Here we investigated the transcriptional effects of prednisone in muscle and adipose tissue when dosed as intermittent once-weekly 1mg/kg i.p. versus once-daily 1mg/kg in WT obese mice. Prior to treatment, mice were fed a high-fat diet for 12 weeks. After 12 weeks of treatment, we performed RNA-seq analysis in muscle (quadriceps) and white adipose tissue (ventral fat pad) to quantitate gene expression at isoform level.

Here we investigated the epigenetic effects of prednisone in muscle when pulsed as single 1mg/kg i.p. dose in mice WT mice at ZT0, i.e. when endogenous corticosterone is low. At 4-hours post-injection (ZT4), we performed ChIP-seq analysis in muscle tissue for the glucocorticoid receptor (GR), as well as BMAL1, clock factor active in the light-phase. We also performed ChIP-seq for RNApol-II to quantitate changes in RNApol-II recruitment. We repeated the GR and RNApol-II analyses in muscles from BMAL1-knockout mice.

Effects of Androstenedione, Ethinyl Estradiol, and Prednisone on Gene Expression in the Male Mouse Liver

Project description:The goal of this study was to provide insights into the adverse outcome pathways that underlie the liver tumor effects of androstenedione in the mouse liver by comparing gene expression patterns to two liver noncarcinogens, ethinyl estradiol and prednisone. B6C3F1 male mice were exposed for 7 days to control diet or diet containing ethinyl estradiol (EE2) at 1, 2.5, 5, or 10 ppm or prednisone (PRED) at 1.25, 2.5, 5, 25 ppm. Androstenedione (A4) was administered in 0.5% carboxymethyl cellulose by gavage at 2, 10, 20, and 50 mg/kg/day. Separate groups of mice were exposed to 50 mg/kg/day A4, 10 ppm EE2 or 25 ppm PRED for 28 days.
