Regional Immune Response to Trichostrongylis colubriformis - Gene Expression during Challenge of Immunized Sheep

ABSTRACT: Afferent Lymph was collected from up to 5 pen raised, nematode naïve, outbreed Romney sheep (Tag No’s 1001, 1002, 1003, 1004, 1006) over a 12 week period. During this time a Pre-Infection sample was collected, the sheep were immunised by 3 truncated (2 week) T. colubiformis infections (40,000 Tc) followed by a Challenge Infection with 40,000 T. colubiformis L3 (Challenge Infection weeks 1-3). The Challenge Infection will enable us to determine changes in gene expression when the sheep gut rejects the parasitic nematodes.

Characterization of host responses during Pseudomonas aeruginosa acute lung infection in the lungs and blood and after treatment with the synthetic immunomodulatory peptide IDR-1002

Project description:Pseudomonas aeruginosa is an opportunistic pathogen that causes nosocomial pneumonia and infects patients with cystic fibrosis. P. aeruginosa lung infections are difficult to treat due to bacterial resistance to antibiotics, and strains with multi-drug resistance are becoming more prevalent. Here we examined the use of a small host defense peptide, innate defense regulator 1002 (IDR-1002), in an acute P. aeruginosa lung infection in vivo. IDR-1002 significantly reduced the bacterial burden in the bronchoalveolar lavage fluid (BALF) as well as MCP-1 in the BALF and serum, KC in the serum, and IL-6 in the BALF. RNA-Seq was conducted on lungs and whole blood and the effects of P. aeruginosa, IDR-1002, or the combination of P. aeruginosa and IDR-1002 were evaluated. Differential gene expression analysis showed that P. aeruginosa increased multiple inflammatory and innate immune pathways as well as affected hemostasis, matrix metalloproteinases, collagen biosynthesis, and various metabolism pathways in the lungs and/or blood. Infected mice treated with IDR-1002 had significant changes in gene expression compared to untreated infected mice, with fewer differentially expressed genes associated with the inflammatory and innate immune responses to microbial infection, and treatment also affected morphogenesis, certain metabolic pathways, and lymphocyte activation. Overall, these results show that IDR-1002 was effective in treating P. aeruginosa acute lung infections and associated inflammation.

Expression data of Hepatocellular Carcinoma

Project description:Expression profiling of Xenografts of Hepatocellular Carcinoma Keywords: Human Cancer Briefly, primary HCCs obtained during liver resection were minced into fine fragments capable of passing through an 18-gauge needle, mixed in a 1:1 (v/v) ratio with Matrigel (Collaborative Research, Bedford, MA) and injected in the subcutaneous flanks of 8-week-old male severe combined immune deficient (SCID/SCID, The Jackson Laboratory, Harbor, ME) mice. Six to eight mice were injected for each primary tumor. Growth of established tumor xenografts was monitored twice weekly by vernier caliper measurement of tumor length (a) and width (b), and tumor volume was calculated as (a x b2) / 2. Serial passages of xenograft lines were obtained by dissecting tumors from sacrificed animals and reinjecting the dissociated tumor cells into successive generations of SCID mice as described above. Seven xenograft lines were studied in this report (2-1318, 5-1318(1), 5-1318(3), 2006, 26-1004, 26-1004(cirr), and 30-1004). Xenografts 2-1318 and 5-1318(1) were derived from Hep B-positive patients.

Mycobacterium bovis-BCG vaccination induces a specific pulmonary transcriptome biosignature in mice

Project description:In the present study, we applied microarray technology to define a biosignature from the whole genome expression in lung and spleen samples after BCG vaccination and M. bovis infection of BALB/c mice. The aims were two-fold, namely to define biosignatures that could predict vaccine success before challenge, and biomarker patterns that correlated with anamnestic protective responses following exposure to virulent M. bovis. Further, Our aim was to define these markers to be detectable without in vitro antigenic challenge. After BCG vaccination, we defined a specific pulmonary gene expression signature related to the connective tissue development and function network that predicted vaccine success before M. bovis challenge. In addition, a Th17-related cytokine profile was found that correlated with vaccine-induced protective immunity following infection with virulent M. bovis in the lung as well as additional genes that were up-regulated in the spleens of vaccinated animals post-infection that was related to neutrophil biology and inflammation. This study has therefore prioritized both biomarkers predicting vaccination success before challenge and bio-signatures that are associated with protective immune responses that will be useful to evaluate future vaccine candidates. Two groups of 20 mice each were immunised by a single intradermal injection of 2 x 10 to 5 CFU of M. bovis BCG (Vaccinated), or Hanks buffered salt solution (HBSS) (Unvaccinated). Six weeks later 5 mice from each group were euthanized for immunological analyses and the remaining mice from each group were challenged with approx 600 CFU M. bovis via the intranasal route. At days 3 and 14 post challenge five mice per group were euthanized and spleens and lungs harvested.
