ScRNA-seq analysis of SHH medulloblastomas in Math1-Cre/SmoM2 mice with homozygous or heterozygous co-deletion of Bcl2l1 (aka Bcl-xL)

ABSTRACT: We found that medulloblastomas that form in Smo mutant mice grow more slowly when the animals are genetically engineered so that the gene Bcl2l1 is conditionally deleted in the tumor cells. We undertook this study to define changes in cellular heterogeneity and gene expression patterns in the Bcl2l1-deleted medulloblastomas, compared to medulloblastomas with an intact Bcl2l1 allele.

We generated mice with medulloblastomas by crossing Math1-Cre and SmoM2 mouse lines. We generated mice with medulloblastomaswith Cre-conditional Bcl2l1 deletion by breeding Bcl2l1 f/f mice into both Math1-Cre and SmoM2 mouse lines. We harvested tumors at P15 and then isolated RNA for expression array analysis.

Bioinformatic Analysis of the BCL-xL/BCL2L1 Interactome in Patients with Pancreatic Cancer.

Project description:Objectives: The aim of the present study was to analyze the differential gene expression of BCL-xL/BCL2L and the associated genetic, molecular, and biologic functions in pancreatic ductal adenocarcinoma (PDAC) by employing advanced bioinformatics to investigate potential candidate genes implicated in the pathogenesis of PDAC. Materials and Methods: Bioinformatic techniques were employed to build the gene network of BCL-xL, to assess the translational profile of BCL-xL in PDAC, assess its role in predicting PDAC, and investigate the associated biologic functions and the regulating miRNA families. Results: Microarray data extracted from one dataset was incorporated, including 130 samples (PDAC: 69; Control: 61). In addition, the expression level of BCL-xL was higher in PDAC compared to control samples (p < 0.001). Furthermore, BCL-xL demonstrated excellent discrimination (AUC: 0.83 [95% Confidence Intervals: 0.76, 0.90]; p < 0.001) and calibration (R squared: 0.31) traits for PDAC. A gene set enrichment analysis (GSEA) demonstrated the molecular functions and miRNA families (hsa-miR-4804-5p, hsa-miR-4776-5p, hsa-miR-6770-3p, hsa-miR-3619-3p, and hsa-miR-7152-3p) related to BCL-xL. Conclusions: The current findings unveil the biological implications of BCL-xL in PDAC and the related molecular functions and miRNA families.

Project description:Our parallel studies have shown that the OLIG2 inhibitor CT-179 and the TLR7/8 agonist resiquimod delivered in polyoxazoline nanoparticle (Pox-resiquimod) produce significant anti tumor effects when administered to mice with SHH medulloblastoma. To gain insight into the cellular events set in motion by each treatment, we compared sets of replicate mice with SHH medulloblastomas treated with either CT-179 or POx-resiquimod to a common set of untreated control mice with SHH medulloblastoma.

BCL-xL/BCL2L1 is a critical anti-apoptotic protein that promotes the survival of differentiating pancreatic cells from human pluripotent stem cells.

Project description:The differentiation of human pluripotent stem cells into pancreatic cells involves cellular proliferation and apoptosis during cell fate transitions. However, their implications for establishing cellular identity are unclear. Here, we profiled the expression of BCL-2 family of proteins during pancreatic specification and observed an upregulation of BCL-xL, downregulation of BAK and corresponding downregulation of cleaved CASP3 representative of apoptosis. Experimental inhibition of BCL-xL reciprocally increased apoptosis and resulted in a decreased gene expression of pancreatic markers despite a compensatory increase in anti-apoptotic protein BCL-2. RNA-Seq analyses then revealed a downregulation of multiple metabolic genes upon inhibition of BCL-xL. Follow-up bioenergetics assays revealed broad downregulation of both glycolysis and oxidative phosphorylation when BCL-xL was inhibited. Early perturbation of BCL-xL during pancreatic specification also had subsequent detrimental effects on the formation of INS+ pancreatic beta-like cells. In conclusion, the more differentiated pancreatic progenitors are dependent on anti-apoptotic BCL-xL for survival, whereas the less differentiated pancreatic progenitors that survived after WEHI-539 treatment would exhibit a more immature phenotype. Therefore, modulation of the expression level of BCL-xL can potentially increase the survival and robustness of pancreatic progenitors that ultimately define human pancreatic beta cell mass and function.
