PcG/TrxG profiling of differentially aged adipose-derived mesenchymal stem cells

Project description:The microarray reported here is part of a study aimed to investigate whether the gene expression profile of Polycomb, Trithorax and interacting molecules (collectively referred to PcG/TrxG genes) might be related to different states of differentiative potential. In particular , this microarray profiles the expression of PcG/TrxG genes in adipose-derived mesenchymal stem cells isolated from the inguinal fat depot of mice of different age. MSC have been isolated from the inguinal fat pad of FVB mice aged 1, 3, 6, 12 or 24 months.

Interferon Regulatory Factor 7 (IRF7) in aging of adipose-derived mesenchymal stromal cells

Project description:We found that Interferon Regulatory Factor 7 (IRF7) is upregulated in adipose-derived stromal cells (MSC) of 12 months-old (middle-age) mice compared with 1 month-old mice. To go in depth into the role of IRF7 in aging of MSC we knocked-down it in 12 months MSC and assessed the transcriptional changes though whole-genome microarray analysis. We investigated the transcriptional changes induced by the Interferon Regulatory Factor 7 (IRF7) knock-down in mesenchymal stromal cells established from the inguinal fat pad of 12-months-old mice.

Exploration of the Chicken Adipose Tissue

Project description:Adipose tissue is essential for lipid storage and thus, energy regulation and maintenance. Many organs in the body utilize lipids as their primary energy source, such as the heart. In addition, when glucose supplies are insufficient adipose tissue is broken down and fats are utilized systemically as a primary energy source. This is regulated by the endocrine system primarily through the regulation of glucagon and insulin. Adipose tissue is also involved with the immune system particularly in eliciting an inflammatory response through cytokine signaling and the release of interleukins. Chickens have two major fat pads: an abdominal fat pad and a cardiac fat pad. The abdominal fat pad is located on the ventral side, above the GI tract. The cardiac fat pad is situated on the atria of the heart and surrounds the aorta. These fat pads are different in coloration and size which leads to the hypothesis that these fat pads have different functions. The objectives of the project are to (a) identify genes that are unique to each type of adipose tissue when compared to the rest of the body, through the collection of other tissues and (b) to identify genes that are differentially expressed in each type of adipose tissue in comparison to each other to better understand the functioning of these fat pads in the chicken and the pathways that they are involved in at the transcriptomic level.

Translating transcriptome of cancer cells in situ in mesenchymal-rich tumor microenvironment

Project description:A mesenchymal rich stroma such as cancer-associated fibroblasts (CAFs) in breast tumors favors the selection of cancer clones with enhanced bone metastatic ability. To determine the cancer cell transcriptomic response to the mesenchymal stroma, we supplemented experimental mammary tumours with or without exogenous mesenchymal cells. We used bone marrow-derived human mesenchymal stem cells (MSCs) as a source of mesenchymal stroma, as MSCs have been shown to undergo CAF-like differentiation. We engineered the cancer cells to express an EGFP-tagged version of ribosomal protein L10a (EGFP-L10a). This allows the retrieval of cancer cell specific transcripts rapidly from whole tumor lysates by translating ribosome affinity purification (TRAP) and direct profiling of cancer cell gene expression patterns when they are in situ. EGFP-10a+ MDA-MB-231 cells were orthotopically injected into the mammary fat pad with or without 1:1 ratio of MSCs. The mammary tumors were retrieved for TRAP-RNAseq profiling after 3 weeks.
