RNA-seq of mouse tissue samples from animal model: Effects of MMP9 on gene expression.

ABSTRACT: In the present study, RNA-seq technique were performed to explore biological significance of MMP9 in the neuro-cutaneous signaling network of chronic itch. We performed high-throughput sequencing of mouse cheek tissue samples from MMP9 animal models (the MMP9 protein or its vehicle was injected into the right cheek of mice).

Project description:In the present study, RNA-seq technique were performed To explore biological significance of MMP9 in the neuro-cutaneous signaling network of chronic itch. We used MMP-9-IN-1, a specific inhibitor of MMP9, or its vehicle to treat ACD model mice, and conducted high-throughput sequencing on ear tissue samples of mice. It was found that inhibition of MMP9 expression affected the expression of a variety of itching factors and skin inflammatory factors related to the skin pathology of chronic dermatitis. These changes may indicate that MMP9 is closely related to inflammation and pruritus. This finding might help us to better understand the immune-neuro-modulatory mechanism in itch circuits.

We found that when using GSK3's inhibitor SB415286 AD mouse model or non GSK3's inhibitor SB415286 AD mouse model for high-throughput sequencing, using GSK3's inhibitor SB415286 ACD mouse model or non GSK3's inhibitor SB415286 ACD mouse model for high-throughput sequencing，SB415286 affected the expression of a variety of itching factors and skin inflammatory factors related to the skin pathology of human chronic dermatitis. These changes may indicate that GSK3 is closely related to inflammation and pruritus. This finding might help us to better understand the immune-neuro-modulatory mechanism in itch circuits.

Project description:As microRNAs have emerged as important regulators of molecular events occurring at the synapses, new questions about their regulatory effect on the behavior have arisen. In the present study, we elucidated the physiological consequences of miR132-Mmp9 mRNA interaction in vivo. To address this question, we created a new mouse model with a disrupted miR132 binding site in Mmp9 mRNA 3’UTR. In the current manuscript, we show for the first time that the dysregulated specific targeting of miR132 to Mmp9 mRNA in the mouse brain results in the increased level of Mmp9, which affects synaptic plasticity and has an effect on mice behavior. Our data points at the importance of complex and precise regulation of Mmp9 level by miR132 in the brain.

IL-33-activated calcitonin gene-related peptide-producing memory Th2 cells co-operate with somatosensory neurons to induce conjunctival itch

Project description:Allergic conjunctivitis is a chronic inflammatory disease that is characterized by severe itch in the conjunctiva; but how neuro-immune interactions shape the pathogenesis of severe itch remains unclear. We identified a subset of memory-type pathogenic Th2 cells that preferentially expressed Il1rl1-encoding ST2 and Calca-encoding calcitonin gene-related peptide (CGRP) in the inflammatory conjunctiva using a single-cell analysis. The IL-33-ST2 axis in memory Th2 cells controlled the axonal elongation of the peripheral sensory C-fiber and the induction of severe itch. Pharmacological blockade and genetic deletion of CGRP signaling in vivo attenuated scratching behavior. The analysis of giant papillae from patients with severe allergic conjunctivitis revealed ectopic lymphoid structure formation with the accumulation of IL-33-producing epithelial cells and CGRP-producing pathogenic CD4+ T cells accompanied by peripheral nerve elongation. Thus, the IL-33-ST2-CGRP axis directs severe itch with neuro-reconstruction in the inflammatory conjunctiva and is a potential therapeutic target for severe itch in allergic conjunctivitis.

An integrin α3β1-CSTF3 signaling axis regulates alternative polyadenylation of Mmp9 mRNA

Project description:The laminin-binding integrin α3β1 is highly expressed in epidermal keratinocytes, where it coordinates diverse cellular functions and gene expression during skin remodeling. Here, we show that α3β1-MEK/ERK signaling operates in vivo to promote proximal polyadenylation site (PAS) usage in the Mmp9 gene, generating a short, more stable mRNA transcript. Using mice with inducible, epidermis-specific α3 deletion, RNA in situ hybridization revealed that loss of α3β1 increased the long Mmp9 transcript in healing wounds and epidermal tumors. α3β1-MEK/ERK signaling in keratinocytes induced the expression of the cleavage stimulation factor CSTF3, a known regulator of alternative polyadenylation (APA), while CSTF3 knockdown shifted Mmp9 toward distal PAS usage. Moreover, α3 deletion reduced Cstf3 gene expression and altered APA in vivo. Genome-wide DaPars2 analysis identified α3β1-dependent APA across numerous genes, including some encoding components of the keratinocyte secretome. Together, these findings define a novel α3β1-MEK/ERK-CSTF3 axis that orchestrates post-transcriptional gene regulation through APA, revealing α3β1 as a potential target for wound and cancer therapies.
