Gene expression of papillary renal neoplasm with reverse polarity (PRNRP) cases + PRCC cases

ABSTRACT: Between 80% and 90% of renal carcinomas are either clear cell renal cell carcinoma or papillary RCC although there are at least 20 other distinct forms of RCC recognised by the World Health Organization (WHO). Emerging or provisional entities have been considered as variants of PRCC, including papillary renal neoplasm with reversed polarity (PRNRP). However, the molecular basis of PRNRP is unknown. We used microarray analysis to elucidate the gene profile of 10 PRNRP cases and an additional 7 PRCC cases. Cases were only considered for this study after fulfilment of strict inclusive histological and immunohistochemical criteria. We used Affymetrix Clariom D human arrays to elucidate the expression of these samples.

Project description:Eleven TC-RCC cases were selected on the basis of the criteria defined by the 2012 Vancouver Classification of Renal Neoplasia and WHO 2016 classification criteria, and re-reviewed by a panel of expert renal pathologists. Two of these cases, TC-RCC#9 and #10, had a mixed tubulocystic and papillary histology and were macro-dissected into separate PRCC and TC-RCC components. Project description:Renal cell carcinoma is the most common neoplasm of the adult kidney. A few subtypes of RCC include papillary RCC (pRCC), chromophobe RCC (chRCC) and the benign oncocytoma tumor. In some cases, distinguishing between the RCC subyptes is difficult. We performed a mircroRNA (miRNA) microarray to determine differential miRNA expression between pRCC, chRCC, and oncocytoma. We performed a miRNA microarray on 10 tumor samples of each papillary renal cell carcinoma (pRCC), chromophobe renal cell carcinoma (chRCC), and oncocytoma.

Project description:Renal cell carcinomas (RCCs) with Xp11 translocation (Xp11 RCC) constitute a distinctive molecular subtype characterized by chromosomal translocations involving the Xp11.2 locus, resulting in gene fusions between the TFE3 transcription factor with a second gene (usually ASPSCR1, PRCC, NONO, or SFPQ). RCCs with Xp11 translocations comprise up to 1-4% of adult cases, frequently displaying papillary architecture with epithelioid clear cells.

Project description:Papillary renal cell carcinomas (pRCC) are the second most common form of renal carcinoma, after clear cell Renal carcinoma (ccRCC). PRCC account for 10 to 15% of RCC and gather a heterogeneous population with no specific systemic treatment. Pathological classification by Elbe divides pRCC population in two morphologically different subtypes. Type 1 consists of predominantly basophilic cells, whereas type 2 contains mostly eosinophilic cells. Type 1 architecture corresponds with a single line of cells along the papillary axis, whereas type 2 generally exhibits several cell strata on the axis. Furthermore, type 2 cells demonstrate more aggressive characteristics, such as the presence of nucleoli and increased nuclear size. The papillary cores often contain edema fluid, foamy macrophages, and psammoma bodies. Further clinical reports identified that type I tumors are more likely to present as numerous, bilateral , indolent, low grade pRCC, whereas type II are associated with higher grade and poor prognosis related to metastatic evolution.
