans interaction with intestinal epithelial cells

ABSTRACT: To characterize the interaction of Candida albicans with intestinal epithelial cells (C2BBe1) from early fungal adhesion to invasion (up to 6 h) and the late translocation and damage phase (12 -24 h), we conducted dual RNA-sequencing of C. albicans-infected C2BBe1 cells over a 24 h time course, with sampling at 0 h, 45 min, 3 h, 6 h, 12 h, and 24 h.

Interaction of soil amoeba Protostelium aurantium with three pathogenic yeasts Candida spp. (here: C. albicans and C. glabrata)

Project description:We investigated different escape and defense strategies of Candida glabrata and Candida albicans against the natural fungivorous predator Protostelium aurantium using dual RNA-Seq. While C. albicans was found to be very succesfull in preventing from amoeba recognition, C. glabrata in turn, was rapidly taken up but remained undigested with a comparably low transcriptional activity. Trophozoites of the amoeba Protostelium aurantium were fed seperately with the two fungal species and samples for RNA isolation were taken at time points 0 min (control), 30 min and 60 min. At indicated time points samples were shock frozen and used for RNA isolation. Samples for RNAseq were taken at time points 0 min (control), 30 min and 60 min. RNA samples from identical timepoints for C. albicans and C. glabrata were pooled for RNA sequencing. We investigated different escape and defense strategies of Candida spp. against natural fungivorous predator. While C. parapsilosis underwent rapid predation and intracellular killing of the yeast, C. albicans was found to be very succesfull in preventing from amoeba recognition. C. glabrata in turn, was rapidly taken up but remained undigested with a comparably low transcriptional activity.

Project description:Candida albicans is the most common human fungal pathogen causing mucosal and systemic infections, but human anti-fungal immunity remains poorly defined. Expression profiling of Candida-stimulated human peripheral blood mononuclear cells (PBMCs) provides new insights into Candida-specific host defense mechanisms in humans. Total RNA was extracted from PBMCs from healthy human volunteers. PBMCs were stimulated with heat-killed Candida albicans (10^6/ml), non-fungal inflammatory stimuli or RPMI control for 4 or 24 hours. A large number of biological replicates (>20) were included per stimulation condition and duration.

Project description:The fungal pathogen Candida albicans produces dark-pigmented melanin when grown in a basal medium containing 1 mM l-DOPA as melanin substrate. In the widely used C. albicans strain SC5314, melanin appeared after 3-4 days of incubation in l-DOPA medium. The experiment was designed to reveal cadidate genes associated with melanin biosynthesis by expression profiling at different times of growth with and without L-DOPA added to the medium. Expression profiling of C. albicans revealed very few genes significantly up- or down-regulated by growth in l-DOPA.

Transcriptional response of Candida albicans to endothelial cells

Project description:Candida albicans is exposed to a different host environment during different site of infection. Thus, different virulence factors may be active during differenttypes of infection. However,little is known about the C. albicans genes that are required for the initiation and maintenance of candidiasis. To identify potential virulence factors relevant to hematogenously disseminated candidiasis, we determined the transcriptional response of C. albicans to human umbilical vein endothelial cells (HUVECs) in vitro. Keywords: cell interaction Two different Candida albicans strains, CAI4-URA and a clinical isolate 36082, were used to identify the transcriptional response of C. albicans to HUVECs. The strains were incubated with either the HUVECs or bare plastic for 45, 90, and 180 min. C. albicans RNA was extracted and the transcriptional profile of these organisms was analyzed using the C. albicans oligonucleotide microarray. The transcriptional response to HUVECs was compared to that to bare plastic as a control condition. Each time point contains six biological replicates, three of which are from each C. albicans strain.
