Project description:Information for organismal patterning can come from a variety of sources. We investigate the possibility that instructive influences for normal embryonic development are provided not only at the level of cells within the embryo, but also via interactions between embryos. To explore this, we challenge groups of embryos with disruptors of normal development while varying group size. Here, we show that Xenopus laevis embryos are much more sensitive to a diverse set of chemical and molecular-biological perturbations when allowed to develop alone or in small groups, than in large groups. Keeping per-embryo exposure constant, we find that increasing the number of exposed embryos in a cohort increases the rate of survival while incidence of defects decreases. This inter-embryo assistance effect is mediated by short-range diffusible signals and involves the P2 ATP receptor. Our data and computational model emphasize that morphogenesis is a collective phenomenon not only at the level of cells, but also of whole bodies, and that cohort size is a crucial variable in studies of ecotoxicology, teratogenesis, and developmental plasticity.

Mechanics regulates ATP-stimulated collective calcium response in fibroblast cells.

Project description:Cells constantly sense their chemical and mechanical environments. We study the effect of mechanics on the ATP-induced collective calcium response of fibroblast cells in experiments that mimic various tissue environments. We find that closely packed two-dimensional cell cultures on a soft polyacrylamide gel (Young's modulus E = 690 Pa) contain more cells exhibiting calcium oscillations than cultures on a rigid substrate (E = 36 000 Pa). Calcium responses of cells on soft substrates show a slower decay of calcium level relative to those on rigid substrates. Actin enhancement and disruption experiments for the cell cultures allow us to conclude that actin filaments determine the collective Ca(2+) oscillatory behaviour in the culture. Inhibition of gap junctions results in a decrease of the oscillation period and reduced correlation of calcium responses, which suggests additional complexity of signalling upon cell-cell contact. Moreover, the frequency of calcium oscillations is independent of the rigidity of the substrate but depends on ATP concentration. We compare our results with those from similar experiments on individual cells. Overall, our observations show that collective chemical signalling in cell cultures via calcium depends critically on the mechanical environment.

Prevalence of 25 canine behavioral problems and relevant factors of each behavior in Japan.

Project description:Undesirable behaviors exhibited by dogs are called canine behavioral problems. Behavioral problems are common among dogs kept in ordinary homes; hence, veterinarians need to have sufficient knowledge of these problems, so as to advise owners. Among dogs kept in ordinary Japanese homes, only a few behaviors have been investigated regarding prevalence and relevant factors. To improve understanding of behavioral problems, a comprehensive survey is crucial. In this study, we distributed an online survey including general information and questions about 25 canine behaviors that can become problematic to owners, to investigate (1) the prevalence of behavioral problem, (2) the most common behavioral problems, and (3) the relevant factors of each behavior. We collected 2,050 valid responses. Among all dogs, 86.0% exhibited at least one behavioral problem. "Barking at noises inside the house", "Barking at unfamiliar visitors", and "Pica" were the most frequent behavioral problems. To reveal those factors related to the expression of each behavior, we performed multiple logistic regression analyses, with the presence of behaviors as objective variables and dogs' general information as explanatory variables. Breed, age, the presence of other dogs in the household, sex, neutering status, source of acquisition, and type of residence were associated with 19, 16, 5, 4, 4, 4, and 1 behaviors, respectively. This study revealed the prevalence of 25 canine behavioral problems in Japan and relevant factors of each behavior. Our findings will be useful for veterinarians in advising owners about preventing behavioral problems.

Condensin pinches a short negatively supercoiled DNA loop during each round of ATP usage.

Project description:Condensin, an SMC (structural maintenance of chromosomes) protein complex, extrudes DNA loops using an ATP-dependent mechanism that remains to be elucidated. Here, we show how condensin activity alters the topology of the interacting DNA. High condensin concentrations restrain positive DNA supercoils. However, in experimental conditions of DNA loop extrusion, condensin restrains negative supercoils. Namely, following ATP-mediated loading onto DNA, each condensin complex constrains a DNA linking number difference (∆Lk) of -0.4. This ∆Lk increases to -0.8 during ATP binding and resets to -0.4 upon ATP hydrolysis. These changes in DNA topology do not involve DNA unwinding, do not spread outside the condensin-DNA complex and can occur in the absence of the condensin subunit Ycg1. These findings indicate that during ATP binding, a short DNA domain delimited by condensin is pinched into a negatively supercoiled loop. We propose that this loop is the feeding segment of DNA that is subsequently merged to enlarge an extruding loop. Such a "pinch and merge" mechanism implies that two DNA-binding sites produce the feeding loop, while a third site, plausibly involving Ycg1, might anchor the extruding loop.
