Decoding the DNA sequence of MAP1S promoter region interacting with HBx

ABSTRACT: Our previous research found that the X protein encoded by the hepatitis B virus (HBx) induces high levels of MAP1S expression. To determine the binding motif and enrichment of HBx in the promoter region of MAP1S gene, we transfected HEK293T cells with pHBx. The chromatin complex was extracted, followed by immunoprecipitation with HBx antibodies, and DNA sequencing. A binding region 1180bp upstream of the MAP1S transcription initiation site (TSS) was found, demonstrating that HBx up-regulated the expression of MAP1S by interacting with the MAP1S gene promoter region.
