Project description:We evaluated the change in expression of genes after treatment of stellate cells with retinoic acid to understand how the stellate cells can de-differentiate and effect their physiological behaviour in pathological conditions. We then tested the changes in the gene expression in 2D and 3D culture conditions for pancreatic stellate cells and in a pancreatic cancer model. Keywords: gene expression change, dose response, matrix conditions We treated a pancreatic stellate cell line on plastic or Matrigel with 1 or 10 micromolar dose of all-trans retinoic acid (ATRA). RNA was extracted and hybridized on Illumina Human microarrays. We looked for target genes regulated by ATRA and evaluated for dose repsonse and change due to background culture conditions.

RNA was extracted and hybridized on Illumina Human microarrays. We looked for target genes regulated by ATRA and evaluated for time course change.

Project description:The biology of metastatic pancreatic ductal adenocarcinoma (PDAC) is distinct from that of the primary tumor. We detected an upregulation of the secreted axon guidance molecule Netrin-1 (Ntn1) in human and mouse PDAC metastases that signals through its receptor, uncoordinated- 5b (Unc5b) to facilitate metastasis in vitro and in vivo. The mechanism of Ntn1 induction in liver metastasis is mediated by hepatic stellate cell (HSC) secreted retinoic acid through RXR/RAR and Elf3 signaling. Ntn1 is present in PDAC-derived extracellular vesicles (EVs) and facilitates metastasis by inducing HSC activation through both long and short distance intercellular communication. Importantly, administration of a neutralizing monoclonal NTN1 antibody decreased metastases and increased survival in several murine PDAC models (autochthonous and metastatic). Our studies reveal a new metabolic feed-forward loop for metastatic niche formation in the liver involving NTN1 in PDAC-secreted EVs and retinoic acid released from HSCs which is targetable using anti-NTN1 therapy.

Retinoic Acid Receptor Gamma is Required for Proliferation of Pancreatic cancer cells

Project description:Despite of the expectation that retinoic acid receptor could be the potential therapeutic targets for pancreatic cancers, there has been the lack of information about the role and the impact of Retinoic acid receptor gamma (RARγ, RARG) on pancreatic cancer, unlike other two RARs.RARG is commonly over-expressed in human pancreatic cancer and is an independent diagnostic marker predicting the poor prognosis of pancreatic cancer patients. In addition, we demonstrated that the reduction in the expression of RARG in human pancreatic cancer cells dramatically suppress their proliferation and tumor growth in vivo, partially attributable to the down-regulation of tumor-supporting biological processes such as glucose transport and the decreased expression of various oncogenes like MYC and STAT3.

Genome wide analysis of pancreatic cancer cell gene expresison exposed to stromal pancreatic stellate cells in 3D gel matrix

Project description:Capan1 (well differentiated pancreatic cancer cell line) was co-cultured with pancreatic stellate cell line (PS1) embedded in a 3D organotypic model and gene expression was analysed in comparison to cancer cells cultured alone without stellate cells. Pancreatic stellate cells were embedded within a gel matrix composing collagen type I and Matrigel, and cancer cells were seeded on top. The gel was lifted on to metal grid after 24 hour and fed from below. Gels were harvested on day 10 and frozen sections obtained. The cancer cell layer on top of the gel was captured by laser microdissection for RNA extraction.
