ABSTRACT: melanocortin 5 receptor (Mc5r) have various effects in immune responses. However, the involvement of Mc5r in ILC2 functions remains unclear. We identified the genes regulated by Mc5r in lung ILC2s during papain induced acute lung inflammation.

Project description:Mass spectrometry-based proteomics serves as an invaluable tool for pinpointing critical biomarkers and elucidating cellular functions and regulatory pathways. However, the proteomic changes in ILC2s during activation remain unexplored. To characterize the dynamic proteomic profiles of lung-resident ILC2s under physiological and inflammatory conditions, we performed high-resolution liquid chromatography-tandem mass spectrometry (LC-MS/MS) to systematically compare the molecular signatures of FACS-purified pulmonary ILC2 populations (Live CD45+CD90.2+Lineage- ST2+) isolated from two experimental groups: 1) steady-state control mice and 2) papain-challenged mice exhibiting acute airway inflammation (Fig. 1a and Supplementary Fig. 1a). Papain-induced lung inflammation was chosen because papain is a classical protease allergen triggering the release of pro-inflammatory factors including IL-33 to develop ILC2-derived lung inflammation.

Prenatal inflammation remodels lung immunity and function by programming ILC2 hyperactivation

Project description:We examined how prenatal inflammation shapes tissue function and immunity in the lung by reprogramming tissue-resident immune cells from early development. Maternal, but not fetal, type I interferon-mediated inflammation provoked expansion and hyperactivation of group 2 innate lymphoid cells (ILC2s) seeding the developing lung. Hyperactivated ILC2s produced increased IL-5 and IL-13, were associated with acute Th2 bias, decreased Tregs, and persistent lung eosinophilia into adulthood. ILC2 hyperactivation was recapitulated by adoptive transfer of a fetal liver precursor following prenatal inflammation, indicative of developmental programming at the fetal progenitor level. Reprogrammed ILC2 hyperactivation and subsequent lung immune remodeling, including persistent eosinophilia, was associated with worsened histopathology and increased airway dysfunction equivalent to papain exposure, indicating increased asthma susceptibility in offspring. Our data elucidate a potential mechanism by which early-life inflammation results in increased asthma susceptibility, driven by hyperactivated ILC2s that drive persistent changes to lung immunity during perinatal development.

Down-regulation of E protein activity augments an ILC2 differentiation program in the thymus

Project description:Innate lymphoid cells (ILCs) are important regulators in various immune responses. Current paradigm states that all newly-made ILCs originate from common lymphoid progenitors (CLP) in the bone marrow. Id2, an inhibitor of E protein transcription factors, is indispensable for ILC differentiation. Unexpectedly, we found that ectopically expressing Id1 or deleting two E protein genes in the thymus drastically increased ILC2 counts in the thymus and other organs where ILC2 normally reside. Further evidence suggests a thymic origin of these mutant ILC2s. The mutant mice exhibit augmented spontaneous infiltration of eosinophils and heightened responses to papain in the lung and increased ability to expulse the helminth parasite, Nippostrongylus brasiliensis. These results prompt the question whether the thymus naturally has the capacity to produce ILC2s and E proteins restrain such a potential. The abundance of ILC2s in Id1 transgenic mice also offers a unique opportunity for testing the biological functions of ILC2s.

Non-redundant roles for ILC2 and TH2 cells during anamnestic lung Type 2 immunity

Project description:Type 2 immune responses help with wound healing and clearing helminth infections, but the distinct roles of group 2 innate lymphoid cells (ILC2s) and T-helper 2 (TH2) cells are not fully understood. In this study, we examined whether ILC2s and TH2 cells are necessary or sufficient to repair lung injury and prevent re-infection with the parasite Nippostrongylus brasiliensis. Surprisingly, removing TH2 cells before re-infection did not affect lung healing or worm clearance. However, mice lacking ILC2s (LCR1-/-) showed severe IL-17-related lung bleeding and poor parasite clearance. Activated ILC2s expressed high levels of Amphiregulin (Areg) and Tph1, and Areg+ ILC2s were more abundant than TH2 cells in re-infected lungs. Although treatment with recombinant Areg increased TH2 cell numbers and improved worm clearance, it did not reduce lung injury in ILC2-deficient mice. Interestingly, serotonin levels influenced IL-17-driven lung damage regardless of parasite burden. Our findings suggest that while TH2 cells help fight infection, ILC2s play a unique and essential role in repairing lung tissue.

Piezo1-mediated Mechanotransduction Shapes ILC2 Translational Activity, Functions, and Lung Pathogenicity

Project description:Group-2 innate-lymphoid cells (ILC2s) are critical mediators of the type-2 immune responses in multiple lung pathologies. We show that Piezo1, a mechanosensitive ion channel, plays a key role in regulating ILC2 functions by linking mechanical cues to biochemical signaling pathways. Both murine and human ILC2s strongly express Piezo1, and its activation by Yoda1 selectively enhances IL-13 production through calcium influx, which activates the mTOR-S6K pathway. This pathway leads to translational reprogramming, favoring IL-13 translation. Piezo1-deficient in ILC2s impairs this process, reducing IL-13 levels and resulting in attenuated lung inflammation and fibrosis in mouse models of IL-33- or Alternaria alternata-induced airway inflammation and bleomycin-induced fibrosis. These findings position Piezo1 as a critical mediator of ILC2-driven type-2 immune responses and highlight its potential as a therapeutic target for lung diseases characterized by excessive inflammation. This streamlined understanding of Piezo1 function improves focus on its mechanistic role in lung pathology.
