Effect of OMVs on gene expression of primary alveolar epithelial cell

ABSTRACT: The alveolar epithelial cell injury is essential for the initaitation and maintenance of inflammatory of lung in ARDS. Outer membrane vesicles (OMVs) are nanoparticles produced by the extrusion of the outer membrane of gram-negative bacteria deliver a range of bacterial molecules to host cells. Recent findings describe OMVs play vital roles in bacterial colonization, delivery of virulence factors, and disease pathogenesis. We here report OMVs as a key regulator of gene expression in primary alveolar epithelial cell.

Haemophilus influenzae Outer membrane vesicles inhalation contributes to steroid-resistant asthma

Project description:Asthma is the most common chronic respiratory disease. Asthma that cannot be well controlled by steroid treatment is called steroid-resistant asthma. Steroid-resistant asthma accounts for only 5% of all asthma cases, but it accounts for 80% of asthma healthcare costs. Nontypeable Haemophilus influenzae (NTHi), as a Gram-negative bacterium, can release outer membrane vesicles (OMVs) and transfer biomolecules to host cells and the external environment by carrying lipopolysaccharides, proteins, peptidoglycans, outer membrane proteins, cell wall components, proteins, nucleic acids, ion metabolites, and signaling molecules. Thus, it plays a role in obtaining nutrition, stress, toxin delivery, adhesion, host immune surveillance evasion, and host immune response regulation. It becomes an essential way in bacterial pathogenesis. To further clarify whether NTHi OMVs could be inhaled to induce steroid-resistant asthma, we isolated and purified NTHi OMVs. In vivo experiments showed that NTHi OMVs could be inhaled and enter airway epithelial cells. Cosensitization with OVA induces steroid-resistant asthma in mice. Furthermore, through high-throughput sequencing, we found that the NTHi OMVs and OVA co-sensitized mice had significantly enriched inflammatory and immune-related signaling pathways, and the transcription and secretion of IL-1β were increased was the potential cause of SRA.

Inappropriate use of antibiotics exacerbates inflammation through OMVs-induced pyroptosis in multidrug-resistant Klebsiella pneumoniae infection

Project description:The inappropriate use of antibiotics is a severe public health problem worldwide, contributing to the emergence of multidrug-resistant (MDR) bacteria. To explore the possible impacts of the inappropriate use of antibiotics on the immune system, we use Klebsiella pneumoniae (K. pneumoniae) infection as an example and show that imipenem increases the mortality of mice infected by MDR K. pneumoniae. Further studies demonstrate that imipenem enhances the secretion of outer membrane vesicles (OMVs) with significantly elevated presentation of GroEL, which promotes the phagocytosis of OMVs by macrophages that depends on the interaction between GroEL and its receptor LOX-1. OMVs cause the pyroptosis of macrophages and the release of proinflammatory cytokines, which contribute to exacerbated inflammatory responses. We propose that the inappropriate use of antibiotics in the cases of infection by MDR bacteria such as K. pneumoniae might cause damaging inflammatory responses, which underlines the pernicious effects of inappropriate use of antibiotic.

Transcriptomic remodeling of gastric cells by Helicobacter pylori Outer Membrane Vesicles

Project description:Helicobacter pylori, a bacterium that colonizes the human stomach, like all Gram-negative bacteria spontaneously shed outer membrane vesicles (OMVs). OMVs, which act as a delivery system for bacterial components, are involved in bacterial-host interactions and thus contribute to pathogenesis. In this study, to understand the gene expression changes that human gastric epithelial cells might undergo when exposed to H. pylori-OMVs, we profiled the transcriptomic changes of the MKN74 gastric cell line induced by OMVs compared to control cells and H. pylori-infected cells, using the Ion AmpliSeq™ Transcriptome Human Gene Expression Panel. The top enriched pathways in the OMVs challenge condition included amino acid-related metabolic pathways, mitogen-activated protein kinase signaling, autophagy, and ferroptosis. The cell cycle, DNA replication, and repair pathways were the top diminished pathways. The transcriptomic changes induced by OMVs were largely consistent with those of the bacteria, although often at low expression levels, suggesting that their effects will mostly reinforce those of the bacterium itself. Our data provide a valuable portrayal of the transcriptomic remodeling of gastric cells by H. pylori-OMVs, which can be further dissected regarding the underlying molecular mediators and explored to understand the pathobiology of the full-spectrum of H. pylori-mediated diseases.

Genetically engineered hybridized OMVs for treatment of implant-associated infections and relapse prevention through host immunomodulation

Project description:Implant-associated infections (IAIs) are refractory to elimination and local immunosuppressive microenvironment exacerbates the therapeutic difficulty, further causing the persistence and relapse. Therefore, exploring immune-strengthening therapeutics holds great promise to reverse suppressive host immunity and thoroughly eradicate chronic or repetitive infections. Bacterial outer membrane vesicles (OMVs) emerge as a potential immunostimulatory candidate, which is however, still suffer from lacking active targeting capability and non-specific systemic inflammatory storms. Herein, we genetically engineered bone marrow-derived mesenchymal stem cells (BMSCs) to express CXCR4 and isolated the cell membranes (mBMSCCXCR4) to hybridize them with OMVs derived from Escherichia coli (E. coli) to finally produce nanovesicles (mBMSCCXCR4@OMV). Due to the hybridized membrane structure of mBMSCCXCR4, the resulting nanovesicles generated excellent targeting capability towards bone marrow and were obviously uptaken by bone marrow-derived macrophages, triggering the efficient transition to pro-inflammatory M1 status through TLR/NF-κB pathway. This alteration promoted its innate bactericidal capacity and antigen presentation. Subsequent activation of T cells and B cells derived from bone marrow and inhibition of myeloid-derived suppressor cells (MDSCs) facilitated the in vivo clearance of infections through adaptive immunity via mouse models. In addition, mBMSCCXCR4@OMV also boosted memory B cells and bacteria-specific antibody responses. Together, this study warrants our mBMSCCXCR4@OMV to eradicate complicated IAIs and provides a whole-stage protection to prevent the postsurgical relapse, marking a significant immunotherapeutic advancement in the post-antibiotic era.

Porphyromonas gingivalis outer membrane vesicles promote apoptosis via msRNA-regulated DNA methylation in periodontitis

Project description:The outer membrane vesicles (OMVs) produced by Porphyromonas gingivalis (P. gingivalis) contain a variety of bioactive molecules that may be involved in the progression of periodontitis. However, the participation of P. gingivalis OMVs in the development of periodontitis has not been elucidated. Here we isolated P. gingivalis OMVs and confirmed their participation in periodontitis both in vivo and in vitro. Microcomputed Tomography (micro-CT) and histological analysis showed that under the stimulation of P. gingivalis OMVs, the alveolar bone of rats was significantly resorbed in vivo. We found that P. gingivalis OMVs were taken up by hPDLCs (human Periodontal Ligament Cells, hPDLCs) in vitro, then subsequently resulting in apoptosis and inflammatory cytokines releasing which was accomplished by the microRNA-size small RNAs (msRNAs) sRNA45033 in the P. gingivalis OMVs. Through bioinformatics analysis and screening of target genes, Chromobox 5 (CBX5) was identified as the downstream target of screened-out small RNA s45033. Using dual-luciferase reporter assay, overexpression, and knockdown methods, s45033 was confirmed to target CBX5 to regulate hPDLCs apoptosis. In addition, Cleavage Under Targets and Tagmentation (Cut&Tag) analysis confirmed the mechanism that CBX5 regulates apoptosis through the methylation of p53 DNA. Collectively, these findings indicate that the role of P. gingivalis OMVs is immunologically relevant and related to bacterial virulence in the development of periodontitis.

Nanobody engineered and photosensitiser loaded bacterial outer membrane vesicles potentiate anti-tumor immunity and immunotherapy

Project description:Bacterial outer membrane vesicles (OMVs) are promising as antitumor agents, but their clinical application is limited by toxicity concerns and unclear mechanisms. We engineered OMVs with CDH17 tumor-targeting nanobodies, enhancing tumor selectivity and efficacy while reducing adverse effects. These engineered OMVs function as natural stimulator of interferon genes (STING) agonists, activating the cyclic GMP-AMP synthase (cGAS)-STING pathway in cancer cells and tumor-associated macrophages (TAMs). Loading engineered OMVs with photoimmunotherapy photosensitizers further enhanced tumor inhibition and STING activation in TAMs. Combining nanobody-engineered OMV-mediated photoimmunotherapy with CD47 blockade effectively suppressed primary and metastatic tumors, establishing sustained antitumor immune memory. This study demonstrates the potential of nanobody-engineered OMVs as STING agonists and provides insights into novel OMV-based immunotherapeutic strategies harnessing the innate immune system against cancer. Our findings open new avenues for OMV applications in tumor immunotherapy, offering a promising approach to overcome current limitations in cancer treatment.
