Interferon Mediated Bronchial Epithelium Cellular Senescence in Chronic Obstructive Pulmonary Disease

ABSTRACT: Cellular senescence has been implicated in chronic obstructive pulmonary disease (COPD) pathogenesis. We used single cell RNA sequencing (scRNA-seq) to analyze gene expressions in bronchial epthelium, finding increased expression of cellular senescence signature in COPD samples.

Proteomic analysis of lungs in early experimental model of Chronic obstructive pulmonary disease (COPD)

Project description:Rationale: Chronic obstructive pulmonary disease (COPD) is a prevalent respiratory disease lacking disease modifying treatment. The role of CXCL12/CXCR4 axis has been demonstrated in acute exacerbation of COPD. The interest of study early COPD has been recently pointed out. Objectives: To study the role of the CXCL12/CXCR4 axis in both human and mouse model of early COPD. Methods: Blood and lung tissue were obtained from both COPD patients and mice exposed to cigarette smoke (CS) for 10 weeks and intranasal instillations of polyinosinic–polycytidylic acid (poly(I:C)) to mimic exacerbations for 5 weeks. Measurements and Main Results: Exposed mice presented mild airway obstruction, peri-bronchial fibrosis and right heart remodeling. The level of CXCR4 expressing cells was increased in the blood of exposed mice, as well as in the blood of patients with mild COPD. Lung CXCL12 expression was higher both in exposed mice and COPD patients. The densities of fibrocytes expressing CXCR4 were increased in the blood and in the bronchial submucosa of exposed mice. Conditional inactivation of CXCR4 at adult stage as well as pharmacological inhibition of CXCR4 with plerixafor injections improved lung function and inflammation and protected against CS and poly-(I:C)-induced airway and cardiac remodeling. CXCR4-/- and plerixafor-treated mice also had reduced levels of CXCR4-expressing circulating cells and a lower density of peri-bronchial fibrocytes. Conclusions: We demonstrated that targeting CXCR4 has beneficial effects in an animal model of early COPD, and provide a framework to translate preclinical findings to clinical settings within a drug repurposing approach.

RNA sequencing of human bronchial epithelial cell transcriptomes after repopulation and differentiation on bronchial scaffolds from COPD patients and healthy donors

Project description:Chronic obstructive pulmonary disease (COPD) is a serious global health problem characterized by chronic airway inflammation, progressive airflow limitation and destruction of lung parenchyma. Remodeling of the bronchial airways in COPD includes changes in both the bronchial epithelium and the subepithelial extracellular matrix (ECM). To explore the impact of an aberrant ECM on epithelial cell phenotype in COPD we developed a new ex vivo model, in which normal human bronchial epithelial (NHBE) cells repopulate and differentiate on decellularized human bronchial scaffolds derived from COPD patients and healthy individuals. By using transcriptomics, we show that bronchial ECM from COPD patients induces differential gene expression in primary NHBE cells when compared to normal bronchial ECM. The gene expression profile indicated altered activity of upstream mediators associated with COPD pathophysiology, including hepatocyte growth factor, transforming growth factor beta 1 and platelet-derived growth factor B, which suggests that COPD-related changes in the bronchial ECM contribute to the defective regenerative ability in the airways of COPD patients.

Divergent molecular responses of bronchial human airway epithelium to SARS-CoV-2 infection in relation to pre-existing respiratory disease states

Project description:The COVID-19 pandemic has revealed differences in individual susceptibility and severity, with pre-existing respiratory diseases as potential risk factors. Using an in vitro model of differentiated primary bronchial epithelial cells, we investigated the molecular mechanisms of SARS-CoV-2 infection in cystic fibrosis (CF) and chronic obstructive pulmonary disease (COPD). Our study found that CF and COPD airway epithelium had reduced susceptibility to SARS-CoV-2 compared to healthy controls. This was linked to preserved epithelial integrity, lower viral replication, and attenuated antiviral and inflammatory responses. Mechanistically, we identified reduced TMPRSS2 activity as a contributing factor in CF epithelium. Importantly, the baseline upregulation of complement and inflammatory pathways in CF and COPD epithelium appeared to prime an antiviral state prior to infection. Analysis of a COVID-19 patient cohort validated our findings, showing correlations between specific inflammatory markers (CXCL-10, SERPINA1, and CFB) and SARS-CoV-2 infection severity. This study offers insights into SARS-CoV-2 pathogenesis and potential biomarkers for clinical monitoring.
