Effect of YTHDF1 on concanavalin A-induced hepatitis

ABSTRACT: To investigate the effect of YTHDF1 on concanavalin A (ConA)-induced hepatitis , we performed RNA-seq with the total RNA extracted from the moue liver. Ythdf1-/- or wild type mice were intravenously injected with ConA (8 mg/kg) or saline for 8 hours. Then, the livers were collected, homogenized, and subjected to total RNA extraction.

Transcriptomic analyis of the response of porcine PBMC to stimulation with concanavalin A

Project description:One of the top priorities of the swine industry worldwide is to decrease the susceptibility of its animals to infectious diseases. There is therefore now an increased focus on the production of more M-bM-^@M-^\robustM-bM-^@M-^] animals, which have lower susceptibility to a broad range of infectious diseases. One general measure of immune competence is the proliferative response of peripheral blood mononuclear cells (PBMC) to mitogenic stimulation. Concanavalin A (ConA) is a mitogen that predominantly stimulates T lymphocytes within the PBMC fraction of blood. T cells play a crucial role in both cell-mediated and humoral immunity. Variation between individuals in the magnitude of the proliferative response to ConA has been estimated to have moderate heritability, and quantitative trait loci (QTL) for this trait have been identified. However, detailed knowledge of the genomic pathways which coordinate this process in swine PBMC is currently lacking. The recent development of a high density oligonucleotide microarray for the pig research community means that it is now possible to study simultaneously the expression of a large proportion of genes from the pig genome. The objective of this research project was to profile gene expression in ConA-stimulated PBMC to identify the gene networks that control the cellular response to this mitogen. Peripheral Blood Mononuclear Cells (PBMC) from 4 individual biological replicates were used for this experiment. Separate cell aliquots from these animals were grown in culture with the mitogen concanavalin A for 4 different times: 0, 3, 20, and 68 hours. For the hybridization experiment, 3 separate direct comparisons of gene expression were made (0 v 3 hr, 0 v 20 hr and 0 vs 68 hr). Time-point samples from the same individual were always compared on the same array. Each comparison consisted of 8 hybridizations (4 biological replicates; 2 dye-swap technical replicates), making a total of 24 hybridizations.

YTHDF1 deletion in macrophages aggravates LPS-induced inflammation

Project description:To investigate the effect of YTHDF1 on LPS-induced inflammation, we performed RNA-seq with the total RNA extracted from peritoneal macrophages (PMs) with or without YTHDF1 deletion. Ythdf1-/- or wild type mice were intraperitoneally injected with 6% starch broth. Three days later, PMs were isolated and cultured overnight. Then, the PMs were treated by LPS (1 μg/mL) or PBS for 6 hours, followed by RNA extraction and RNA-seq.

MicroRNA-674-5p/5-LO axis is important in concanavalin A-induced acute mouse liver injury

Project description:Eight-week-old male pathogen-free BALB/c mice were used for the induction of concanavalin A (Con A)-induced hepatitis. Animals were acclimated for at least 6-7 days to a 12-hour light/dark cycle in a humidity- and temperature-controlled, specific-pathogen-free environment in autoclaved cages. Mice were randomly assigned to two groups (4 in each group) and received Con A type IV (or saline) intravenously via the tail vein at the dose of 20 mg/kg for 2 hours. The animals were sacrificed for blood and liver collection. Blood was taken for transaminase and cytokine measurements. Livers were excised immediately after sacrifice. Part of the livers were immediately snap-frozen in liquid nitrogen and kept at -80Â°C for total RNA isolation and microRNA microarray analysis. miR-674-5p and other miRNAs were experimentally validated to be dysregulated by Con A in mouse livers. Liver samples from four mice treated with Con A and four mice treated with saline were used to acquire the miRNA expression profiling.

YTHDF1-N6-methyladenosine-ARHGEF2 axis promoted colorectal tumorigenesis and metastasis

Project description:N6-methyladenosine (m6A) governs the fate of RNAs through m6A readers. Colorectal cancer (CRC) exhibits aberrant m6A modifications and expression of m6A regulators. However, little is known about how m6A readers interpret oncogenic m6A methylome for malignant transformation. m6A reader YTHDF1 was overexpressed by copy number gain/amplification in majority of CRCs. YTHDF1 high expression and CNVs predict increased risk of CRC relapse. Transcriptome profiles of YTHDF1-high tumors exhibit highly metastatic features. YTHDF1 promoted CRC tumor cell and organoid proliferation and enhanced metastasis. Ythdf1 knockout dampened tumor growth in carcinogen-induced CRC model. Through multiomic integration, RhoA activator ARHGEF2 was characterized as the key functional YTHDF1 target based on its m6A and YTHDF1-binding signal, translation efficiency changes, protein correlations with YTHDF1 in clinical samples, and disrupted RhoA features by YTHDF1 knockdown. Moreover, YTHDF1-ARHGEF2 co-regulation was observed in YTHDF1-overexpressing metastatic sites and carcinogen-induced Ythdf1-null CRC tumors. ARHGEF2 overexpression significantly rescued RhoA signaling, tumor cell survival and invasiveness impaired by YTHDF1 knockdown both in vitro and in vivo, further confirming the essential function of ARHGEF2.
