Single cell transciptional profiling of a hESC 3D extra-embryonic amnoid model (PGA - post-gastrulation amnoid)

ABSTRACT: The goal of this study was to carry out a comprehensive molecular characterization of transcriptional profile and cell types formed during amniogenesis at the single-cell level. We further explored the transcriptional profiles and cell types formed over time when the extra-embryonic cells were mixed with (primed) hESC representing cells of the inner cell mass epiblast.

Single cell transcriptome profiling of mouse and hESC-derived pancreatic progenitors

Project description:Human embryonic stem cells (hESCs) are a potential unlimited source of insulin-producing β-cells for diabetes treatment. A greater understanding of how β-cells form during embryonic development will improve current hESC differentiation protocols. As β-cells are formed from NEUROG3-expressing endocrine progenitors, this study focused on characterizing the single-cell transcriptomes of mouse and hESC-derived endocrine progenitors. To do this, 7,223 E15.5 and 6,852 E18.5 single cells were isolated from Neurog3-Cre; Rosa26mT/mG embryos, allowing for enrichment of endocrine progenitors (yellow; tdTomato + EGFP) and endocrine cells (green; EGFP). From a NEUROG3-2A-eGFP CyT49 hESC reporter line (N5-5), 4,497 hESC-derived endocrine progenitor cells were sequenced. Differential expression analysis reveals enrichment of markers that are consistent with progenitor, endocrine, or novel cell-state populations. This study characterizes the single-cell transcriptomes of mouse and hESC-derived endocrine progenitors and serves as a resource (https://lynnlab.shinyapps.io/embryonic_pancreas/) for improving the formation of functional β-like cells from hESCs.

Microarrays confirmed the hematoendothelial, endothelial and hematopoietic identity of hPSC-derived CD144+-EBs, BCs and ECs, respectively, and the similarities between hESC-derived cell populations and human EL equivalent populations.

Expression analysis of λH1-hESC derived β-like cells

Project description:Purpose: Pancreatic islet transplantation is an effective cell therapy for type 1 diabetes (T1D), but its clinical application is limited by the shortage of donor pancreata. Among the potential alternatives, the differentiation of human embryonic stem cells (hESc) into insulin-producing β-cells has taken an early lead. However, while the proportion of β-cells obtained through current methods is relatively high, a significant percentage of undefined non-endocrine cell types are still generated. Most importantly, there is the potential for carry-over of non-differentiated cell types that may produce teratomas upon transplantation. In order to address these issues, we sought to modify hESc so that their differentiated progeny could be selectively devoid of tumorigenic cells and enriched for cells of the desired phenotype (in this case, pancreatic β-cells). Methods: mRNA profiles of λH1-hESC derived β-like cells were developed by mRNA sequencing, in triplicate, using Illumina HiSeq PE Cluster Kit v4 and Illumina HiSeq Flow Cell v4 with 50 nt paired end reads plus dual index reads using the Illumina HiSeq SBS kit v4. Sequence reads that passed quality filters were analyzed at the transcript isoform level following alignment using TopHat v2.1.0 followed by exon and gene level counting using Bioconductor easyRNASeq v 2.4.7. Conclusions: Our study demonstrates that λH1-hESC derived β-like cells have a transcriptional expression profile similar to ESC derived β-like cells.

RNA-seq of human embryonic stem cells (hESC) line Amicqui-1 derived and maintained on human amniotic epithelial cells

Project description:Conventional conditions to maintain human embryonic stem cells (hESC) imply the use of inactive mouse embryonic fibroblast (iMEF) as a feeder layer. However, it has suggested that the culture of hESC on iMEF could be an artifact that does not correspond to the in vitro counterpart of the human epiblast. We previously derived and maintained human embryonic stem cells (Amicqui-1 hESC line) on a feeder layer of human amniotic epithelial cells (hAEC). However, the mechanisms involved in the interaction between both cell types to promote the pluripotency still remain unknown. To elucidate if the transcription profile of hESC on hAEC differs from conventional conditions on iMEF, we carried out RNA-seq of Amicqui-1 hESC on both feeder layer conditions (AMIQ hESC-iMEF and AMIQ hESC-hAEC) and on their respective conditioned media (AMIQ hESC-hAEC-CM and AMIQ hESC-iMEF-CM). In this experiment we included H1-hESC-iMEF and hAEC alone.

Expression data from undifferentiated human embryonic stem cells (hESC) and Day 3.5 mesodermal progenitor (CD326neg CD56+) population

Project description:Our understanding of how mesodermal tissue is formed, has been limited by the absence of specific and reliable markers of early mesoderm commitment. We report that mesoderm commitment from human embryonic stem cells (hESC) is initiated by Epithelial to Mesenchymal transition (EMT) as shown by gene expression profiling and by reciprocal changes in expression of the cell surface proteins, EpCAM/CD326 and NCAM/CD56. Molecular and functional assays reveal that CD326negCD56+ cells, generated from hESC in the presence of activin A, BMP4, VEGF and FGF2, represent a novel, multi-potent mesoderm-committed progenitor population. CD326negCD56+ progenitors are unique in their ability to generate all mesodermal lineages including hematopoietic, endothelial, mesenchymal (bone, cartilage, fat, fibroblast), smooth muscle and cardiomyocytes, while lacking the pluripotency of hESC. CD326negCD56+ cells are the precursors of previously reported, more lineage-restricted mesodermal progenitors. These findings present a novel approach to study how germ layer specification is regulated, and offer a unique target for tissue engineering. We used microarrays to compare gene expression profile of early mesodermal progenitors with undifferentiated hESC (H9 line). Mesoderm induction from hESC was initiatiated with combination of morphogens and growth factors including activin A, bone morphogenic protein 4, basic fibroblast growth factor and vascular endothelial growth factor. Proposed mesodermal progenitor population was isolated by FACS on day 3.5 of culture based on the presence of CD56 expression and the absenbce of CD326 expression.
