High-throughput explorations of Drosophila models of AKT1 variants associated with juvenile ovarian granula cell tumors provide insights into their pathogenesis mechanisms.

ABSTRACT: To investigate the effect of AKT1 mutants discovered in juvenile granulosa-cell tumors, we generated UAS-AKT1 transgenic flies (Drosophila melanogaster), expressing either the human wild type or a mutant form of AKT1. Next, we performed high-throughput analyses of the transcripts deregulated after mild over-expression of human WT and mutated AKT1 variants in Drosophila follicular cells. Site-specific transformation of pUASTattB-AKT1-3xFLAG constructs into the 51C fly line (y1 M{vas-int.Dm}ZH-2A w*; M{3xP3-RFP.attP'}ZH-51C)(Reference BDSC Stock # 24482) was performed by Bestgene, Inc. (CA, U.S.A.).
