ScRNAseq of Hematopoietic stem cell populations from human cord blood

ABSTRACT: Single cell transcriptomic profiling (sc RNA-seq) of the two Human Hematopoietic Stem Cell Populations from cord blood: 18LinnegCD34posCD133pos (hereafter CBC1:CD34pos ) and 18LinnegCD34negCD133pos HSCs (hereafter CBC3:CD34neg). Purpose: to compare the single cells transcriptomic profiles of the two Human Hematopoietic Stem Cell Populations: CD34pos HSCs and CD34neg HSCs in order to find unique homing molecules profile. Results provide insight of highly expressed adhesion molecules in CD34neg HSCs and their crucial role in interacting with the bone marrow microenvironment.

Project description:Hematopoietic stem cell transplantation (HSCT) is successfully applied since the late 1950s, however, its efficacy still needs to be improved. A promising strategy is to transplant high numbers of pluripotent hematopoietic stem cells (HSCs). Therefore, an advanced ex vivo culture system is needed that supports the proliferation and maintains the pluripotency of HSC to override possible limitations in cell numbers gained from donors. To model the natural HSC niche in vitro and thus, to amplify high numbers of undifferentiated HSCs, we used an optimized HSC cell culture medium in combination with artificial 3D bone marrow-like scaffolds made of polydimethylsiloxane (PDMS). After 14 days in vitro (DIV) cell culture, we performed transcriptome and proteome analysis of the whole cell populations. Ingenuity pathway analysis (IPA) indicated that our 3D PDMS cell culture scaffolds activated interleukin, SREBP, mTOR and FOXO signaling pathways as well as the HSC metabolism, which we confirmed by ELISA, Western blot and metabolic flux analysis. These molecular signaling pathways and HSC metabolism are well known to promote the expansion HSCs and are involved in their pluripotency maintenance. After selection and enrichment of immature CD34-positive/CD38-negative HSCs using FACS sorting, we could confirm our findings by another proteome analysis followed by IPA. Thus, we could show that our 3D bone marrow-like PDMS scaffolds activate key molecular signaling pathways to amplify the numbers of undifferentiated HSC efficiently ex vivo.

Human hematopoietic leukemic k562 cells: control Vs CXCR4 constitutively active mutant cells

Project description:The low homing and engraftment efficiency especially in context of low availability of donor CD34+ HSPCs is a major challenge, which limits the clinical applications of HSC therapies. SDF-1/CXCR4 axis plays a principle role in the homing and engraftment of hematopoietic stem/progenitor cells. Activation of CXCR4 induces cell trafficking and homing to the marrow microenvironment, where it retains hematopoietic stem cells in close contact with marrow stromal cells. The aim of our study is to understand the mechanisms of other molecules acting along with CXCR4 to provide new insights in the regulation of cell phenotypes. Rad21-mediated NF-κB signaling limits hematopoietic stem cell self-renewal in aging and inflammation

Project description:MTD project_description Inflammation and decreased stem cell function characterize organism aging, yet the relationship between these factors remains incompletely understood. This study shows that aged hematopoietic stem and progenitor cells exhibit increased ground-stage NF-κB activity, which enhances their responsiveness to undergo differentiation and loss of self-renewal in response to inflammation. The study identifies Rad21/cohesin as a critical mediator of NF-κB signals, by increasing chromatin accessibility of inter-/intra-genic and enhancer regions. Rad21/NF-κB are required for normal differentiation, but limit self-renewal of hematopoietic stem cells (HSCs) during aging and inflammation in an NF-κB dependent manner. HSCs from aged mice fail to downregulate Rad21/cohesin and inflammation/differentiation inducing signals in the resolution phase after acute inflammation. and The inhibition of cohesin/NF-κB is sufficient to revert the hypersensitivity of aged HSPCs to inflammation-induced differentiation. During aging, myeloid-biased HSCs with disrupted and naturally occurring reduced expression of Rad21/cohesin are increasingly selected over lymphoid-biased HSCs. Together, Rad21/cohesin mediated NF-κB signaling limits HSPC function during aging and selects for cohesin deficient HSCs with myeloid skewed differentiation.

single cell RNA seq of mouse bone marow-derived hematopoietic stem progenitor cells

Project description:Hematopoietic stem cells (HSCs) have been considered to progressively lose the self-renewal and differentiation potentials towards the commitment to each blood lineage. However, recent studies have suggested megakaryocyte progenitors are generated in a close relation with HSCs. In this study, we identified CD201-CD48- or CD48+ CD150+CD34-Kit+Sca-1+Lin- cells as new early megakaryocyte progenitors (MegP) in adult mouse bone marrow by single-cell (sc) transplantation, sc colony assay, sc RT-PCR, and sc RNA-sequencing. These MegP had little repopulating potential in vivo but formed megakaryocyte colonies in vitro. Transcriptome analysis suggested that these MegP lie downstream of HSCs and upstream of multipotent progenitors in a megakaryocyte differentiation pathway. The polyinosinic-polycytidylic acid (polyIC) injection and long-term reconstitution data suggested that a proportion of MegP expand after inflammation and transplantation. Here we propose a new model of HSC differentiation where HSCs give rise to common myeloid progenitors and MegP with little repopulating activity.

Gene expression profiling of novel hematopoietic stem cell subpopulations defined by CD35 expression and progenitor populations in human adult hematopoiesis

Project description:Cannonical hematopoietic stem cell (HSC) population is phenotypically defined as Lin- CD34+ CD38- CD45RA- CD90+ in human adult bone marrow. We recently identified novel HSC subpopulation defined as CD35+ HSCs. We performed microarray analysi to clarify the characteristics of CD35+ HSCs in comparison with other stem/progenitor populations. As compared to CD35- HSCs and progenitor populations, CD35+ HSCs showed lower expression of cell-cycle- or lineage-affiliated genes.suggesting their dormant or immature state.

Regulatory Role of S100A6 in Hematopoetic Stem Cells

Project description:The fate options of hematopoietic stem cells (HSCs) include self-renewal, differentiation, migration and apoptosis, but the interaction between intracellular Ca2+ and cytoplasmic chaperon protein in regulating fate options of long term-HSCs (LT-HSC) is unknown. We created a S100A6 conditional knockout mouse model in the hematopoietic system and our studies showed that in S100A6KO, the number of LT-HSCs was significantly reduced and HSCs engrafted poorly. After 5FU challenge, the frequency of S100A6KO HSCs remained significantly low. Our data showed that S100A6 failed to self-renew through Akt pathway in an intracellular calcium (Cai2+)-dependent manner. Expression profiling of S100A6KO obtained from gene signatures revealed that cytosolic calcium level and proteins translocation to mitochondria were decreased. Mitochondrial oxidative phosphorylation was impaired in S100A6KO. Proteomic data indicated Hsp90 protein and chaperonin family were reduced. Our findings demonstrated that S100A6 regulates fate options of HSCs self-renewal through integrating Akt signaling, specifically governing mitochondria metabolic function and protein quality.
