Project description:The Purpose of this study is to screen for microRNAs regulated by hypoxia in neonatal rat cardiomyocytes. 3 biological replicates were perfomed for each each group, normoxia and hypoxia. One normoxic and one hypoxic sample was hybridized to each chip for a total of 6 2-color experiments. A dye swap for each oxygen treatment were performed.

Project description:Genome-wide gene expression analysis at different stages of cardiomyocyte differentiation (undifferentiated mouse embryonic stem cells, neonatal mouse cardiomyocytes and adult mouse cardiomyocytes). Results provide important information on the differential expressed genes between undifferentiated mouse embrionic stem cells (mES) and mouse cardiomyocytes (CM) and also between cardiomyocytes from neonatal (CMp) and adult stages (CMa). This dataset allowed us to compare the expression profile of mES, CMp and CMa with the epigenetic profile of histone methylation generated with ChIP-seq experiments. Electric stimulation of neonatal rat ventricular cardiomyocytes

Project description:Study on changes in gene expression in primary cultures of neonatal rat ventricular cardiomyocytes to electric stimulation. Through comparing non-stimulated, stimulated and blebbistatin supplemented and stimulated cultures we set out to identify the genes that are specifically activated by electric pulsing separate from cardiomyocyte contractions. After initial recovery phase, primary cultures of neonatal rat ventricular cardiomyocytes were cultured for 3 days without pulsing, with electric pulsing or with electric pulsing combined with blebbistatin. Per treatment: 3 arrays, with samples obtained from 3 separate series of cardiomyocyte isolation and culturing. Per array: sample prepared from pooled (1:1) RNA from duplicate experiments.

Project description:FGF21 belongs to the FGF superfamily and the highest expression of the FGF21 transcript was found in the liver. The initial studies identified FGF21 as an endocrine hepatokine with crucial roles in regulating lipid, glucose and energy metabolism. More recent studies have indicated a role for FGF21 in cardiovascular stress and diseases.The goal of our study is to analyze the effect of FGF21 on ischemic cardiomyocytes transcriptome using microarray. Total RNA was extracted from cultured neonatal mice cardiomyocytes after exposure to hypoxia (in 2% oxygen, 5% CO2 and 93% N2) for 12 h in the absence or presence of FGF21. Samples were processed for hybridization to the Mouse Gene 2.0 ST Array (Affymetrix). We sought to find out the differentially expressed genes regulated by FGF21 in hypoxic cardiomyocytes.
