Single-cell gene expression profile of the postnatal mouse arcuate nucleus in the hypothalamus

Project description:m6A, the most prevalent internal modification on mRNAs, plays important roles in the nervous system. Whether neurogenesis in the hypothalamus, a region critical for controlling appetite, is regulated by m6A signaling, especially in humans, remains unclear. Here we showed that deletion of m6A writer Mettl14 in the mouse embryonic hypothalamus led to adult obesity, with impaired glucose-insulin homoeostasis and increased energy intake. Mechanistically, deletion of Mettl14 leads to hypothalamic arcuate nucleus neurogenesis deficits with reduced generation of feeding-related neurons and dysregulation of neurogenesis-related m6A-tagged transcripts. Deletion of m6A writer Mettl3, or m6A reader Ythdc1, shared similar phenotypes. METTL14 or YTHDC1 knockdown also led to reduced generation of feeding-related neurons in human brain subregion-specific arcuate nucleus organoids. Our studies reveal a conserved role of m6A signaling in arcuate nucleus neurogenesis in mice and human organoids and shed light on the developmental basis of epitranscriptomic regulation of food intake and energy homeostasis.

Proteomic profiling of the hypothalamic arcuate nucleus of bull calves offered either a high or moderate plane of nutrition in early life

Project description:Enhanced nutrition during the early calfhood period has been shown to hasten the onset of puberty in bull calves. The hypothalamic-pituitary-ovarian biochemical signalling axis regulates reproductive development in bulls, with the dynamics of the arcuate nucleus region of the hypothalamus, in particular, central to mediating the relationship between metabolic status with reproductive development. However, the precise molecular mechanisms regulating the influence of metabolic status on this signalling axis in bull calves is yet to be fully elucidated. The objective of this study was to determine the effect of an enhanced plane of nutrition during early life, up to 12 weeks of age, on the proteomic profile of the arcuate nucleus of bull calves. Bull calves were offered either a high or moderate plane of nutrition.

Refined RIP-seq protocol for epitranscriptome analysis with low input materials

Project description:We optimized the key parameters of m6A MeRIP-seq, including the starting amount of RNA, RNA fragmentation, antibody selection, MeRIP washing/elution conditions, methods for RNA library construction and the bioinformatics analysis pipeline. With the optimized immunoprecipitation conditions and a post-amplification rRNA depletion strategy, we were able to profile the m6A epitranscriptome using 2 µg of total RNA. We identified ~12,000 m6A peaks with a high signal/noise ratio from two lung adenocarcinoma (ADC) patient tumors. Through integrative analysis of the transcriptome, m6A epitranscriptome and proteome data in the same patient tumors, we identified dynamics at the m6A level that accounts for the discordance between mRNA and protein levels in these tumors.
