NAT10 as an acetyltransferase to promote erythropoiesis


ABSTRACT: Understanding erythropoiesis involves comprehending the process of erythroid differentiation, yet there are still numerous mysteryin this field.The purpose of this research was to look at NAT10-mediated regulation of gene expression during erythroid differentiation. NAT10, an acetyltransferase, has been shown to be highly expressed in the early stages of erythroid differentiation and to play an important role in stimulating erythroid differentiation.We identified ac4C modification on RAC2 and GDF15 mRNAs by acetylated RNA immunoprecipitation sequencing (acRIP-seq) and ribosome profiling sequencing (Ribo-seq) combined with RNA sequencing, which are significant downstream targets of NAT10 to improve erythropoiesis . Collectively, our data suggest that NAT10 promotes erythropoiesis by acetylating RAC2 and GDF15 mRNA associated with erythroid progenitors, thereby promoting erythroid differentiation by increasing RNA stability and translation efficiency. This study reveals unexpected patterns in the regulation of erythroid differentiation, advances our understanding of the regulatory network for erythroid differentiation from multiple perspectives, and identifies potential clinical targets for controlling erythroid regeneration.
