Project description:Regeneration of alveolar bone is an essential step in restoring healthy function following tooth extraction. Growth of new bone in the healing extraction socket can be variable and often unpredictable when systemic comorbidities are present, leading to the need for additional therapeutic targets to accelerate the regenerative process. This study examined the effect of signaling through the TAM (Tyro3, Axl, Mertk) family of receptors on alveolar bone regeneration. We performed RNA sequencing on human alveolar bone stem cells treated with a pan-TAM inhibitor in vitro. We also performed RNA sequencing on tissue isolated from the extraction socket of WT and Mertk-\- mice.

Project description:Previous studies have suggested that Bmp4 is a key Msx1-dependent mesenchymal odontogenic signal for driving tooth morphogenesis through the bud-to-cap transition. Whereas the bud stage tooth developmental arrest in Msx1-/- mutant mice was accompanied by reduction in mesenchymal Bmp4 mRNA expression, we show that depleting functional Bmp4 mRNAs in the tooth mesenchyme, through neural crest-specific gene inactivation in Bmp4f/f;Wnt1Cre mice, caused mandibular molar developmental arrest at the bud stage but allowed maxillary molars and incisors to develop to mineralized teeth. We show that the Wnt inhibitors Dkk2 and Wif1 were much more abundantly expressed in the mandibular than maxillary molar mesenchyme in wildtype embryos and that Dkk2 expression was significantly unregulated in the tooth mesenchyme in Bmp4f/f;Wnt1Cre embryos. In addition, expression of Osr2, which encodes a zinc finger protein that antagonizes Msx1-mediated activation of odontogenic mesenchyme, is significantly upregulated in the molar mesenchyme in Bmp4f/f;Wnt1Cre embryos. Msx1 heterozygosity enhanced maxillary molar developmental defects whereas Osr2 heterozygosity rescued mandibular first molar morphogenesis in Bmp4f/f;Wnt1Cre mice. Moreover, in contrast to complete lack of supernumerary tooth initiation in Msx1-/-Osr2-/- mutant mice, Osr2-/-Bmp4f/f;Wnt1Cre compound mutant mice exhibit formation and subsequent arrest of supernumerary tooth germs that correlated with down regulation of Msx1 expression in the tooth mesenchyme. Taken together, our data indicate that, while reduction in mesenchymal Bmp4 expression alone could not account for the tooth bud arrest phenotype in Msx1-/- mutant mice, Bmp4 signaling synergizes with Msx1 and antagonizes Osr2 to activate mesenchymal odontogenic activity to drive tooth morphogenesis and sequential tooth formation. E13.5 mouse embryos tooth germs were microdissected by laser capture microdissection (LCM), and the mandibular molar and maxillary molar were separated. 3 pairs of control and mutant samples were pooled for the RNA extraction.

Project description:The alveolar ridge loss and absorption strongly affects the following implantation and prosthodontic treatment. It is a clinical challenge and research hotspot to sustain the alveolar bone mass. Interestingly, the patients affected with Tricho-dento-osseous (TDO) syndrome present high density of skull and jaw bones, and can remain sufficient bone mass after tooth extraction in a long time. The mechanism of this phenomenon is of highly study value. To explore DLX3-the disease-causing gene of TDO syndrome, we constructed the mouse alveolar loss and healing model. After tooth extraction, gelatin sponge with GFP or with adenovirus vector that can suppress the expression of Dlx3 gene was put in normal murine alveolar socket, respectively. RNA sequencing of the alveolar bone of these two groups was performed to explore the downstream molecular alterations.

Previous studies suggest that Msx1 activates Bmp4 expression in the developing tooth mesenchyme to drive early tooth organogenesis. Whereas Msx1−/− mice exhibit developmental arrest of all tooth germs at the bud stage, however, mice with neural crest-specific inactivation of Bmp4 (Bmp4ncko/ncko), which lack Bmp4 expression in the developing tooth mesenchyme, showed developmental arrest of only mandibular molars. We recently demonstrated that deletion of Osr2, which encodes a zinc finger transcription factor expressed in a lingual-to-buccal gradient in the developing tooth bud mesenchyme, rescued molar tooth morphogenesis in both Msx1−/− and Bmp4ncko/ncko mice. In this study, through RNA-seq analyses of the developing tooth mesenchyme in mutant and wildtype embryos, we found that Msx1 and Osr2 have opposite effects on expression of several secreted Wnt antagonists in the tooth bud mesenchyme. Remarkably, both Dkk2 and Sfrp2 exhibit Osr2-dependent preferential expression on the lingual side of the tooth bud mesenchyme and expression of both genes was up-regulated and expanded into the tooth bud mesenchyme in Msx1−/− and Bmp4ncko/ncko mutant embryos. We show that pharmacological activation of canonical Wnt signaling by either lithium chloride (LiCl) treatment or by inhibition of Dkk in utero was sufficient to rescue mandibular molar tooth morphogenesis in Bmp4ncko/ncko mice. Furthermore, whereas inhibition of Dkk alone was insufficient to rescue tooth morphogenesis in Msx1−/− mice, pharmacological inhibition of Dkk in combination with genetic inactivation of Sfrp2 and Sfrp3 rescued maxillary molar morphogenesis in Msx1−/− mice. Together, these data reveal a novel mechanism that the Bmp4-Msx1 pathway drives tooth organogenesis by activating Wnt signaling via regulation of the secreted Wnt antagonists.

Project description:It is well established that tooth morphogenesis initiates at embryonic day 10.5 (E10.5), characterized by the specification of a single molar germ in each jaw quadrant. However, the developmental mechanisms underlying the establishment of spatially distinct maxillary and mandibular identities along the jaw axis prior to this stage remain elusive. In Wnt1-Cre;pMes-Bmp4 transgenic mice, a pathological model recapitulating human syngnathia (resulting in bilateral jaw fusion), we demonstrate here that BMP4 overexpression in cranial neural crest cells (CNCCs) disrupts early molar patterning with 100% penetrance, manifesting as the formation of only one molar germ within the unilateral fused jaw structure, establishing this model as a valuable tool for investigating the patterning of maxillary and mandibular molar germs during jaw development. Systematic analysis of wild-type embryos revealed the emergence of a single molar primordium in the unilateral first pharyngeal arch (PA1) by E9.0. This primordium undergoes segregation to form the maxillary and mandibular molar between E9.5-E9.75, a process synchronized with the anteroposterior patterning of PA1. Comparative studies with the mutant model indicated that this developmental segregation depends on coordinated migration of CNCC-derived molar primordium and a BMP4/FGF8-dependent gene regulatory network (GRN) at the maxillomandibular junction. Pathologically elevated BMP4 signaling upregulates the p38 intracellular pathway in the mutant jaws, perturbing the finely tuned GRN and disrupting post-migratory CNCC migration. These perturbations collectively lead to defective molar segregation in syngnathia mouse model. Our findings elucidate fundamental mechanisms governing early molar patterning and may provide molecular explanations for syngnathia-associated molar oligodontia in humans.

Project description:Periodontal granulation tissue is formed during the wound healing process and contributes to subsequent repair or regeneration. However, periodontitis causes the presence of unwanted mass of persistently inflamed granulation tissues in periodontal osseous defects, are routinely eliminated and discarded for anticipated better treatment outcomes. Nevertheless, the emerging evidence of microbiome and histological profiles strongly encourages further studies on this considerable issue and the potential clinical implications. Here, we collected the granulation tissues from pocket wall via excision (PT) and osseous defect (GT) during periodontal surgery, and those attaching on the extracted root surfaces (RT) and remaining in the extraction sockets (ST) during tooth extraction. Subsequently, these samples were used for histological assessment, 16S rRNA sequencing of intra-tissue microbiome and single-cell sequencing (scRNA-seq). The origin-based distinguishment of granulation tissue was well reflected by the clinical and biological characteristics. Conceivably, the periodontal parameters at the involved teeth of RT and ST were more deteriorated than those of GT and PT, which was also reflected with the enrichment of genera of periodontopathogens like Porphyromonas and the depletion of commensals such as Rothia and Neisseria in RT and ST. Further scRNA-seq revealed higher proportion of mesenchymal stem cells and fibroblasts while fewer natural killer cells in GT than those in both PT and ST, in line with elevated wound healing scores and decreased inflammatory response scores. Pseudotime trajectory analysis demonstrated the apparent differentiation of stroma cells among these tissues. Histological examination exhibited a less extent of inflammatory infiltration and CD4+ and NKG2D+ immunoreaction in GT, as compared to other tissue types. Collectively, these findings show the salubriousness of osseous GT and deleteriousness of socket-originated RT and ST in terms of clinical, microbiome and cellular attributes. This work could facilitate the development of novel strategies for potentiating and enhancing periodontal treatment outcomes.
