Project description:Dicamba is an auxin-like herbicide that can stimulate the production of ethylene and ABA biosynthesis. The subsequent stomatal closure and build-up of reactive oxygen species is hypothesized to contribute to plant death. In order to further understand the herbicide dicamba's mode of action at the molecular level, we used microarrays to dissect global gene expression in Arabidopsis seedlings 10 hrs after dicamba treatment. 4 day old Arabidopsis seedlings (a Columbia line containing a -800GSTF8::LUC reporter) were flooded with water or 7mM dicamba for 40 minutes before the treatment was poured off. Ten hours after the start of the treatment, whole seedlings were collected

Project description:Background: The auxin herbicides 2,4-D and dicamba are commonly used for management of horseweed (Erigeron canadensis L, (syn: Conyza canadensis L)). Halauxifen-methyl is a new auxin herbicide and recently commercialized to control several broadleaf weed species under a range of sites and environmental conditions. While synthetic auxin herbicides have been used for over 70 years, the precise mode of action that leads to plant death has yet to be clearly characterized. As new chemical families are discovered and the use of these herbicides continues to increase, it is imperative to understand how synthetic auxin active ingredients work within the plant to cause an herbicidal effect. Results: At 1 hour after treatment (HAT), 48 genes were consistently upregulated across the three herbicides, many of which are involved in the auxin-activated signaling pathway and response to auxin. At 6 HAT, 735 genes were upregulated by all herbicide treatments including genes associated with hormone signaling, metabolism, transport, senescence, and gene expression. The GO terms representing the 501 genes downregulated in all herbicide treatments were broadly categorized under two major groups: ATP and photosynthesis. At 6 HAT, over 50% of the genes differential expressed among the three herbicide treatments were unique to a single active ingredient. Conclusion: This research presents a first look into the differential gene expression profiles in horseweed following a foliar application of synthetic auxin compounds that represent three unique chemical families. While there are an abundance of transcriptome similarities induced by each herbicide that accounts for the general auxin herbicide response, distinct gene expression changes exclusive to each compound cannot be ignored as a contributor to the mode of action.

To further investigate whether GPA1 is involved in the regulation of gene expression in response to ABA, we examined the induction of known ABA-inducible genes in the gpa1 mutant and compared it to wild-type. We found significant differences in levels of ABA-induced expression between wild-type and gpa1 mutant. In order to systematically investigate GPA1 involvement in ABA signalling leading to gene expression, we are requesting the transcriptome analysis of the gpa1 mutant in response to ABA.In detail, 2 week old wild-type and gpa1 plants grown in the 16/8 hrs light and dark cycle will be treated with either ABA or with a control solution for 3 hours. 10 plants will be used per sample to produce RNA, to give 4 samples, one for each chip: gpa1-1 mutant in the presence (chip1) or absence of ABA (chip2) and wild-type (WS2) in the presence (chip3) or absence of ABA (chip4).

Project description:Chloroplast-nuclear retrograde signaling is viewed as a mechanism for inter-organelle communication. Here we show the SAL1-PAP (3′-phosphoadenosine 5′- phosphate) retrograde pathway functions more broadly in guard cells, interacting with abscisic acid (ABA) signaling at least in part via exoribonucleases. Unexpectedly, PAP bypasses the canonical signaling components ABA Insensitive 1 (ABI1) and Open Stomata 1 (OST1) by priming an alternative pathway that restores ABA-responsive gene expression, ROS bursts, ion channel function and stomatal closure in ost1-2. This alternative pathway up-regulates lowly expressed Calcium Dependent Protein Kinases (CDPKs) which have the capacity to activate the key slow anion channel SLAC1 in response to ABA-mediated and ost1-2 independent calcium release. The role of PAP in priming an alternative pathway to bypass components previously considered essential for stomatal closure demonstrates how a chloroplast signal can have broader roles as a secondary messenger to directly intersect with and tune hormone signaling.

Project description:Understanding how developmental and environmental signals are integrated to produce specific responses is one of the main challenges of modern biology. Hormones and, most importantly, interactions between different hormones serve as crucial regulators of plant growth and development, playing central roles in the coordination of internal developmental processes with the environment. Herein, a combination of physiological, genetic, cellular, and whole-genome expression profiling approaches has been employed to investigate the mechanisms of interaction between two key plant hormones, ethylene and auxin. Quantification of the morphological effects of ethylene and auxin in a variety of mutant backgrounds indicates that auxin biosynthesis, transport, signaling and response are required for the ethylene-induced growth inhibition in roots but not in hypocotyls. Analysis of the activation of early auxin and ethylene responses at cellular level, as well as of global changes in gene expression in the wild type versus auxin and ethylene mutants, suggests a simple mechanistic model for the interaction between these two hormones in roots. This model not only implies existence of several levels of interaction but also provides a likely explanation for the strong ethylene response defects observed in auxin mutants. Abstract:
BACKGROUND: 24-hour biological clocks are intimately
connected to the cellular signalling network, which complicates
the analysis of clock mechanisms. The transcriptional regulator
TOC1 (TIMING OF CAB EXPRESSION 1) is a founding component of
the gene circuit in the plant circadian clock. Recent results
show that TOC1 suppresses transcription of multiple target
genes within the clock circuit, far beyond its
previously-described regulation of the morning transcription
factors LHY (LATE ELONGATED HYPOCOTYL) and CCA1 (CIRCADIAN
CLOCK ASSOCIATED 1). It is unclear how this pervasive effect of
TOC1 affects the dynamics of the clock and its outputs. TOC1
also appears to function in a nested feedback loop that
includes signalling by the plant hormone Abscisic Acid (ABA),
which is upregulated by abiotic stresses, such as drought. ABA
treatments both alter TOC1 levels and affect the clock's timing
behaviour. Conversely, the clock rhythmically modulates
physiological processes induced by ABA, such as the closing of
stomata in the leaf epidermis. In order to understand the
dynamics of the clock and its outputs under changing
environmental conditions, the reciprocal interactions between
the clock and other signalling pathways must be integrated.
RESULTS: We extended the mathematical model of the plant clock
gene circuit by incorporating the repression of multiple clock
genes by TOC1, observed experimentally. The revised model more
accurately matches the data on the clock's molecular profiles
and timing behaviour, explaining the clock's responses in TOC1
over-expression and toc1 mutant plants. A simplified
representation of ABA signalling allowed us to investigate the
interactions of ABA and circadian pathways. Increased ABA
levels lengthen the free-running period of the clock,
consistent with the experimental data. Adding stomatal closure
to the model, as a key ABA- and clock-regulated downstream
process allowed to describe TOC1 effects on the rhythmic gating
of stomatal closure. CONCLUSIONS: The integrated model of the
circadian clock circuit and ABA-regulated environmental sensing
allowed us to explain multiple experimental observations on the
timing and stomatal responses to genetic and environmental
perturbations. These results crystallise a new role of TOC1 as
an environmental sensor, which both affects the pace of the
central oscillator and modulates the kinetics of downstream
processes.

Project description:Coordinated regulation of protection mechanisms against environmental abiotic stress and pathogen attack is essential for plant adaptation and survival. Initial abiotic stress can interfere with disease resistance signaling. Conversely, initial plant immune signaling may interrupt subsequent ABA signal transduction. However, the processes involved in cross talk between these signaling networks have not been determined. By screening a 9,600 compound chemical library, we identified a small molecule [5-(3,4-Dichlorophenyl)Furan-2-yl]-Piperidin-1-ylMethanethione that rapidly down-regulates ABA-dependent gene expression and also inhibits ABA-induced stomatal closure. Transcriptome analyses show that DFPM also stimulates expression of plant defense-related genes. Plate grown 12-day-old seedlings were transferred into 6 well plates with 1:5000 (V/V) DMSO in water as a control, 30uM DFPM, and 10uM ABA in water as a treatment for 6 hours. DFPM was added 30 min prior to ABA treatment. RNA was extracted using Trizol (Invitrogen, Carlsbad, CA, USA) and further purified using RNeasy Plant RNA purification kit (QIAgen, Valencia, CA, USA). Three biological replicates of ATH1 oligonucleotide arrays were hybridized with labeled samples from 1) wild-type Columbia (WT) untreated, 2) WT with 30uM DFPM treatment, 3) WT with 10uM ABA treatment, 4) WT with 30uM DFPM and 10uM ABA treatment. Each biological replicate was prepared by combining 7 independently-treated samples. Seedling treatments, 4 agent, 1 genotype/variation sets.
